Spondyloarthritis (SpA) and psoriatic arthritis (PsA) are interrelated, immune-mediated inflammatory diseases characterized by significant clinical heterogeneity. Early differential diagnosis, accurate disease activity assessment, and the development of personalized treatment strategies remain significant clinical challenges. Single-omics approaches have provided only a limited view of this complexity, highlighting the need for integrative strategies. This review systematically synthesizes findings from multi-omics studies-including genomics, transcriptomics, proteomics, and microbiomics-in Sp

Post-traumatic stress disorder (PTSD) is a profound neurobiological disorder in which dysregulation of the neuroendocrine system represents a core pathophysiological feature. This Review synthesizes recent advances in PTSD neuroendocrine research to delineate a comprehensive and dynamic pathological landscape that extends beyond traditional single-axis frameworks. We first examine the central role of the hypothalamic-pituitary-adrenal (HPA) axis, highlighting the well-known paradox of enhanced central drive accompanied by low peripheral cortisol levels and discussing feedback dysregulation med

Vascular cognitive impairment and dementia (VCID) encompasses a heterogeneous spectrum of cognitive disorders driven by cerebrovascular pathology and represents a major contributor to late-life cognitive decline. VCID is highly prevalent and frequently coexists with Alzheimer's disease pathology. Despite this, it remains poorly defined in clinical practice and lacks approved disease-modifying therapies. Therapeutic development has been hindered by biological heterogeneity, challenges in patient stratification, and a historical emphasis on neurodegenerative targets that inadequately address vas

This narrative review, prepared in accordance with the Scale for the Assessment of Narrative Review Articles, surveys next-generation dental implant surface modifications for osseointegration, from nanoengineering to clinical translation. Peri-implantitis affects approximately 19.53% of patients and 12.53% of implants. Because peri-implant tissue destruction reflects a dysregulated host inflammatory response to an unavoidable, lifelong microbial challenge rather than bacterial colonization alone, the design target is shifting from surfaces that are simply bactericidal to surfaces that regulate

The retina, as an extension of the central nervous system, shares a common embryological origin with the brain. In Alzheimer's disease (AD), studies of human tissue and animal models have revealed that hallmark AD pathologies, including amyloid-β (Aβ) deposits and pathological tau protein tangles, also appear in the retina. These findings, coupled with advances in high-resolution retinal imaging techniques, suggest the potential to detect and characterize AD-related molecular and structural changes in the retina. However, retinal findings across different AD mouse models have significant discr

Tuberculosis (TB), a disease caused by Mycobacterium tuberculosis, remains one of the major causes of death from infection worldwide, with over a million associated deaths each year. The study of biomarkers for TB is critical for advancing our understanding and management of the disease. Biomarkers, defined as measurable indicators of biological states or conditions, are invaluable for the diagnosis, prognosis and treatment monitoring of TB. Clinical studies have provided critical knowledge on the matter but are also notoriously constrained by economical, ethical and sampling limitations. The

Mitophagy is a selective autophagic process responsible for the elimination of damaged or dysfunctional mitochondria, playing a critical role in maintaining mitochondrial quality control and cellular homeostasis. Dysregulated mitophagy has been implicated in the pathogenesis of numerous diseases, including neurodegenerative diseases, cancer, cardiovascular diseases, metabolic disorders, inflammatory and immune diseases, and musculoskeletal diseases. In recent years, nanotechnology-based approaches for the targeted modulation of mitophagy have emerged as promising therapeutic strategies due to

Circulating cell-free DNA (cfDNA) has attracted increasing attention as a minimally invasive biomarker for assessing radiation-induced tissue injury. Radiation exposure can alter cfDNA concentration, fragmentation profiles, and methylation patterns. These changes may reflect cellular damage, tissue origin, and temporal responses to irradiation. During radiotherapy, however, cfDNA can arise from tumor cells, irradiated normal tissues, endothelial cells, immune cells, and hematopoietic cells. This mixed origin complicates the interpretation of radiation-related signals. Current evidence is deriv

The prevalence of Alzheimer's disease and other dementias is increasing as populations live longer lifespans. Imaging is becoming a key component of the workup for patients with cognitive impairment or dementia. Integrated PET-MRI provides a unique opportunity for same-session multimodal characterization with many practical benefits to patients, referring physicians, radiologists, and researchers. The impact of integrated PET-MRI on clinical practice for early adopters of this technology can be profound. Classic imaging findings with integrated PET-MRI are illustrated for common neurodegenerat

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common pediatric liver disease, affecting approximately 10% of children. Its prevalence is rising at an alarming rate, with cases increasingly identified even in early childhood. While MASLD shares key features across the lifespan, its earlier onset reflects developmental vulnerabilities and unique mechanistic drivers. Perinatal influences, including maternal obesity, gestational diabetes, and early-life nutritional exposures, play a central role by disrupting metabolic programming, driving mitochondrial dysfunction,

The etiology of mood and schizophrenia spectrum disorders remains largely unknown. In recent years, several studies have focused on the role of the microbiome-gut-brain-axis (MGBA) in its etiology, providing several novel insights. The communication within MGBA involves various pathways leading through the vagus nerve and the bloodstream mediators. The latter are represented by short-chain fatty acids (SCFAs), produced by gut microbiota from dietary fiber. To date, several physiological and pathophysiological roles in the periphery and the central nervous system for SCFAs have been suggested.

