A range of bacteria biofilm models exist for the testing of antibiotics. However, many of these are limited to a single experimental output, such as colony-forming units or metabolic activity. Furthermore, many biofilm models do not reflect the biological and physiochemical properties of the human host environment. This is an important issue in many conditions, but most noticeably in cystic fibrosis (CF). A large proportion of people with CF suffer from both chronic and intermittent infections, and in vitro, antibiotic susceptibility testing poorly correlates with patient treatment outcomes. S

Small cell lung cancer (SCLC) remains among the most aggressive thoracic malignancies and continues to present major therapeutic challenges despite advances in multimodality treatment. Limited-stage SCLC (LS-SCLC), representing approximately one-third of cases, is conventionally managed with concurrent chemoradiotherapy delivered with curative intent. Radiation therapy (RT) is central to treatment; however, rapid treatment related anatomical and volumetric changes may alter target geometry and reduce the accuracy of plans generated from baseline imaging. Adaptive RT (ART) has emerged as an ima

Pea (Pisum sativum L.), a major legume crop, is affected by various parasites including the pea aphid (Acyrthosiphon pisum Harris). The pea aphid is composed of multiple biotypes, each one being able to feed and reproduce on one or a few legume species. To understand the pea defense mechanisms to a pea adapted and a non-adapted A. pisum biotype, we studied the early molecular responses of four pea genotypes with contrasted levels of resistance, which are controlled primarily by the ApRVII locus. We found that major defense-related phytohormones and their derivatives in pea did not show clear r

Cytomegaloviruses (CMVs) are host species-specific and have adapted to their respective mammalian hosts during co-evolution. Host-adaptation is reflected by "private genes" that have specialized in mediating virus-host interplay and have no sequence homologs in other CMV species, although biological convergence has led to analogous protein functions. They are mostly organized in gene families evolved by gene duplications and subsequent mutations. The host immune response to infection, both the innate and the adaptive immune response, is a driver of viral evolution, resulting in the acquisition

The deep sea, which encompasses the largest habitat on Earth, presents a set of extreme and unique environmental conditions, including high hydrostatic pressure, near-freezing temperatures, and perpetual darkness. These conditions pose significant challenges to the survival and energy management of its inhabitants. Deep-sea organisms have evolved a range of bioenergetic adaptations to negotiate these harsh conditions, ensuring efficient energy acquisition and utilization. This review examines the multifaceted strategies employed by deep-sea animals, focusing on three key areas: energy input, d

Coxsackievirus A6 (CVA6) is a key pathogen causing hand, foot and mouth disease (HFMD). However, there are currently no specific antiviral drugs or vaccines for treating infections caused by CVA6. In this study, human rhabdomyosarcoma (RD), African green monkey kidney (Vero), and human embryonic lung diploid fibroblast (KMB17) cells were used to isolate CVA6 from 327 anal swab and fecal samples obtained during HFMD monitoring between 2009 and 2017. The VP1 genes of the isolates were sequenced and genotyped, and the biological characteristics of the representative CVA6 strains were analyzed. A

Minimal residual disease (MRD) has emerged as a central biomarker in hematologic malignancies, enabling highly sensitive detection of tumor persistence beyond conventional morphologic assessment and serving as an increasingly important surrogate endpoint in clinical trials. Despite these advances, MRD remains predominantly conceptualized as a quantitative variable reflecting residual tumor burden below assay detection thresholds. While this paradigm has enabled standardization of response criteria and cross-trial comparisons, it does not fully explain key clinical observations, including heter

BACKGROUND: Restless legs syndrome (RLS) is a common condition affecting 3% of the world's population. Pharmacological treatments do not cure RLS, and symptoms substantially affect everyday life. However, knowledge of biopsychosocial (BPS) factors influencing patients' everyday lives remains limited. This study aimed to explore and describe BPS barriers and facilitators influencing everyday life among patients with RLS.
METHODS: A deductive, descriptive qualitative design grounded in the BPS model was employed. Semi-structured telephone interviews were conducted with 28 strategically selected

Chronic obstructive pulmonary disease (COPD) is increasingly recognised as a systemic disorder that extends beyond the lungs, representing a clinical model of accelerated ageing. Frailty, a multidimensional syndrome characterised by reduced physiological reserve and increased vulnerability to stressors, is highly prevalent among individuals with COPD and contributes to adverse outcomes. Both conditions share common biological mechanisms, including chronic inflammation, oxidative stress, endocrine dysregulation, mitochondrial dysfunction and skeletal muscle atrophy. Exercise acts as a physiolog

The interactions between a host and microbe drive the health and disease status of the host. Of importance is the cause of dysbiosis in the presence of a pathogen, and critically, the relationship between the host and pathogen may evolve over time through response and adaptation. For immunocompromised individuals, dual infections are prevalent and contribute to disease severity and treatment options. Here, we explore the global reprogramming of host cells in response to immediate and established microbial infections with the human fungal pathogen Cryptococcus neoformans and the nosocomial bact

