Comparison of isometamidium chloride and homidium bromide as prophylactic drugs for trypanosomiasis in cattle at Nguruman, Kenya.

The duration of prophylaxis provided by isometamidium chloride and homidium bromide, each at a dose rate of 1 mg kg-1 bodyweight, was compared in a 12-month field trial involving groups of 30 zebu cattle in south-west Kenya. The trial took place between February 1990 and February 1991 and included several months of high trypanosome challenge. Cattle in the prophylaxis groups were retreated on a group basis when 10% of the group had become infected since the previous group treatment. On this basis the mean intervals between retreatment were 7.5 +/- 1.9 and 4.6 +/- 2.1 weeks for the isometamidiu

Dysregulation of photic induction of Fos-related protein in the biological clock during experimental trypanosomiasis.

The mammalian suprachiasmatic nuclei of the hypothalamus (SCN) serve as pacemaker for circadian rhythms and the immediate-early gene c-fos is known to be induced by photic stimulation in the SCN of rodents. We studied the induction of Fos-related protein following a light pulse in rats infected with Trypanosoma brucei. This parasite causes in humans African sleeping sickness, a neuropsychiatric syndrome that involves changes of endogenous biological rhythms. Fos-like immunoreactivity after photic stimulation was dramatically reduced in the SCN of trypanosome- infected rats during the subjectiv

[Evaluation of the parasitologic technics used in the diagnosis of human Trypanosoma gambiense trypanosomiasis in the Ivory Coast].

The investigators carried out a comparative evaluation of twelve or parasitological techniques used nowadays in the diagnosis of human trypanosomiasis and parasite isolation in the lymph fluid, blood and cerebro-spinal fluid (CSF). The tests were performed on 64 seropositive suspects selected with TESTRYP-CATT among 661 attendants examined at the Projet de Recherches Cliniques sur la Trypanosomiase (PRCT), Daloa, Côte-d'Ivoire. The study showed that the sensitivity of the different techniques varies between 17.2% (for CSF inoculation to Mastomys) and 84.5% (for the anion exchange centrifugatio…

Histopathological and immunohistochemical study of the brain in the acute and chronic phases of experimental trypanosomiasis cruzi in dogs.

A histopathological and immunohistochemical study was undertaken of the brains of 12 young purebred Pinscher dogs, eight of which were inoculated intraperitoneally with the Colombiana strain of Trypanosoma cruzi. The four non-infected dogs formed the control group. An immunoperoxidase method was employed for demonstrating amastigotes of T. cruzi in tissue sections. Three infected dogs died in the acute phase; the other five were sacrificed at different stages of the chronic phase, one of them having survived for almost three years after the inoculation. Histopathological and immunohistochemica

[A trial treatment with eflornithine of trypanosomiasis caused by Trypanosoma brucei gambiense in the Peoples Republic of the Congo].

In a multiclinic trial in Brazzaville, Congo, 14 patients with late-stage Trypanosoma brucei gambiense trypanosomiasis were treated with eflornithine. All cases had previously been treated with one or several courses of melarsoprol. Eflornithine treatment consisted of 400 mg/kg/day intravenously for 14 days followed by 300 mg/kg/day orally for 21 days. After treatment all patients had a disappearance of trypanosomes from cerebrospinal fluid (CSF), a normalization of CSF WBC count, and, where present prior to study, a clear, rapid and lasting amelioration of neurological signs. Neither clinical…

Studies on the influence of non-steroidal anti-inflammatory drugs upon trypanosomiasis in goats and sheep.

The non-steroidal anti-inflammatory drug (NSAID) flurbiprofen caused a rise in parasitaemia in goats infected with Trypanosoma vivax, Trypanosoma congolense and Trypanosoma brucei. All trypanosome-infected goats treated with flurbiprofen showed many dividing trypanosomes. This also included the short-stumpy forms of T. brucei. In T. vivax-infected goats flurbiprofen treatment resulted in 100% mortality in the acute and chronic stages of the infection. The increase in parasitaemia of T. brucei infected goats, treated with flurbiprofen, was not associated with an increase in mortality. The incre

Efficacy of combinations of difluoromethylornithine and bleomycin in a mouse model of central nervous system African trypanosomiasis.

