Filariasis is a common disabling parasitic disease in this region and cytological diagnosis is often not required. Cytology has important role in diagnosis of sub-clinical filariasis. Most cases of cytologically diagnosed filariasis are clinically unanticipated. Microfilaria, ova and fragments of adult worm of Wuchereria bancrofti, in exfoliative as well as aspiration cytology have been reported and are useful in cytological detection of bancroftian filariasis. Microfilaria is frequently detected in association with neoplasm, although the role in tumorogenesis is controversial. The objective o…

Filariasis is a major public health problem throughout many regions of the tropics. The disease is caused by several species of filarial nematode including Wuchereria bancrofti and Brugia malayi, the agents of lymphatic filariasis, and Onchocerca volvulus, the cause of 'riverblindness'. Disease caused by these worms varies depending on the tissue location of the parasite, and is associated with episodes of acute and chronic inflammation. These pathologies, including elephantiasis and blindness, rank among the most disabling in the world. Studies aimed at characterizing the molecular nature of …

Floating layers of polystyrene beads suffocate mosquito larvae and pupae and inhibit egg laying. The layers are very durable in breeding sites with water contained within walls, as in wet pit latrines and soakage pits. In some areas such pits constitute an important breeding site for Culex quinquefasciatus. Trials have been conducted in communities in Zanzibar, Tanzania, and in Tamil Nadu, India, where such mosquito populations were the vectors of Wuchereria bancrofti. In each case, treatment of all the pits with polystyrene beads was integrated with mass treatment of the people with antifilar…

The age- and sex-specific distributions of human infections with Wuchereria bancrofti were investigated at two sites in the Varanasi region of north India: one a rural, agricultural area (Chiraigaon) and the other an urban-slum area (Sunderpur). A random clinical and parasitological survey revealed that the prevalence of microfilaraemia and elephantiasis in the urban area (14% and 7.3%, respectively) were both higher than in the rural area (9% and 3.1%, respectively). In both areas, prevalence of microfilaraemia generally increased with age, to a maximum in those aged 20-29 years, and then dec…

Wuchereria bancrofti in Pernambuco was first documented in 1952 (Azevedo & Dobbin 1952), and since then it has been reported in surveys carried out in selected areas of Recife. Several surveys were carried out from 1981 to 1991 by SUCAM. In the 1985 SUCAM's report the disease is considered under control. The CPqAM Filariasis Research Program was established in 1985 and a filarial survey was carried out in the town of Olinda, Greater Recife. In order to verify the real epidemiological situation, a study was conducted in the city of Recife. 21/36 of the Special Zones of Social Interest (ZEIS), w…

It is apparent that there are many similarities among the various filariae. Besides a common life cycle, with an arthropod vector and human hosts, there are similarities in the diseases that they produce. This clinical picture takes one of two main courses: (1) characteristic disease produced by the presence of adult nematodes in their target tissue, which distinguishes typical cases for each of the filariae, and (2) the systemic hypersensitivity reactions to the circulating microfilaria, which tend to be similar. The characteristic feature of Wuchereria bancrofti is genital disease with funic…

The responses of cats to Brugia pahangi, which parasitizes them in nature, mimic those of humans to Brugia malayi and Wuchereria bancrofti in many important respects. It is likely that many of the features of the relationship between host and parasite that can be studied in cats infected with B. pahangi also apply to humans, but for a variety of practical reasons cannot be demonstrated in humans. Both immunologically and parasitologically there is a profound difference between B. pahangi-infected cats that have microfilariae in their blood and those that either do not become microfilaraemic or…