Efforts to understand biological functions and develop management schemes specific to Bos indicus-influenced cattle raised in tropical and subtropical environments are critical to meet the increasing global demand for protein. In the United States, B. indicus breeds are mostly used to generate B. indicus × B. taurus crosses with increased thermal and parasite tolerance, while retaining some productive characteristics of B. taurus cattle. Although crossbreeding represents a proven strategy to improve cattle adaptation almost immediately, research has also attempted to identify B. taurus genetic

Moulting is fundamental to decapod growth and development, yet its molecular regulatory mechanisms and relationship with environmental adaptive evolution remain incompletely elucidated. Through comparative genomic and transcriptomic analyses, we identified six cuticle-associated gene families (CP8, CP AMP1A, pro-resilin, CP AM1199, CP575, and CP1246) that underwent significant expansion in decapods, displaying species-specific distribution patterns. Notably, the CP1246 family, specifically expanded in crabs and enriched with calcification-related cuticle domains, likely represents a key evolut

Tuberculosis is the leading cause of death worldwide from a single infectious agent; it has also been declared a threat to humanity by the World Health Organization. New insights indicate that the innate immune response plays a crucial role in determining the outcome of the infection. In this study, we assessed the role of macrophages in the innate immune response through a simple mathematical model. Our results confirm that macrophages provide the primary protective response against Mycobacterium tuberculosis. However, they also highlight the importance of other innate cells in the outcome of

Escherichia coli is a commensal of the intestine of most mammals, but also an important human pathogen. Within a healthy human its population structure is highly dynamic, where typically a dominant E. coli strain is accompanied by several low abundance satellite strains. However, the factors underlying E. coli strain dynamics and evolution within hosts are still poorly understood. Here, we colonised germ-free immune-competent (wild-type) or immune-compromised (Rag2KO) mice, with two phylogenetically distinct strains of E. coli, to determine if strain co-existence and within-strain evolution ar

Single neurons function along a spectrum of neuronal operating modes whose properties determine how the output firing activity is generated from synaptic input. The auditory brain stem contains a diversity of neurons, from pure coincidence detectors to pure integrators and those with intermediate properties. We investigated how intrinsic spike initiation mechanisms regulate neuronal operating mode in the avian cochlear nucleus. Although the neurons in one division of the avian cochlear nucleus, nucleus magnocellularis, have been studied in depth, the spike threshold dynamics of the tonically f

Aneuploidy is common both in tumor cells responding to chemotherapeutic agents and in fungal cells adapting to antifungal drugs. Because aneuploidy simultaneously affects many genes, it has the potential to confer multiple phenotypes to the same cells. Here, we analyzed the mechanisms by which Candida albicans, the most prevalent human fungal pathogen, acquires the ability to survive both chemotherapeutic agents and antifungal drugs. Strikingly, adaptation to both types of drugs was accompanied by the acquisition of specific whole-chromosome aneuploidies, with some aneuploid karyotypes recover

We show that genetic recombination can be a powerful mechanism for escaping suboptimal peaks. Recent studies of empirical fitness landscapes reveal complex gene interactions and multiple peaks. However, classical work on recombination largely ignores the effect of complex gene interactions. Briefly, we restrict to fitness landscapes where the global peak is difficult to access. If the optimal genotype can be generated by shuffling genes present in the population, then recombination will produce the genotype. If, in addition, recombination is sufficiently rare, then the proportion of the genoty

Natural model systems evolving under extreme environmental pressures provide the opportunity to advance our knowledge of how the craniofacial complex evolves in nature. Unlike traditional models, natural systems are less inbred, and, therefore, better model the complex variation of the human population. Owing to the nature of certain craniofacial aberrations in blind Mexican cavefish, we suggest that this organism can provide new insights to a variety of craniofacial changes. Diverse cranial features have evolved in natural cave-dwelling Astyanax fish, which have thrived in the unforgiving dar

In this paper we analyse the possibility that the early hominin Ardipithecus ramidus had vocal capabilities far exceeding those of any extant non-human primate. We argue that erect posture combined with changes in craniofacial morphology, such as reduced facial and jaw length, not only provide evidence for increased levels of pro-sociality, but also increased vocal ability. Reduced length of the face and jaw, combined with a flexed cranial base, suggests the larynx in this species was situated deeper in the neck than in chimpanzees, a trait which may have facilitated increased vocal ability. UNLABELLED: Infections with Sudan virus (SUDV), a member of the genus Ebolavirus, result in a severe hemorrhagic fever with a fatal outcome in over 50% of human cases. The paucity of prophylactics and therapeutics against SUDV is attributed to the lack of a small-animal model to screen promising compounds. By repeatedly passaging SUDV within the livers and spleens of guinea pigs in vivo, a guinea pig-adapted SUDV variant (SUDV-GA) uniformly lethal to these animals, with a 50% lethal dose (LD50) of 5.3 × 10(-2) 50% tissue culture infective doses (TCID50), was developed. Animals infected with SU
