Spinal cord injury-induced expression of TrkA, TrkB, phosphorylated CREB, and c-Jun in rat lumbosacral dorsal root ganglia.

Previous studies have demonstrated increased expression and phosphorylation of tyrosine kinase receptor (TrkA, TrkB) in lumbosacral DRG after chronic (6 weeks) spinal cord (T8-T10) injury. This study examined the effects of acute SCI (48 hours, 2 weeks) on TrkA and TrkB expression and phosphorylation, and CREB and c-Jun expression in DRG. A significant increase in the number of TrkA- (1.5-3-fold; P < or = 0.05), TrkB- (1.3-2.0-fold; P < or = 0.05), and phosphorylated Trk (pTrk)-immunoreactive (1.5-3-fold; P < or = 0.05) cells was observed in the L1, L6, and S1 DRG 48 hours, 2, or 6 weeks after

Distribution of trigeminohypothalamic and spinohypothalamic tract neurons displaying substance P receptor-like immunoreactivity in the rat.

Primary afferent neurons containing substance P (SP) are apparently implicated in the transmission of noxious information from the periphery to the central nervous system, and SP released from primary afferent neurons acts on second-order neurons with the SP receptor (SPR). In the rat, nociceptive information reached the hypothalamus not only through indirect pathways but also directly through trigeminohypothalamic and spinohypothalamic pathways. Thus, in the present study, the distribution pattern of trigeminohypothalamic and spinohypothalamic tract neurons showing SPR-like immunoreactivity (

Anti-inflammatory properties of the vagus nerve: potential therapeutic implications of vagus nerve stimulation.

Brain and viscera interplay within the autonomic nervous system where the vagus nerve (VN), containing approximately 80% afferent and 20% efferent fibres, plays multiple key roles in the homeostatic regulations of visceral functions. Recent data have suggested the anti-inflammatory role of the VN. This vagal function is mediated through several pathways, some of them still debated. The first one is the anti-inflammatory hypothalamic-pituitary-adrenal axis which is stimulated by vagal afferent fibres and leads to the release of cortisol by the adrenal glands. The second one, called the choliner

CNS sites activated by renal pelvic epithelial sodium channels (ENaCs) in response to hypertonic saline in awake rats.

In some patients, renal nerve denervation has been reported to be an effective treatment for essential hypertension. Considerable evidence suggests that afferent renal nerves (ARN) and sodium balance play important roles in the development and maintenance of high blood pressure. ARN are sensitive to sodium concentrations in the renal pelvis. To better understand the role of ARN, we infused isotonic or hypertonic NaCl (308 or 500mOsm) into the left renal pelvis of conscious rats for two 2hours while recording arterial pressure and heart rate. Subsequently, brain tissue was analyzed for immunohi

Sensory innervation of the rat kidney and ureter as revealed by the anterograde transport of wheat germ agglutinin-horseradish peroxidase (WGA-HRP) from dorsal root ganglia.

The sensory innervation of the rat kidney and ureter was investigated in wholemount preparations and sectioned materials by labeling the afferent nerve fibers with wheat germ agglutinin-horseradish peroxidase (WGA-HRP) transported anterogradely from dorsal root ganglia. Labeled fibers were seen in large numbers in the ureter and in the lining of the renal pelvis, where they were located in the adventitia, smooth muscle, subepithelial connective tissue, and epithelium. Most of the fibers in the ureter and ureteropelvic junctional zone traveled parallel to the long axis of the organ. In contrast

Closed-loop coupling of both physiological spindle model and spinal pathways for sensorimotor control of human center-out reaching.

The development of new studies that consider different structures of the hierarchical sensorimotor control system is essential to enable a more holistic understanding about movement. The incorporation of more biological proprioceptive and neuronal circuit models to muscles can turn neuromusculoskeletal systems more appropriate to investigate and elucidate motor control. Specifically, further studies that consider the closed-loop between proprioception and central nervous system may allow to better understand the yet open question about the importance of afferent feedback for sensorimotor learn

Regulation of vasopressin release by co-released neurotransmitters: mechanisms of purinergic and adrenergic synergism.

Arginine vasopressin (AVP) neurons of the hypothalamo-neurohypophseal system (HNS) are innervated by numerous afferent pathways carrying information about two physiologically important parameters: blood volume/pressure and osmolality. These pathways use a variety of neurotransmitters/neuropeptides. In order to understand normal and pathological regulation of VP secretion, the mechanisms underlying integration of these complex afferent signals by the AVP neurons must be understood. The importance of neurotransmitter interactions in determining hormone release is highlighted by the finding that …

The gut feeling in motion sickness.

Motion sickness is a nearly universal response to the unnatural motions that are experienced when traveling by means other than the body's own faculties; in artificial realities; and in micro- and partial gravity environments. Despite being a known malady since ancient times, its underlying mechanisms, as well as the marked interpersonal variability in susceptibility, remain incompletely understood. While efferent brain-to-body signaling pathways involved in motion sickness have been previously described, recent findings on the functional role of the gut's (neuro)epithelial cells and microbiom

Plasticity of the afferent innervation of the airways: the role of ion channels.

Neuronal pathways associated with cough exhibit remarkable plasticity that can result in a persistent and uncontrollable urge to cough during disease. Afferent neurones involved in detecting tussive stimuli are polymodal, i.e. they respond to several types of stimuli including acid, inflammatory mediators such as bradykinin and mechanical stimuli. The pattern of action potential discharge following the encounter of the nerve terminal with a tussive stimulus is likely to determine the magnitude of the urge to cough and cough itself. The discharge pattern in sensory neurones is determined by mul

Three Rostromedial Tegmental Afferents Drive Triply Dissociable Aspects of Punishment Learning and Aversive Valence Encoding.

