We report the first study in non-human primates of the safety and immunogenicity of a recombinant vaccine designed to prevent amebic liver abscess. In a pilot study, a recombinant vaccine containing the serine rich Entamoeba histolytica protein (SREHP) attached to a maltose binding protein (SREHP/MBP), which has been shown to be effective in preventing amebic liver abscess in rodent models of infection, was used to immunize two African Green Monkeys. Of 115 patients with symptomatic intestinal amebiasis, 56 were treated with Ro 7-0207 and 59 with metronidazole in a double blind study. Results of treatment were similar in the two groups and side effects were of low intensity except in one patient who received Ro 7-0207 and developed numbness of hands and tongue, difficulty in speaking, and headache. These symptoms disappeared after termination of treatment.

Results are reported of antibodies to E. histolytica antigen obtained by enzyme immunoassay, complement fixation, indirect hemagglutination and latex agglutination in sera of persons with varying exposure to infection by E. histolytica. The data are compared to findings in non-exposed persons and in patients with amebic liver abscess. From the results it is concluded that the value of serodiagnosis in amebiasis is higher in patients, who have traveled in tropical areas for a limited period, than in residents of the tropics.
