BACKGROUND: DNA piezoelectric biosensors have become a promising tool in molecular medicine since they do not require any label or staining. Here, a DNA piezoelectric biosensor based on a quartz crystal microbalance (QCM) was created to identify abnormal genes causing α-thalassemia 1 (SEA deletion).
METHODS: The functionalized gold electrode of the quartz crystal was coated with avidin and the biotinylated DNA probe was attached. The target gene causing α-thalassemia 1 was amplified and hybridized with the immobilized probe. DNA hybridization was indicated by changes in the quartz resonance f

The renal Na-K-Cl cotransporter (NKCC2) is selectively expressed in the apical membranes of cells of the mammalian kidney, where it is the target of the clinically important loop diuretics. In contrast, the "secretory" NKCC1 cotransporter is localized in the basolateral membranes of many epithelia. To identify the sorting signal(s) that direct trafficking of NKCCs, we generated chimeras between the two isoforms and expressed these constructs in polarized renal epithelial cell lines. This analysis revealed an amino acid stretch in NKCC2 containing apical sorting information. The NKCC1 C terminu

N-acetylglucosaminidase (NAG) is particularly active in mammalian spermatozoa and appears to be involved in fertilization. Although it is assumed that this enzyme is acrosomal, previous results from our laboratory suggest the presence of NAG at the sperm plasma membrane level. The present study attempted to analyse the subcellular distribution of this enzyme in human spermatozoa. Sperm were incubated under different conditions and NAG activity measured in the soluble extracts and cell pellets using a specific fluorometric substrate. A significant proportion of NAG activity was released when sp

DNA microarrays are useful tools to study changes of gene expression in response to a treatment with drugs. Here, we describe the optimization of conditions for the cDNA synthesis and hybridization protocols to be used for a low-density DNA microarray called 'Rat HepatoChips.' This DNA microarray with 59 carefully selected genes could be used to study changes in gene expression levels due to a treatment with xenobiotic. These 59 genes (including 8 housekeeping genes) have been selected among potential toxic markers involved in basic cellular processes and drug metabolism related genes. Using t

BACKGROUND: Acute myeloid leukemia (AML) remains challenging to treat and often requires intensive chemotherapy. In contrast to other hematologic malignancies, the development of effective antibody-based and cellular immunotherapies for AML has been limited by the scarcity of suitable target antigens.
METHODS: We applied a phage display-based whole-cell panning method in which Fab-phage were biotinylated and captured, followed by next-generation sequencing (NGS), bioinformatic, and statistical analyses. Target deconvolution was performed using a CRISPR-Cas9 knockout library, fluorescence-acti

High-quality regeneration of large bone defects depends on the coordinated integration of immunomodulation, osteogenic signal presentation, and structural support. However, conventional bioinks often struggle to simultaneously fulfill these key requirements, particularly in terms of the effective utilization of low-dose bone morphogenetic protein 2 (BMP-2) and matching biomechanical interfaces. Inspired by the shaping behavior of traditional porcelain clay, we developed a clay-like microsphere-based hydrogel bioink that integrates biomimetic immunomodulation, low-dose osteogenic factor deliver

Antigen presentation via MHC class I molecules (MHC-Is) is a turning point in the establishment of immune responses to endogenous threats, such as viruses. From their synthesis to their cell surface display, MHC-Is travel to many compartments, including the endoplasmic reticulum, Golgi, and endosomes. They come close to a plethora of molecules, some of which regulate directly or indirectly their folding and trafficking. While many of these proteins are well characterized, such as those found in the peptide loading complex, others remain to be discovered. The proxiome can be studied using proxi

An in situ nanozyme assembly amplification strategy is reported based on the programmable bioaffinity-driven assembly of streptavidin-Pd@Pt and biotin-Pd@Pt nanozymes, enabling ultrasensitive colorimetric immunoassay of protein biomarkers at sub-femtomolar levels. In this design, streptavidin-Pd@Pt nanozymes are first anchored onto the immunocomplexes through standard biotinylated antibody recognition, followed by the recruitment of biotin-Pd@Pt nanozymes via strong streptavidin-biotin interactions. This stepwise and localized assembly process induces the formation of high-density Pd@Pt nanozy

OBJECTIVE: In some patients with membranous nephropathy (MN) receiving rituximab therapy, treatment failure may be partly related to the development of anti-rituximab antibodies (ADAs). This study aimed to establish a highly sensitive time-resolved fluoroimmunoassay (TRFIA) for detecting ADAs in these patients.
METHOD: A streptavidin-coated microplate was used as the solid phase, with biotinylated rituximab as the capture antibody and Eu3+ -labeled rituximab as the detection antibody. A bridging assay was developed to detect ADAs, and the method was optimized, validated, and preliminarily app

