Diastereoselective Synthesis of trans-β-Amino Cyclohexyl Ketones and trans-3-Amino-4-acyl-tetrahydropyrans from Intramolecular C-Alkylation of ω-Halo-Substituted β-Sulfinamido Ketones.

Reaction of ω-halo-substituted nonracemic β-sulfinamido ketones with NaH afforded the β-amino cyclohexyl ketones in excellent yields and diastereoselectivity, via an intramolecular C-alkylation, α to the carbonyl group. The reaction was found to be general and can be applied for the synthesis of different cyclohexyl amino ketones and tetrahydropyrans possessing amine and acyl substitutions.

Design of a Photoredox Catalyst that Enables the Direct Synthesis of Carbamate-Protected Primary Amines via Photoinduced, Copper-Catalyzed N-Alkylation Reactions of Unactivated Secondary Halides.

Despite the long history of SN2 reactions between nitrogen nucleophiles and alkyl electrophiles, many such substitution reactions remain out of reach. In recent years, efforts to develop transition-metal catalysts to address this deficiency have begun to emerge. In this report, we address the challenge of coupling a carbamate nucleophile with an unactivated secondary alkyl electrophile to generate a substituted carbamate, a process that has not been achieved effectively in the absence of a catalyst; the product carbamates can serve as useful intermediates in organic synthesis as well as bioact

Alkylating Agent-Induced FANCL Impairment Damages Pulmonary Arterial, Venous, and Microvascular Endothelial Cells and Predisposes to PAH/PVOD.

Alkylating chemotherapies such as mitomycin C (MMC) and cisplatin (CDDP) are associated with pulmonary arterial hypertension (PAH) with pulmonary veno-occlusive disease (PVOD) features, yet the underlying mechanisms remain unclear. Using established MMC and newly developed CDDP rat models, we found that both agents induced PAH/PVOD, as evidenced by elevated right ventricular systolic pressure, pulmonary arterial, venous and microvascular remodeling, and right ventricular dysfunction, with MMC causing more severe pathology. In cultured pulmonary arterial, venous, and microvascular endothelial c

Ring-opening decarbonylative C(sp3)-C(sp3) cross-electrophile coupling of cyclic imides with unactivated alkyl chlorides.

Herein we report a nickel-mediated decarbonylative cross-electrophile coupling of N-Boc succinimides and glutarimides with unactivated alkyl chlorides. The transformation proceeds via selective endocyclic N-C(O) activation, which opens a new entry point into C(sp3)-C(sp3) cross-electrophile coupling and, through incorporation of the ring-opened imide scaffold, establishes a highly modular platform to rapidly build molecular complexity. In situ halide exchange enables the use of abundant alkyl chlorides, while broad functional group tolerance grants access to structurally diverse α- and β-subst

Distinct Source-Sink Patterns and Vertical Consumption of Alkyl and Aryl Organophosphate Esters in the Remote Ocean and Its Marginal Sea.

Organophosphorus esters (OPEs) have been widely used as flame retardants and plasticizers and are globally ubiquitous. In the marine environment, OPEs primarily originate from continental sources and are initially confined to marginal seas before reaching remote oceans. However, their differing biogeochemical fates in these two regions remain unclear. This study investigated regional variations in the biogeochemical regulation mechanisms of OPEs (alkyl and aryl) across the Western Pacific and the South China Sea (112°E to 163°E) by analyzing their air-sea interface processes and vertical profi

Ultrafast Photochemical Reaction Dynamics of a Cyclic (Alkyl)(Amino)Carbene-Carbon Disulfide Dimer Probed by Femtosecond Infrared Spectroscopy.

The ultrafast photochemical reaction dynamics of a cyclic(alkyl)(amino)carbene-carbon disulfide (CAAC-CS2) dimer containing two adjacent S-S bonds were investigated using femtosecond time-resolved infrared spectroscopy in combination with multireference electronic structure calculations. Time-resolved vibrational spectra and global kinetic analysis reveal that photoexcitation of the S-S n → σ* transition at 375 nm induces subpicosecond (<0.3 ps) homolytic cleavage of one S-S bond, generating a bis-thiyl diradical intermediate. This intermediate undergoes two competing pathways: recombination t

P-Directed and Lewis Acid-Assisted Pd-Catalyzed Si─Me Bond Activation Enabling Alkyl Exchange at Silicon.

A novel mode of Si─Me bond activation has been unveiled through the cooperative action of a P-directing group, a palladium catalyst and a Lewis acid. The pivotal role of the Lewis acid in enabling the activation of the strong and low-polarity Si─Me bond was elucidated via stoichiometric experiments. According to DFT calculations, the Lewis acid acts as a Me- acceptor and triggers the formation of the key cationic silyl Pd complex responsible for the alkyl exchange at Si. This unprecedented Si─Me bond transformation has been demonstrated in a four-step sequence, enabling the stepwise upgrading

SAR Studies around the SULT1A1-Activated Alkylator YC-1 as Cytotoxic Agents against Biliary Tract Cancer Cells.

A quantitative high-throughput screen using biliary tract cancer cell lines had identified the small-molecule YC-1 as being selectively cytotoxic against the IDH1-mutant cell lines with high expression of SULT1A1. We discuss the structure-activity relationship study of YC-1 analogs and identify the key structural motifs that are essential for this cytotoxicity. We highlight the narrow SAR around the furfuryl alcohol that has been reported as a critical motif that is activated to a reactive electrophile by the sulfotransferase enzyme SULT1A1. Drug-like properties of key analogs are evaluated, i

Dynamic and structural insights into hydrogen-bonded interpolymer complexes of poly(2-alkyl-2-oxazolines) with poly(carboxylic acids).