Psychiatric disorders represent a particularly challenging area of medicine to study using non-human animals. They arise from a complex interaction between genetic and environmental factors and are diagnosed using clinical interviews and diagnostic criteria based largely on self-reported symptoms and with significant heterogeneity seen within patient populations. Anxiety and major depressive disorder are the most diagnosed psychiatric disorders, with prescriptions of antidepressants exceeding 80 million in the UK in 2024. Despite more than 70 years of research, we do not understand how antidep

Hearing loss represents a highly prevalent and debilitating sensory disorder affecting roughly one in five people worldwide. In a majority of patients with congenital hearing loss, genetic mutations cause the disease. Up until recently, therapeutic options for individuals with hearing loss were limited to hearing aids and different types of auditory implants. However, after numerous years of intensive basic and translational research, gene therapy strategies are now being investigated in clinical trials. First results show significant hearing improvement in treated patients, highlighting gene

Rotator cuff disease is one of the most common human tendinopathies and can lead to significant shoulder dysfunction. Despite efforts to improve symptoms in patients with rotator cuff tears and healing rates after rotator cuff repair, high rates of failed healing and persistent shoulder morbidity exist. Increasing interest has been placed on the utilization of orthobiologics-scaffolds, cell-based augmentation, platelet right plasma (platelet-rich plasma), and small molecule-based strategies-in the management of rotator cuff disease and the augmentation of rotator cuff repairs. This is a comple

IMPORTANCE: Chimeric antigen receptor (CAR) T cells are T lymphocytes that are genetically engineered to express a synthetic receptor that recognizes a tumor cell surface antigen and causes the T cell to kill the tumor cell. CAR T treatments improve overall survival for patients with large B-cell lymphoma and progression-free survival for patients with multiple myeloma.
OBSERVATIONS: Six CAR T-cell products are approved by the US Food and Drug Administration (FDA) for 6 hematologic malignancies: B-cell acute lymphoblastic leukemia, large B-cell lymphoma, follicular lymphoma, mantle cell lymph

Dry eye disease (DED) is an emerging global health concern with meibomian gland dysfunction (MGD) being the most common subtype of DED. Despite being quite prevalent, the pathophysiological mechanisms governing MGD are poorly understood. Animal models for MGD can be a valuable resource to advance our understanding of this entity and explore novel diagnostic and therapeutic modalities. Although a lot of literature on rodent MGD models exists, a comprehensive review on rabbit animal models is lacking. Rabbits offer a great advantage over other animals as models for studying both DED and MGD. Rab

BACKGROUND/AIM: Prostate cancer (PCa) is one of the most common types of cancer in men. Prostate-specific antigen (PSA) is currently the only biomarker used to screen for the risk of developing PCa. Because PSA tests may show false positives, identifying novel PCa-specific biomarkers would improve prediction and diagnosis at an early stage. Previously, we identified a number of genes/microRNAs (miRNAs) in prostate tissue as potential biomarkers of chronic prostatitis in a rat model of chemical-induced prostatitis. The current study aimed to evaluate their potential for use as translational, di

Visual impairment from corneal stromal disease affects millions worldwide. We describe a cell-free engineered corneal tissue, bioengineered porcine construct, double crosslinked (BPCDX) and a minimally invasive surgical method for its implantation. In a pilot feasibility study in India and Iran (clinicaltrials.gov no. NCT04653922 ), we implanted BPCDX in 20 advanced keratoconus subjects to reshape the native corneal stroma without removing existing tissue or using sutures. During 24 months of follow-up, no adverse event was observed. We document improvements in corneal thickness (mean increase

Translational research has improved patient care over the last decade. In dermatology, this research often requires human tissue for laboratory analysis. The skin biopsy remains the gold standard for tissue acquisition, but the procedure comes with a small risk of bleeding and infection. It also causes scarring and anxiety in certain populations. These risks and concerns may affect participation rates in translational studies, which can require multiple biopsies. Minimally invasive procedures may mitigate these risks and concerns. We queried the PubMed database for all minimally invasive techn

OBJECTIVE: Preterm birth (PTB) is the leading cause of infant morbidity and mortality worldwide. Canada and Japan each have strengths that can inform clinical decision-making, research, and health care policy regarding the prevention of PTB and its sequelae. Our objectives were to: 1) compare PTB rates, risk factors, management, and outcomes between Japan and Canada; 2) establish research priorities while fostering future collaborative opportunities; and 3) undertake knowledge translation of these findings.
METHODS: We conducted a literature review to identify publications that examined PTB r
