Protocol for the assessment of the impact of mycotoxins and glyphosate residues on the gut microbiome and resistome of European fallow deer.

Here, we present a protocol to describe the bacteriome of the intestinal content of toxin-exposed fallow deer. We describe steps for measuring fecal mycotoxin (deoxynivalenol, zearalenone, fumonisin B1, and aflatoxin B1) levels using liquid chromatography-mass spectrometry, as well as serum glyphosate. We then detail a short-read shotgun DNA sequencing-based bioinformatic pipeline for the toxin level-associated analysis of the bacteriome and resistome and the construction of metagenome-assembled bacterial genomes. This protocol has potential applications in further toxin level-associated metag

Recombinant AflQ: a novel fungal-derived protein with potent antibacterial properties.

Mycotoxins, especially aflatoxins produced by fungi of the genus Aspergillus, threaten global food security and public health due to their carcinogenic and immunosuppressive effects. To exploit Key enzymatic steps in the aflatoxin biosynthetic pathway for therapeutic applications, the aflQ gene (also Known ordA) -wich encodes acytochrome P450 monooxygenase capable of generating reactive oxidative species -was targeted as a candidate antimicrobial agent. This study aims to extract and characterize the aflQ gene (known as ordA) from the Aspergillus niger fungus isolated from grain silos in Anbar…

Public health strategies for hepatocellular carcinoma: from risk factors to prevention and control.

Hepatocellular carcinoma (HCC) remains a major global health burden, ranking as one of the leading causes of cancer-related deaths worldwide. This review synthesizes current evidence on HCC epidemiology, highlighting both modifiable and non-modifiable risk factors, including chronic hepatitis B and C virus infections, metabolic dysfunction-associated steatotic liver disease (MASLD), excessive alcohol consumption, aflatoxin exposure, and genetic susceptibility. These diverse etiologies reflect not only biological mechanisms but also broader social and environmental determinants of health, empha

Molecular characterization of hepatocarcinogenesis using mouse models.

Hepatocellular carcinoma (HCC) is a deadly disease, often unnoticed until the late stages, when treatment options become limited. Thus, there is a crucial need to identify biomarkers for early detection of developing HCC, as well as molecular pathways that would be amenable to therapeutic intervention. Although analysis of human HCC tissues and serum components may serve these purposes, inability of early detection also precludes possibilities of identification of biomarkers or pathways that are sequentially perturbed at earlier phases of disease progression. We have therefore explored the opt

Naturally occurring toxic substances in foods.

Numerous chemical toxins, including normal components of natural foods, e.g., mycotoxins, and toxic chemicals as contaminants such as pesticides, fertilizers, food additives, and preservatives, which are potentially toxic to humans, are discussed. Potential toxicity, the hazard to man represented by most of these chemicals, may be low because the concentration in food may be low. The gap in our knowledge of long-term effects makes rational decisions as to allowable levels of these substances a major problem. On the other hand, nitrosamines and aflatoxins are toxins for which there exists a vol…

Optimizing HCV Management Among Rohingya Refugees in Cox's Bazar, Bangladesh: Addressing Cofactors and Environmental Risks to Mitigate Long-Term Liver Disease Complications.

Hepatitis C virus (HCV) infection is increasingly reported among Rohingya refugees in Cox's Bazar, with active infection rates in adults reaching 20% by 2023. The risk of liver disease progression and hepatocellular carcinoma (HCC) may be amplified by coexisting factors such as chronic malnutrition, coinfections, aflatoxin exposure, metabolic disorders, and environmental toxins. Despite WHO-led efforts, data on these risk factors remain fragmented, and access to care is limited. This study underscores the need for comprehensive surveillance, epidemiological research, and long-term prevention s

Cholesterol gallstones and cancer of gallbladder (CAGB): molecular links.

There is a known association between cholesterol gallstones and cancer of gall bladder (CAGB). However, the exact relation is not clear. It is proposed they are linked at molecular level by the activity of the orphan nuclear receptors (ONRs) and ABC transporter pumps involved in cholesterol and xenobiotic efflux from the liver into bile. There is evidence that these two pathways are closely interlinked and influence each other. Genetic and environmental factors that upregulate these systems can lead to the simultaneous pumping of cholesterol (which precipitate as gallstones) and a food carcino

[Carcinogenic fungi (author's transl)].

An association of carcinogenicity of toxic substances with fungi is shortly documented and discussed. The actions of the aflatoxin B1 (AFB1) are particularly examined; but it is emphasized that other carcinogenic mycotoxins are more frequent and therefore more dangerous. The man is unable to live estranged from them, because they are a great part in ecological systems and in the work-place, resulting a potentially major threat. The range of food and feedstuffs potentially affected has been increased in recent years. In effect contamination is liable to occur during transport, storage and food

Divergent volatile metabolomes and flavor attributes in rice fermented by Aspergillus oryzae and Aspergillus flavus.

INTRODUCTION: The generally recognized as safe (GRAS) fungus Aspergillus oryzae has been used for millennia in the production of traditional Asian fermented foods and beverages. Domestication has led to genomic and phenotypic adaptations that distinguish A. oryzae from its wild relative, Aspergillus flavus. While differences between these species have been partially characterized, their comparative production of volatile compounds during food fermentation remains poorly understood. METHODS: We evaluated alpha-amylase activity, aflatoxin production, taste attributes using an electronic tongue,

Novel techniques for the mass production of nutritionally improved, fungus-treated lignocellulosic biomass for ruminant nutrition.

BACKGROUND: Laboratory-scale experiments have shown that treatment with selective lignin-degrading white-rot fungi improves the nutritional value and ruminal degradability of lignocellulosic biomass (LCB). However, the lack of effective field-applicable pasteurization methods has long been recognized as a major obstacle for scaling up the technique for fungal treatment of large quantities of LCB for animal feeding. In this study, wheat straw (an LCB substrate) was subjected to four field-applicable pasteurization methods - hot-water, formaldehyde fumigation, steam, and hydrated lime - and cult

Simultaneous detection of AFB1 and aflD gene by "Y" shaped aptamer fluorescent biosensor based on double quantum dots.

The developed method for simultaneous detection of aflatoxin B1 (AFB1) and aflD genes can effectively monitor from the source and reduce the safety problems and economic losses caused by the production of aflatoxin, which can be of great significance for food safety regulations. In this paper, we constructed a sensitive and convenient fluorescent biosensor to detect AFB1 and aflD genes simultaneously based on fluorescence resonance energy transfer (FRET) between quantum dots (QDs) and a black hole quenching agent. A stable "Y" shaped aptasensor was employed as the detection platform and a doub

Gut microbes define liver cancer risk in mice exposed to chemical and viral transgenic hepatocarcinogens.

BACKGROUND AND AIMS: Hepatocellular carcinoma (HCC) frequently results from synergism between chemical and infectious liver carcinogens. Worldwide, the highest incidence of HCC is in regions endemic for the foodborne contaminant aflatoxin B1 (AFB1) and hepatitis B virus (HBV) infection. Recently, gut microbes have been implicated in multisystemic diseases including obesity and diabetes. Here, the hypothesis that specific intestinal bacteria promote liver cancer was tested in chemical and viral transgenic mouse models. METHODS: Helicobacter-free C3H/HeN mice were inoculated with AFB1 and/or He