The formation of interpolymer complexes (IPCs) between poly(2-alkyl-2-oxazolines) (PAOx) and poly(methacrylic acid) (PMAA) in solutions has been investigated using polarized luminescence relaxation and small-angle X-ray scattering (SAXS) structural methods. The results reveal that hydrophobic interactions contribute to the formation of IPCs in both aqueous and methanol solutions, influenced by the length of the side group in PAOx. The relaxation times, which can be used to quantify the intramolecular mobility of the luminescently labeled poly(methacrylic acid) (PMAA*), increase from 80 to 680

Physical characterization of PEGylated exosome constructs: Size, charge, and morphology changes in non-specific alkylating N-terminal reactions.

Small extracellular vesicles, commonly referred to as exosomes, withhold a promising future in the pharmaceutical industry as carriers for targeted drug delivery due to their high specificity and bioavailability when compared to synthetic-based vectors. They, however, present some limitations for systematic administration because of natural organism defenses and their high-water solubility, ultimately making it difficult for them to reach the intended target. To improve the delivery capacity of these nanoparticles, the possibility for the construction of PEGylated versions was explored in this

Synthesis of γ-Lactams via Palladium-Catalyzed C(sp3)-H Bond Activation of Alkyl Sulfonamides with Substituted Alkenes.

A methodology for the γ-butyrolactam scaffolds via ligand-enabled C(sp3)-H bond functionalization of sulfonamides with olefins has been demonstrated. The protocol has been found to be compatible with several activated and unactivated olefins, and the desired lactams were formed in excellent yields. A plausible mechanism has been described to account for the desired lactamization reaction as well as supported by mechanistic investigation including a 1H NMR study and isolation of a palladacycle intermediate.

C(sp3)-N Coupling of Nonactivated Alkyl Boronic Pinacol Esters by the Merger of Amino Radical Transfer and Copper Catalysis.

Metal-catalyzed C(sp3)-N coupling remains a formidable synthetic challenge, especially with alkyl electrophiles, due to competing pathways such as slow oxidative addition, β-hydride elimination, and alkyl isomerization. While alkyl halides are commonly used, their limitations have spurred interest in alternative coupling partners. Alkyl boronic pinacol esters (Bpins), though stable and widely accessible, are underutilized in C-N coupling due to their poor transmetalation reactivity and susceptibility to protodeborylation and isomerization. Here, we report a dual catalytic strategy that merges

Copper(I)-free syntheses of [11C/18F]trifluoromethyl ketones from alkyl or aryl esters and [11C/18F]fluoroform.

Herein, we report a copper(I)-free method for labeling the trifluoroacetyl group with positron-emitting carbon-11 (t1/2 = 20.4 min) or fluorine-18 (t1/2 = 109.8 min) as part of our exploration of radiolabeled fluoroforms to access new radiolabeled chemotypes of interest for tracer development. Treatment of alkyl esters and aryl esters, containing electron-donating or electron-withdrawing groups, with [11C/18F]fluoroform in the presence of strong base, gave [11C/18F]trifluoromethyl ketones as novel radiolabeling synthons in moderate to high yields within 15 minutes.

Catalytic Aerobic Carbooximation of Alkenes Using Secondary Nitroalkanes as Both α-Nitro Alkyl Radical and Nitrogen Monoxide Sources.

Aerobic nitro-nitrite isomerization of secondary nitroalkanes is postulated to proceed via the intermediacy of the α-nitro alkyl radical, where the corresponding Nef-type products, ketones, and nitrogen monoxide can be obtained as byproducts. To explore the catalytic aerobic carbooximation of alkenes using secondary nitroalkanes, phase-transfer catalysis of KSeCN and TBAI has been developed. The current aerobic carbooximation of alkenes utilizes nitroalkanes as both radical and nitrogen monoxide sources in water without external oxidants and prefunctionalized nitroalkanes.

Air and Water Stable Bicyclic (Alkyl)(Amino)Carbene Stabilized Phosphenium Cation: Reactivity and Selective Fluoride Ion Affinity.

The synthesis and reactivity of an air and water stable Bicyclic (alkyl)(amino)carbene (BICAAC) stabilized phosphenium cation (1) is reported. Air and water stable phosphenium cation are rare in the literature. Compound 1 is obtained by reaction of BICAAC with Ph2PCl in THF followed by anion exchange with LiOTf. The reduction and oxidation of 1 yielded corresponding α-radical phosphine species (2) and BICAAC stabilized phosphenium oxide (3) respectively. All compounds are well characterized by single crystal X-ray diffraction studies. The Lewis acidity of compounds 1 and 3 are determined by co

Visible-Light-Induced Difunctionalization of Alkenyl Ketones with α-Carbonyl Alkyl Bromide: Concomitant Installation of C-C Bonds.

The visible-light-promoted difunctionalization of alkenyl ketones has been developed for easy access of various tetralones, cyclopropane, or alkenyl migration compounds. With fac-[Ir(ppy)3] as the photocatalyst, alkenyl ketones captured the α-carbonyl alkyl radical and evolved through intramolecular cyclization and the elimination of a proton to give the difunctionalized products. This strategy is characterized by good yields, mild reaction conditions, and outstanding functional group tolerance.