Persistence of reward seeking despite punishment or other negative consequences is a defining feature of mania and addiction, and numerous brain regions have been implicated in such punishment learning, but in disparate ways that are difficult to reconcile. We now show that the ability of an aversive punisher to inhibit reward seeking depends on coordinated activity of three distinct afferents to the rostromedial tegmental nucleus (RMTg) arising from cortex, brainstem, and habenula that drive triply dissociable RMTg responses to aversive cues, outcomes, and prediction errors, respectively. Cortical projections to the superior colliculus in prosimian galagos (Otolemur garnetti).

The superior colliculus (SC) is a key structure within the extrageniculate pathway of visual information to cortex and is highly involved in visuomotor functions. Previous studies in anthropoid primates have shown that superficial layers of the SC receive direct inputs from various visual cortical areas such as V1, V2, and middle temporal (MT), while deeper layers receive direct inputs from visuomotor cortical areas within the posterior parietal cortex and the frontal eye fields. Very little is known, however, about the corticotectal projections in prosimian primates. In the current study we i

The anatomy and physiology of the ocular motor system.

Accurate diagnosis of abnormal eye movements depends upon knowledge of the purpose, properties, and neural substrate of distinct functional classes of eye movement. Here, we summarize current concepts of the anatomy of eye movement control. Our approach is bottom-up, starting with the extraocular muscles and their innervation by the cranial nerves. Second, we summarize the neural circuits in the pons underlying horizontal gaze control, and the midbrain connections that coordinate vertical and torsional movements. Third, the role of the cerebellum in governing and optimizing eye movements is pr

Vertical alignment in monkeys with unilateral IV section: effects of prolonged monocular patching and trigeminal deafferentation.

We investigated monocular viewing and trigeminal (V) deafferentation on the vertical deviation (VD) in monkeys following intracranial IV section. Two monkeys wore a patch for four to six weeks, one over the paretic eye and the other over the normal eye following IV section. Two other monkeys had combined IV and V section with the paretic eye patched postlesion. In monkeys with IV section alone, the VD lessened within the first week postlesion but then increased gradually with the same eye still patched. Thus binocular viewing was unnecessary for the later VD increase. With combined IV and V se

Transient urinary retention in acute right lateral medullary infarction.

Urinary disturbance has rarely been reported in patients with a medullary lesion. We evaluated a patient with acute infarction in the right lateral medulla who had voiding difficulty as an initial manifestation. Urodynamic study showed detrusor areflexia on voiding, and the voiding symptom completely recovered 2 weeks after the stroke onset. The urinary retention in this patient might be caused by interruption of descending fibers from facilitatory pontine tegmentum micturition centers.

Neurophysiology of Rett syndrome.

Rett syndrome is a neurodevelopmental disorder that in most cases is consequent to a mutation in the MECP2 gene. The central nervous system is the primary organ system involved in Rett syndrome. Neurophysiologic evaluations provide information concerning the developmental aspects of Rett syndrome and the character and extent of involvement of the central, peripheral, and autonomic nervous system pathways. Evoked potentials typically demonstrate intactness of peripheral auditory and visual pathways and suggest dysfunction of central or "higher" cortical pathways. Somatosensory evoked potentials

Trigeminocardiac reflex. A clinical phenomenon or a new physiological entity?

The trigemino-cardiac reflex (TCR) is defined as the sudden onset of parasympathetic dysrhythmia, sympathetic hypotension, apnea or gastric hypermotility during stimulation of any of the sensory branches of the trigeminal nerve. The sensory nerve endings of the trigeminal nerve send neuronal signals via the Gasserian ganglion to the sensory nucleus of the trigeminal nerve, forming the afferent pathway of the reflex arc. This afferent pathway continues along the short internuncial nerve fibers in the reticular formatio to connect with the efferent pathway in the motor nucleus of the vagus nerve

Target tissue influences the peripheral trajectory of mouse primary sensory olfactory axons.

Primary olfactory neurons situated in the nasal septum project axons within fascicles along a highly stereotypical trajectory en route to the olfactory bulb. The ventral fascicles make a distinct dorsovental turn at the rear of the septum so as to reach the olfactory bulb. In the present study we have used a brain and nasal septum coculture system to examine the role of target tissue on the peripheral trajectory of olfactory sensory axons. In cultures of isolated embryonic nasal septa, olfactory axons form numerous parallel fascicles that project caudally in the submucosa, as they do in vivo.

Modulation of cochlear afferent response by the lateral olivocochlear system: activation via electrical stimulation of the inferior colliculus.

The olivocochlear (OC) efferent innervation of the mammalian inner ear consists of two subdivisions, medial (MOC) and lateral (LOC), with different peripheral terminations on outer hair cells and cochlear afferent terminals, respectively. The cochlear effects of electrically activating MOC efferents are well known, i.e., response suppression effected by reducing outer hair cells' contribution to cochlear amplification. LOC peripheral effects are unknown, because their unmyelinated axons are difficult to electrically stimulate. Here, stimulating electrodes are placed in the inferior colliculus

Small-fiber dysfunction in trigeminal neuralgia: carbamazepine effect on laser-evoked potentials.

BACKGROUND: In patients with trigeminal neuralgia, results of clinical examination of sensory function are normal. Reflex and evoked potential studies have already provided information on large-afferent (non-nociceptive) function. Using laser-evoked potentials (LEP), the authors sought information on small-afferent (nociceptive) function. METHODS: The brain potentials evoked by CO(2)-laser pulses directed to the perioral and supraorbital regions were studied in 67 patients with idiopathic or symptomatic trigeminal neuralgia and 30 normal subjects. Of the 67 patients, 49 were receiving carbama
