Smoking and Air Pollution as Pro-Inflammatory Triggers for the Development of Rheumatoid Arthritis.

INTRODUCTION: Smoking is now well recognized not only as a risk factor for rheumatoid arthritis (RA), but also as a determinant of disease activity, severity, response to therapy, and possibly mortality. METHODS: Studies, mostly recent, which have provided significant insights into the molecular and cellular mechanisms which underpin the pathogenesis of smoking-related RA, as well as the possible involvement of other types of outdoor and indoor pollution form the basis of this review. RESULTS: Smoking initiates chronic inflammatory events in the lungs. These, in turn, promote the release of

A 30-year retrospective on the respirometric method for measuring instantaneous metabolic fuel use in fish.

In 1996, based on the ideas of Max Kleiber, we proposed a respirometric method in this journal for the quantitative determination of the roles of protein, lipid and carbohydrate as substrates for fueling aerobic metabolism in fish on an instantaneous basis. Here I provide a 30-year retrospective on its performance, explaining how it works, methodological challenges, the applications for which it has been used, its strengths and potential flaws, and the important issues to be addressed going forward. The approach is based on the simultaneous measurement of the rates of O2 consumption (ṀO2), CO2

Enabling protein-rich microalgae production from anaerobic digestate through direct aeration stripping for gaseous nitrogen and carbon supply.

Efficient removal and recovery of nitrogen (N) from N-rich waste streams are essential to mitigate their environmental impact and to enhance the sustainability and circularity of N-demanding processes, such as the production of protein-rich microalgal biomass. This study investigated the integration of a direct aeration stripping process, recovering gaseous ammonia and CO2 from anaerobic digestate, with the growth of a mixed microalgal culture in a photobioreactor, targeting the conversion of the recovered gases into protein-rich microalgae while preventing cross-contamination from the waste d

Mimicking Kidney Flow Shear Efficiently Induces Aggregation of LECT2, a Protein Involved in Renal Amyloidosis.

Aggregation of leukocyte cell-derived chemotaxin 2 (LECT2) causes ALECT2, a systemic amyloidosis that affects the kidney and liver. Homozygosity of the I40V LECT2 mutation is believed to be necessary but not sufficient for the disease. Previous studies established that LECT2 fibrillogenesis is greatly accelerated by loss of its single bound zinc ion and stirring or shaking. These forms of agitation are often used to facilitate protein aggregation, but they create heterogeneous shear conditions, including air-liquid interfaces that denature proteins, that are not present in the body. Here, we d

The flavonoids of Sophora flavescens exerts anti-inflammatory activity via promoting autophagy of Bacillus Calmette-Guérin-stimulated macrophages.

Tuberculosis (TB), a highly infectious air-borne disease, has remained a global health problem. Conventional treatment and preventions such as antibiotics and Bacilli Calmette-Guerin (BCG) vaccine can be unreliable. In view of the increasing prevalence of anti-TB drug resistance, adjunctive therapy may be necessary to shorten the recovery time. We have previously shown that flavonoids in the medicinal herb Sophora flavescens exhibit anti-inflammatory and bactericidal activities. The aim of this study was to investigate the molecular and cellular characteristics of flavonoids of S. flavescens (

Quantitative proteomics reveals an altered cystic fibrosis in vitro bronchial epithelial secretome.

Alterations in epithelial secretions and mucociliary clearance contribute to chronic bacterial infection in cystic fibrosis (CF) lung disease, but whether CF lungs are unchanged in the absence of infection remains controversial. A proteomic comparison of airway secretions from subjects with CF and control subjects shows alterations in key biological processes, including immune response and proteolytic activity, but it is unclear if these are due to mutant CF transmembrane conductance regulator (CFTR) and/or chronic infection. We hypothesized that the CF lung apical secretome is altered under c

VARA attenuates hyperoxia-induced impaired alveolar development and lung function in newborn mice.

