Toxoplasma gondii DNA in Tissues of Anadromous Arctic Charr, Salvelinus alpinus, Collected From Nunavik, Québec, Canada.

BACKGROUND: Toxoplasma gondii is a very common zoonotic parasite in humans and animals worldwide. Human seroprevalence is high in some regions of Canada's North and is thought to be associated with the consumption of traditionally prepared country foods, such as caribou, walrus, ringed seal and beluga. While numerous studies have reported on the prevalence of T. gondii in these animals, in the general absence of felid definitive hosts in the North there has been considerable debate regarding the source of infection, particularly in marine mammals. It has been proposed that fish could be involv

Whole-Genome Sequencing-Based Characterization of Listeria monocytogenes from Fish and Fish Production Environments in Poland.

Listeria monocytogenes, an important foodborne pathogen, may be present in different kinds of food and in food processing environments where it can persist for a long time. In this study, 28 L. monocytogenes isolates from fish and fish manufactures were characterized by whole genome sequencing (WGS). Core genome multilocus sequence typing (cgMLST) analysis was applied to compare the present isolates with publicly available genomes of L. monocytogenes strains recovered worldwide from food and from humans with listeriosis. All but one (96.4%) of the examined isolates belonged to molecular serogr

Comparison of the toxicity of two chelated copper algaecides and copper sulfate to non-target fish.

New pesticide products are reviewed by the United States Environmental Protection Agency for possible effects to non-target aquatic organisms. The required toxicity data are for the active ingredient only, and fail to include toxicity of the mixture of other ingredients found in these pesticides. These ingredients may increase the toxicity of the active ingredient to non-target organisms. Our study compares the toxicity of two formulations of chelated copper algaecides with each other, and to a copper sulfate algaecide. We were particularly interested in the effects of a surfactant that is pre

Production of triploid transgenic zebrafish, Brachydanio rerio (Hamilton).

Eggs of zebrafish Brachydanio rerio were heat-shocked just after fertilisation to induce triploidy and subsequently subjected to microinjection with the linear recombinant plasmid containing the Rous Sarcoma Virus-Long Terminal Repeat (RSV-LTR) promoter linked to the rainbow trout (Oncorhynchus mykiss) growth hormone (GH)cDNA(pRSVrtGH). Slot and Southern analyses showed the extra chromosomal transfer of pRSVrtGH. Expression of pRSVrtGH was detected in the triploid transformants by RNA slot, Northern and Western analyses.

A century of allopatry: plasticity and rapid selection shape phenotypic trait variability under contrasting environments.

Allopatric isolation under contrasting environments can drive rapid phenotypic divergence, even over contemporary timescales. Rapid changes in morphology or physiology can allow organisms to adapt to biotic and abiotic characteristics of their habitats. While studying metabolism, growth and resource needs may allow us to understand adaptation to several selective pressures, these traits are rarely jointly considered. We investigated morphological, growth and metabolic divergence in two allopatric populations of Arctic charr (Salvelinus alpinus) sharing a common evolutionary origin but inhabiti

Toxicity assessment of 6PPDQ and Its hydroxy metabolites in fish cell lines and coho salmon.

N-(1,3-Dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPDQ), a transformation product of the tire antidegradant 6PPD, is associated with acute mortality in sensitive salmonids. Although hydroxylation is typically considered an initial detoxification step prior to conjugation, the toxicity of specific hydroxylated metabolites remains poorly understood. In this study, we evaluated the cytotoxicity of 6PPDQ, quinone ring-hydroxylated 6PPDQ-1-OH, and phenyl ring-hydroxylated p‑hydroxy-6PPDQ using three salmonid-derived cell lines (coho salmon CSE 119, rainbow trout RTG 2, and Chinook salmon…

An updated transketolase activity assay to probe thiamine utilization in fish.

Thiamine (vitamin B1) deficiency is causative of reproductive failures and population declines in lake trout, Atlantic salmon, and other predatory fish species. Transketolase (TKT) is the rate-limiting enzyme of the non-oxidative phase of the pentose-phosphate pathway. TKT is critical for mediating the availability of sugars to return to glycolysis and for synthesizing NADPH and R5P, which are needed to maintain the cellular oxidation state and to produce biomolecules necessary for successful growth and reproduction. TKT activity provides a measure of functional thiamine availability since it …

Interaction of carboxylated CdSe/ZnS quantum dots with fish embryos: Towards understanding of nanoparticles toxicity.

Due to colloidal instability even with protective coatings, nanoparticles tend to aggregate in complex environments and possibly interact with biota. In this study, visualization of quantum dots (QDs) interaction with rainbow trout (Oncorhynchus mykiss) embryos was performed. Studies on zebrafish (Danio rerio) and pearl gourami (Trichogaster leerii) embryos have shown that QDs interact with embryos in a general manner and their affects are independent on the type of the embryo. It was demonstrated that carboxylated CdSe/ZnS QDs (4 nM) were aggregating in accumulation media and formed agglomera…

Invasion of Ceratomyxa shasta (Myxozoa) and comparison of migration to the intestine between susceptible and resistant fish hosts.

The myxozoan parasite Ceratomyxa shasta infects salmonids causing ceratomyxosis, a disease elicited by proliferation of the parasite in the intestine. This parasite is endemic to the Pacific Northwest of North America and salmon and trout strains from endemic river basins show increased resistance to the parasite. It has been suggested that these resistant fish (i) exclude the parasite at the site of invasion and/or (ii) prevent establishment in the intestine. Using parasites pre-labeled with a fluorescent stain, carboxyfluorescein succinimidyl diacetate (CFSE), the gills were identified as th…

Cloning, localization and functional properties of a cGMP-gated channel in photoreceptor cells from fish pineal gland.

The perception of photic information and its translation into a rhythmic melatonin signal differ considerably among vertebrates. In the fish pineal gland, melatonin biosynthesis is controlled directly by the natural light/dark cycle. There are indications that the mechanisms of phototransduction are similar in the retinal and pineal photoreceptor cells. Here we report the molecular cloning of a novel ionic cyclic guanosine monophosphate (cGMP)-gated channel from trout pineal photoreceptors. The deduced amino acid sequence exhibits a high sequence homology to cyclic nucleotide-gated-3 (CNG) cha…

[Botulism in Osterdalen in 1831].

The earliest report we have had of an outbreak of botulism in Norway is from 1934, of five cases caused by contaminated salted and dried ham. All patients survived. This paper describes an early outbreak of botulism in the county of Hedmark in 1831 on the basis of a previously unknown report to the county governor from district public health officer Peter Heiberg (1778-1849). Two cases were reported, one of them fatal. The contaminated food was salted and fermented trout or char.

The bloodless blood knot.

Just as it is difficult to describe adequately the exhilaration one feels when using a fly rod to land a trout caught from a mountain stream, there is also a tremendous amount of satisfaction in the successful completion of an obstetric operation. Until recently, we were woefully ignorant of how fly fishing expertise could benefit pregnancy. We report with great pride an instance in which fly fishing knot skill was essential to successful placement of a cervical cerclage for a woman with an incompetent cervix.

Pathogen coinfections in wild and farmed salmonids: A systematic review.

Coinfections have been reported in a wide range of organisms, including wild and farmed salmon and trout. However, the effects of coinfections on host resistance to pathogens remain poorly understood. A primary pathogen can modulate the host's immune response to subsequent infections, either by suppressing or priming the immune system. However, in some cases, pathogens may compete within the same host, potentially reducing or neutralizing the severity and impact of the disease. This systematic review examines the impacts of coinfections on farmed and wild salmonids, synthesizing evidence from …