DL-alpha-Difluoromethylornithine, a polyamine biosynthesis inhibitor, and bleomycin, a currently used antineoplastic agent, have each previo … by demonstrating that melarsoprol, an arsenical and currently the only drug used for human African trypanosomiasis involving the brain, was also curative for these brain infections. Neither difluoromethylornithine nor bleomycin alone was curative for the brain infections; however, many combinations of the two drugs were found to be 100% curative with no evidence …

[Symptoms of human trypanosomiasis at the first diagnostic phase in the People Republic of Congo (author's transl)].

A study of clinical symptoms of trypanosomiasis has been carried out on 317 patients diagnosed actively in the field by lymphatic gland puncture and fluorescent antibody test in two foci of People's Republic of the Congo. The presence and frequency of the signs have been evaluated according to the focus, age, sex, period, the level of disturbance of C.S.F. and the results of parasitology. Thus, the authors demonstrate that: --9 p. 100 of patients show symptoms; --symptomatology of the woman is richer than that of the man; --several symptoms are closely related to age (fever, glands hypertrophy…

Susceptibility to African trypanosomiasis of N'Dama and Zebu cattle in an area of Glossina morsitans submorsitans challenge.

The use of trypanotolerant livestock is considered to be an important strategy for the control of African animal trypanosomiasis. In order to define the extent of the differences in susceptibility and productivity, 10 Zebu cows (a breed considered trypanosusceptible) and 10 N'Dama cows (a breed recognised for trypanotolerance) were exposed to a natural field challenge from Glossina morsitans submorsitans Newstead. The animals were two-and-a-half to three years old and had not been previously exposed to trypanosomiasis. All Zebu died of trypanosomiasis within eight months of first exposure. In …

Pleomorphism and the problem of recrudescent parasitaemia following treatment with salicylhydroxamic acid (SHAM) in African trypanosomiasis.

The curative action of a single dose of salicylhydroxamic acid (SHAM) in combination with glycerol was investigated in mice infected with a variety of monomorphic and pleomorphic African trypanosomes. The treatment of mice infected with monomorphic members of the sub-genus Trypanozoon was always successful and resulted in a complete cure. Treatment of animals with pleomorphic infections resulted in an initial clearance of the parasitaemia which later recrudesced. Both chronic and acute infections with Trypanosoma vivax were radically cured with SHAM plus glycerol, while infections with T. cong

Tsetse movement in wind fields: possible epidemiological and entomological implications for trypanosomiasis and its control.

This paper presents evidence that tsetse flies (Glossina) can be dispersed by wind. This dispersal in West Africa is suggested to be along a south-west north-east axis. The implications of wind dispersal of Glossina for chemical and genetic control operations is discussed. Field experiments necessary to test this hypothesis are recommended. A study of human trypanosomiasis foci in West Africa has revealed that foci are orientated in roughly parallel lines in a south-west north-east direction. This directionally was significant when compared with 7 other compass points. It is proposed that foci…

The role of the host immune response in the development of tissue lesions associated with African trypanosomiasis in mice.

A variety of tissue lesions occurs in African trypanosomiasis, in the pathogenesis of which direct toxic effects of the parasite as well as immunological mechanisms may be involved. The purpose of the present study was to evaluate the role of the host immune response in inducing tissue damage in this disease and particularly in the production of lesions in striated muscle. The development of muscle lesions in T. brucei infection was studied in several groups of mice with different forms of immunodeficiency, as well as in normal mice. In the normal mice, foci of intense inflammation and necrosi…

[Comparative study of immunoenzyme technics, ELISA and on formed antigens, in the immunologic diagnosis of African trypanosomiasis].

The enzyme linked immunosorbent assay (ELISA) for the serodiagnosis of human african trypanosomiasis is described, comparatively with the immunoperoxidase technique using blood streamforms of Trypanosoma brucei as antigen. Practical aspects for measuring the amount of antibody in serum samples from infected rabbits and from patients with african sleeping sickness are discussed. ELISA does offer important advantages over the other method used namely: -- quantitative estimates are possible, -- it bends itself readily to large scale series, owing the fact that it can be fully automatic. These qua…

[Value of indirect immunofluorescence in the diagnosis of "Trypanosoma gambiense" trypanosomiasis. 46 cases (author's translation)].

Several indirect fluorescents reactions were performed on 46 trypanosomepositive specimens which had been confirmed by parasitologic diagnos … individuals possessing a serum titer of I:640 or higher are shown to need definitive treatment for trypanosomiasis, while those with titers of I:320 are those who may either be treated with Lomidine or tested regularly for change in serum titer.