Examination of blood from randomly selected residents in four subdivisions of Puerto Limón, Costa Rica, demonstrated Wuchereria bancrofti infection in 1.8% of 2,879 persons. Previous studies in four other subdivisions of the city revealed infection in 2.7% of 2,338 persons. In both studies the microfilaremia rate in males was approximately twice that in females. Distribution by race was similar in the two surveys, with microfilaremia in 1% of 1,052 white and 4.1% of 1,286 black residents in the first survey and 0.8% of 2,216 white and 5.4% of 663 black residents in the second. The clinical and parasitologic aspects of filariasis were investigated in 459 inhabitants of a South Pacific island endemic for subperiodic Wuchereria bancrofti filariasis. Episodes of filarial fevers, usually with concomitant lymphangitis and/or lymphadenitis, were experienced by 26%. Lymphobstructive lesions, manifest in the later decades of life, were present in 12 with elephantiasis and 24 with hydroceles. Microfilaremia was detectable by membrane filtration of 1 ml of blood in 33% of persons, with a quarter of these having less than 50 microfilariae/ml. Microfilaremia and filarial fevers …

A sensitive and specific diagnostic kit is crucial for the detection of human lymphatic filariasis at the early stage of infection as the existing diagnostic tools are inefficient and expensive. In the present study, we have cloned and expressed Brugia malayi HSP70 (BmHSP70) protein and characterized it as a potential antigen for diagnosis of the asymptomatic microfilariae stage of Wuchereria. bancrofti infection using ELISA, western blot, and bioinformatics tools. The antigenic efficacy of BmHSP70 was also compared with ScHSP70. The BmHSP70 and ScHSP70 peptide showed highly antigenic in natur…

The Culicidae family is represented by approximately 3,610 species, among which many are involved in transmission of pathogens and parasites, thus presenting great medical-veterinary importance. In Brazil, the state of Pernambuco is considered an endemic area for many diseases vectored by mosquitoes, such as human and canine filariasis, caused by Wuchereria bancrofti and Dirofilaria immitis, respectively. The aim of this study was to evaluate the occurrence of immature forms of culicids, over an one-year period, in a specific area of northeastern region of Brazil. Culicids larvae were collecte…

Lymphatic filariasis is endemic in the coastal areas of Kenya, with four major foci identified in the early 1970s. The prevalence and intensity of Wuchereria bancrofti infection, together with antifilarial antibody responses, were assessed in a historically highly endemic focus along the River Sabaki, in Malindi district. The prevalences of microfilaraemia and antigenaemia (detected by Og4C3 ELISA) were >20% and >40%, respectively, and both increased steadily with age. The high prevalences of antifilarial IgG1 (86%) and IgG4 (91%) responses indicate that most people living in this setting are …

Although ultrasonography has allowed 'nests' of live adult worms and dilated lymphatics to be detected in the early stages of infection with Wuchereria bancrofti, previous attempts to locate such adult-worm nests in brugian filariasis have been unsuccessful. In this study, the successful location of live adult Brugia malayi parasites, in the lymphatics of the axilla, thigh, epitrochlear region and/or popliteal fossa of children aged 3-15 years, is described for the first time. The 'filarial dance sign' (FDS), which indicates the presence of live adult worms, was observed in six children with m…

Filarial nematode parasites are a serious cause of morbidity in humans and animals. Identification of filarial infection using traditional morphologic criteria can be difficult and lead to misdiagnosis. We report on a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)-based method to detect and differentiate a broad range of filarial species in a single PCR. The first internal transcribed spacer 1 (ITS1) along with the flanking 18S and 5.8S ribosomal DNA (rDNA) were isolated and cloned from Wuchereria bancrofti, Brugia malayi, and Brugia pahangi. Sequence analysis id…

The NOW Filariasis card test is a useful tool for rapid field diagnosis of Wuchereria bancrofti infection, based on detection of specific circulating filarial antigen (CFA) in the patients' blood. Concern has been raised that a high proportion of infection negative individuals develop false positive reactions in these tests when the test cards are left for a prolonged period before being examined. We carried out a survey in an endemic Tanzanian village to investigate this phenomenon. Individuals who were positive in the NOW Filariasis test at 10 min after specimen application were also positiv…
