The DZHK Feasibility Explorer: Supporting Cohort Discovery and Feasibility Assessment for Cardiovascular Research.

INTRODUCTION: The secondary use of clinical study data and biological samples holds substantial potential for advancing biomedical research. The German Centre for Cardiovascular Research (DZHK) research platform made the secondary use of study data possible through standardized data collections and centralized data management. However, identifying suitable datasets and estimating cohort sizes prior to formal data access requests remains challenging due to restricted access to data and metadata. To address this gap, we developed the Feasibility Explorer (FE), a privacy-preserving tool that enab

A 1024-Channel 0.8V 23.9-nW/Channel Event-Based Compute In-Memory Neural Spike Detector.

The increasing data rate has become a major issue confronting next-generation intracortical brain-machine interfaces (iBMIs). The scaling number of recording sites requires complex analog wiring and lead to huge digitization power consumption. Compressive event-based neural frontends have been used in high-density neural implants to support the simultaneous recording of more channels. Event-based frontends (EBF) convert recorded signals into asynchronous digital events via delta modulation and can inherently achieve considerable compression. But EBFs are prone to false events that do not corre

Catalytic co-pyrolysis of polylactic acid and high-density polyethylene over hierarchical zeolite catalysts.

Catalytic fast co-pyrolysis of polylactic acid (PLA) and high-density polyethylene (HDPE) was investigated using a tandem micro-reactor/gas chromatogram spectrometry (TMR-GC/MS) to selectively produce BTEX (benzene, toluene, ethylbenzene, and xylene). Non-catalytic co-pyrolysis revealed strong synergistic suppression of oxygenated products, which was most pronounced at a PLA/HDPE mass ratio of 0.75/0.25, originating from efficient hydrogen transfer between oxygen-rich PLA-derived intermediates and hydrogen-rich HDPE-derived species. To enhance deoxygenation and aromatization, hierarchical HZSM

Characterization of archaeal promoters using explainable and web-based CNN model.

BACKGROUND: Archaea, the third realm of life, shows unique genetic and metabolic adaptations that enable its survival in harsh environments. Despite its biotechnological and evolutionary importance, limited research has been done on archaeal promoter prediction compared to bacteria and eukaryotes. Current tools have shortcomings such as relying solely on DNA duplex stability feature encoding, high false-positive rates, low precision and lack of publicly accessible archaeal promoter prediction tool. RESULTS: To overcome these limitations, we present “iProm-Archaea”, a CNN-based tool for precise

Single-nucleus multi-omics of Parkinson's disease reveals a glutamatergic neuronal subtype susceptible to gene dysregulation via alteration of transcriptional networks.

The genetic architecture of Parkinson's disease (PD) is complex and multiple brain cell subtypes are involved in the neuropathological progression of the disease. Here we aimed to advance our understanding of PD genetic complexity at a cell subtype precision level. Using parallel single-nucleus (sn)RNA-seq and snATAC-seq analyses we simultaneously profiled the transcriptomic and chromatin accessibility landscapes in temporal cortex tissues from 12 PD compared to 12 control subjects at a granular single cell resolution. An integrative bioinformatic pipeline was developed and applied for the ana

Interfacial Coordination and Dimensional Confinement Enable Continuous Thin MOF-on-MOF Membranes for Ion Sieving.

Metal-organic frameworks (MOFs) with crystallographically defined pores and chemically programmable internal structures are attractive candidates for regulating ion transport at the molecular scale. Yet, it is still difficult to grow MOF-on-MOF structures as continuous thin membranes because the secondary phase usually forms either thick coatings or discontinuous particles, both of which hinder ion transport. In this work, we found that Cu-O-Zr coordination at the interface can guide UiO-66 to grow continuously on the CuOx-BDC scaffold even under severe lattice mismatch, producing a thin integ

Photonic-Crystal-Integrated Plasmonic Hydrogel Microcylinder Arrays for Enhanced and Fouling-Resistant SERS.

Surface-enhanced Raman scattering (SERS) enables highly sensitive molecular identification but can be compromised by plasmonic-surface fouling and inefficient utilization of excitation light. Here, we report a strategy that combines molecular-size-selective transport with spectrally selective excitation recycling to enhance SERS sensitivity while resisting surface fouling. A colloidal photonic crystal (PC) reflector was fabricated by shear-assisted casting of monodisperse silica particles in a photocurable medium, with its stopband tuned to the 633 nm excitation wavelength by controlling parti

Phase-Engineered MoS2 Quantum Monolayer Catalysts.