We have recently shown that a combination of vitamin A (VA) and retinoic acid (RA) in a 10:1 molar ratio (VARA) synergistically increases lung retinoid content in newborn rodents, more than either VA or RA alone in equimolar amounts. We hypothesized that the increase in lung retinoids would reduce oxidative stress and proinflammatory cytokines, resulting in attenuation of alveolar simplification and abnormal lung function in hyperoxia-exposed newborn mice. Newborn C57BL/6 mice were exposed to 85% O₂ (hyperoxia) or air (normoxia) for 7 or 14 days from birth and given vehicle or VARA every other

Immunohistochemical surfactant protein-A expression: fatal drowning vs. postmortem immersion.

The postmortem diagnosis of drowning continues to be one of the most difficult in forensic pathology because of unspecific autopsy findings. It must be always remembered that disposal of a victim body in water is not unknown in homicide. The most important physiological consequence in fatal drowning is hypoxemia. The air-liquid interface of alveoli and distal airways of the mature lung are lined with a thin layer of lung surfactant, composed of phospholipids, proteins and neutral lipids. Surfactant components are synthesized and/or incorporated into lung surfactant in alveolar type II cells, a

[Dynamic changes in vascular endothelial growth factor and endothelial nitric oxide synthase in lungs of premature rats after hyperoxia exposure].

OBJECTIVE: Recent studies suggest that the disruption of vascular endothelial growth factor (VEGF) and endothelial nitric oxide synthase (eNOS) functions plays a pivotal role in the pathogenesis of bronchopulmonary dysplasia (BPD). The aim of this study was to investigate the changes of VEGF and eNOS expression in the lungs of premature rats exposed to moderate hyperoxia in order to explore possible relationships with BPD. METHODS: Premature rats delivered by hysterotomy at 21 days gestation were randomly continuously exposed to moderate hyperoxia (60% FiO2) and room air. The rats were sacrif

Wet lung leading to RDS: the lung ultrasound findings and possible mechanisms - a pilot study from an animal mode.

BACKGROUND: Clinically, the lung ultrasound (LUS) showed wet lung could cause respiratory distress syndrome (RDS) in newborns. This work aimed to investigate LUS changes over time and its potential mechanism as alveolar fluid increase in a rabbit model. METHODS: A total of 35 New Zealand Rabbits were randomly assigned to seven groups. Models of various alveolar fluid levels were induced by infusion of different volumes of normal saline (NS) via the endotracheal tube. LUS was performed before NS infusion, immediately after NS infusion and 4 h after NS infusion. To appraise LUS changes and its

Exposure to the three structurally different PCB congeners (PCB 118, 153, and 126) results in decreased protein expression and altered steroidogenesis in the human adrenocortical carcinoma cell line H295R.

Polychlorinated biphenyls (PCB), synthetic, persistent organic pollutants (POP), are detected ubiquitously, in water, soil, air, and sediments, as well as in animals and humans. PCB are associated with range of adverse health effects, such as interference with the immune system and nervous system, reproductive abnormalities, fetotoxicity, carcinogenicity, and endocrine disruption. Our objective was to determine the effects of three structurally different PCB congeners, PCB118, PCB 126, and PCB 153, each at two concentrations, on the steroidogenic capacity and proteome of human adrenocortical c

Interaction of recombinant surfactant protein D with lipopolysaccharide: conformation and orientation of bound protein by IRRAS and simulations.

Effective innate host defense requires early recognition of pathogens. Surfactant protein D (SP-D), shown to play a role in host defense, binds to the lipopolysaccharide (LPS) component of Gram-negative bacterial membranes. Binding takes place via the carbohydrate recognition domain (CRD) of SP-D. Recombinant trimeric neck+CRDs (NCRD) have proven valuable in biophysical studies of specific interactions. Although X-ray crystallography has provided atomic level information on NCRD binding to carbohydrates and other ligands, molecular level information about interactions between SP-D and biologic

Attenuation of cadmium chloride induced cytotoxicity in murine hepatocytes by a protein isolated from the leaves of the herb Cajanus indicus L.