Single-atom catalysts with 100% atomic efficiency represent a frontier in heterogeneous catalysis, yet they face low loading density of active species (normally < 5.0 wt.%), poor stability, and difficulties in low-cost synthesis. Here, we present a new type of catalyst model-phase-engineered quantum-monolayer catalyst, as a complementary catalyst architecture that offers high-density accessible atoms and enables versatile phase/interphase engineering for catalysis. As a prototype (in our 13-type quantum monolayer library), our fabricated MoS2 quantum-monolayer catalysts are one molecule-layer

Conformational switch of NPC1 epitomizes a lumen-to-membrane alternating access model for nonpolar substrates.

Niemann-Pick disease type C (NPC) arises from failure of the NPC1 and/or NPC2 proteins to mobilize lysosomal cholesterol, yet the mechanism by which the complex drives sterol translocation lacks a thorough understanding. Here, we capture an NPC1-NPC2 complex conformation at lysosomal pH using a bis-sterol ligand (JM046), fortifying NPC2 engagement with NPC1 and enabling structural characterization of an alternative transport state. In this complex, the sterol moiety is absent from the NPC1 central tunnel, and the neck site adopts an expanded architecture that resembles a previously observed NP

High-performance stretchable conductive film on complex microscale structures via spontaneous fusion of liquid metal.

Conformal fabrication of stretchable conductive film on micro-structured surfaces is important for flexible electronics. Existing methods based on material design and structural engineering suffer from limitations in stretchability, conductivity, and geometrical compatibility. Here, we propose a universal strategy for fabricating high-performance stretchable conductive film on complex micro-structured surfaces. By precisely controlling the interparticle liquid bridge force and the critical rupture strength of liquid metal (LM) particle shells, the spontaneous fusion of LM particles is realized

miniMTI: minimal multiplex tissue imaging enhances biomarker expression prediction from histology.

Virtual multiplexing from routine histology has advanced rapidly, yet morphology alone provides limited access to molecular state, imposing an intrinsic ceiling on H&E-only inference. Here, we introduce miniMTI, a molecularly anchored virtual staining framework that determines the minimal set of experimentally measured markers required, alongside H&E, to accurately reconstruct large multiplex tissue imaging (MTI) panels while preserving biologically and clinically relevant information. miniMTI learns from paired same-section H&E-MTI data using a unified multimodal generative model that can con

Room-Temperature Tuning and Probing of Fermi Polarons in Atomically Thin Semiconductors on a Plasmonic Metasurface.

The Fermi polaron, arising from interactions between a mobile impurity and a degenerate Fermi sea, is a many-body quasiparticle that provides a sensitive probe of strongly correlated electronic phases in atomically thin semiconductors. In doped transition-metal dichalcogenides, the attractive and repulsive polaron branches are well established in monolayers. However, extending active control and quantitative, branch-resolved probing to stacked geometries has remained elusive because spectral quenching and weak optical contrast restrict access to Fermi polaron signatures. Here, we integrate ele

Plasmonic trimer nanoarrays with probe-trapping sites for SERS detection of urinary copper in Wilson's disease.

Cu2+ plays a critical role in the onset of Wilson's disease (WD), and precise analysis of Cu2+ levels in human metabolites is essential for early clinical diagnosis. However, existing clinical detection such as organ biopsies and mass spectrometry are costly, complex and unsuitable for rapid screening. Surface-enhanced Raman scattering (SERS) offers a rapid, label-free, and noninvasive alternative to conventional assays. Nevertheless, SERS fundamentally depends on highly localized electromagnetic "hot spots", which occupy only a very small fraction of the nanoparticle surface (less than 1%), m

An open-source 3D printing system enabling in-situ freeze-thaw processing of hydrogels.

Hydrogels are widely used in tissue engineering and scaffold fabrication due to their excellent biocompatibility, while 3D printing excels at creating complex architectures. Among various techniques, low-temperature extrusion-based 3D printing has shown promise for hydrogel scaffold fabrication, as rapid solidification under controlled cooling can improve shape retention during deposition. However, commercial low-temperature DIW systems are often expensive, closed-source, and difficult to customize. Moreover, most require a two-step post-processing workflow involving external freeze-thaw cycle

Bridging sepsis physiology and biosensor engineering: a clinical perspective on electrochemical cytokine sensor design.

Sepsis remains one of the most critical challenges in contemporary intensive care medicine, characterized by profound immunological and physiological heterogeneity. Cytokines have gained increasing attention as dynamic biomarkers that can complement conventional markers and bedside clinical assessment, enabling more refined diagnosis, risk stratification, and monitoring of treatment response. Most current work on cytokine biosensing is organized around a technological ambition: continuous, minimally invasive, wearable measurement. In this Perspective, we invert that starting point and derive s