Cadmium has been recognized as a strong environmental pollutant. Exposure to this heavy metal occurs through the intake of foodstuffs, drinking water and also via the inhalation of air. Present study was conducted to evaluate the protective effect of a 43 kDa protein, isolated from the leaves of the herb Cajanus indicus, against cadmium-induced cytotoxicity in hepatocytes. For this study, cadmium chloride (CdCl(2)) has been used as the source of cadmium. Treatment of hepatocytes with 800 microM CdCl(2) for 3 h caused significant reduction in cell viability in association with the increased lev

The Role of Endoplasmic Reticulum Stress in Extrinsic Skin Aging and Protective Strategies of Natural Plant Products.

BACKGROUND: The complicated process of extrinsic skin aging is brought on by long-term exposure to ultraviolet (UV) radiation, air pollution, and other environmental stressors. Its primary manifestations include deep wrinkles, skin laxity, abnormal pigmentation, epidermal barrier dysfunction, chronic inflammation, and telangiectasia. According to recent research, endoplasmic reticulum stress (ERS) is an important stress mechanism involved in extrinsic skin aging. Through the production of reactive oxygen species (ROS), disruption of Ca2+ homeostasis, and defects in protein folding, extrinsic e

Life in the margins: the effect of immersion/emersion and tidal cycle on the North Atlantic limpet Patella vulgata protein synthesis rates.

Biological processes in intertidal species follow tidal rhythms that enhance survival and fitness. Whereas tidal effects on behaviour and metabolic rates have been widely studied, impacts on other key process such as protein synthesis are still poorly understood. To date, no studies have examined the effect of immersion/emersion and tidal cycles on protein synthesis rates (ks). Patella vulgata is an intertidal limpet present in North-Eastern Atlantic rocky shores from high to low shore. Previously reported P. vulgata respiration and heart rate measurements suggest aerobic metabolism is maintai

Development and Evaluation of an Eco-Friendly Hand Sanitizer Formulation Valorized from Fruit Peels.

Hand sanitizer usage has proven to be a common and practical method for reducing the spread of infectious diseases which can be caused by many harmful pathogens. There is a need for alcohol-free hand sanitizers because most hand sanitizers on the market are alcohol-based, and regular use of them can damage the skin and can be hazardous. India is the world's largest producer of fruits and one of the major problems after fruit consumption is their peels, causing waste management problems and contributing to the formation of greenhouse gases leading to air pollution and adding to the problem of c

The Protective Effects of Endogenous PACAP in Oxygen-Induced Retinopathy.

Pituitary adenylate cyclase-activating polypeptide (PACAP) is a neuropeptide having trophic and protective functions in neural tissues, including the retina. Previously, we have shown that intravitreal PACAP administration can maintain retinal structure in the animal model of retinopathy of prematurity (ROP). The purpose of this study is to examine the development of ROP in PACAP-deficient and wild-type mice to reveal the function of endogenous PACAP. Wild-type and PACAP-knockout (KO) mouse pups at postnatal day (PD) 7 were maintained at 75% oxygen for 5 consecutive days then returned to room

Impact of room air resuscitation on early growth response gene-1 in a neonatal piglet model of cerebral hypoxic ischemia.

Early growth response gene-1 (Egr-1) is up-regulated by hypoxia-ischemia (HI) and reactive oxygen species (ROS) in adult animals, functioning as a master switch in inflammation and thrombogenesis. We hypothesized that resuscitation from HI with 100% O2 would result in greater Egr-1 expression, ROS, and cell death (CD) in the brains of newborn piglets than 21% O2. Two control groups breathed 21% O2 for 1 h followed by 21% or 100% O2 for 1 h. Two HI groups underwent carotid artery occlusion and breathed 8-12% O2 for 1 h followed by occlusion release and 21% or 100% O2 for 1 h. Brain Egr-1 mRNA a