Respiratory diseases are challenging to diagnose rapidly and accurately due to diverse pathogens, overlapping symptoms, and high infection rates. Here, we developed a suspension array based on magnetic microspheres encoded with blue fluorescent protein (mTagBFP) and Cy5-labeled anti-Tag ssDNA (MB@mTagBFP&anti-Tag-Cy5) for simultaneous detection of five core respiratory pathogens. Multiplex PCR was performed using 5'-Tag-labeled upstream primers and 5'-biotinylated downstream primers, yielding dual-labeled products (Tag and biotin). The resulting complexes (MB@mTagBFP&anti-Tag-Cy5-Tag-dsDNA-phy

The Getah virus (GETV) is a mosquito-borne pathogen that infects diverse hosts, including pigs, horses, and humans, which can cause swine reproductive disorders such as abortion and stillbirth, posing a potential threat to animal and public health. Therefore, there is an urgent need for efficient and accurate serological diagnostic methods for surveillance and control of GETV. However, commercial diagnostic kits for swine GETV infection remain unavailable. In this study, we developed a novel enzyme-linked immunosorbent assay (ELISA) based on a GETV-specific epitope peptide (E2EP3) for serologi

Alzheimer's disease (AD), caused by amyloid-β (Aβ) aggregation-derived senile plaques, is among the most prevalent neurodegenerative disorders. Recently, Roche and Lilly announced the development of an AD diagnosis that utilizes phosphorylated tau (p-tau) and apolipoprotein E epsilon 4 allele as biomarkers, rather than Aβ. Additionally, the National Institute on Aging and Alzheimer's Association Research Framework suggests that p-tau molecules can serve as critical biomarkers for AD diagnosis. Here, we quantified p-tau181 in human plasma using a novel electrochemical assay method called the El

In vitro fertilization (IVF) faces challenges in evaluating embryo quality and in determining the genetic health of embryos. Key biomarkers in the culture medium, including nucleic acids and proteins, offer promising avenues for noninvasive assessment. However, small sample volumes, low biomolecule concentrations, and potential contaminants complicate the reliable detection of genetic indicators. In this context, we developed a noninvasive preimplantation genetic testing (niPGT) approach using a superparamagnetic particles-enhanced surface plasmon resonance (SPR) imaging biosensor capable of d

RNA-based therapeutics have revolutionized precision medicine due to their unprecedented potency, specificity, and adaptability. However, the inherent limited stability of RNA, including mRNA used in vaccines, is a major obstacle to the full realization of their potential. This instability, coupled with the centralized nature of vaccine production, currently limits the generation of RNA therapeutics at the point of care, which will otherwise fully harness the potential of these agents. Here, a microfluidic platform is presented for on-demand, personalized synthesis of modified mRNA stabilized

Bacterial gene expression from sites of infection are poorly studied due to low levels of bacterial mRNA present in clinical samples. Here, we develop Transcript-Capture Seq, which uses customizable biotinylated probes generated in-house to enrich bacteria-specific RNA from host samples before Next Generation Sequencing (NGS). This method results in a >200-fold increase in bacterial mRNA reads from mixed samples and allows analysis of the complete bacterial transcriptome from clinical samples. We apply Transcript-Capture to models of tuberculosis (TB) infection as well as sputum samples from T

This article describes the carbohydrate composition of early and mature placentas from bitches, detected by lectin histochemistry. Formalin-fixed placental samples from 11 mixed-breed bitches have been assigned to the 'early' or the 'mature' placenta group, processed by the routine histological technique and labelled with a panel of 14 biotinylated lectins. The glycan distribution was almost completely preserved over pregnancy. Cytotrophoblast cells expressed high-mannose N-linked residues and N-acetyl-D-glucosamine, whereas mannose N-linked residues, N-acetyl-D-glucosamine and β- and α-D-gala

Porcine circoviruses (PCVs) contain four types: PCV1, PCV2, PCV3, and PCV4, all of which can infect pigs. Among them, PCV1 is non-pathogenic, and PCV2 can cause porcine circovirus diseases (PCVD) or porcine circovirus-associated diseases (PCVAD). Although the pathogenicity of PCV3 and PCV4 is still controversial, increasing evidence shows that PCV3 and PCV4 can cause PCV-related disease. However, mixed infection of PCV2, PCV3, and PCV4 with other pathogens often occurs in large-scale pig breeding, bringing severe economic losses to the global pig industry. In this study, the soluble recombinan

Reactive oxygen species (ROS) that are overproduced in certain tumors can be considered an indicator of oxidative stress levels in the tissue. Here, we report a magnetic resonance imaging (MRI)-based probe capable of detecting ROS levels in the tumor microenvironment (TME) using ROS-responsive manganese ion (Mn2+)-chelated, biotinylated bilirubin nanoparticles (Mn@bt-BRNPs). These nanoparticles are disrupted in the presence of ROS, resulting in the release of free Mn2+, which induces T1-weighted MRI signal enhancement. Mn@BRNPs show more rapid and greater MRI signal enhancement in high ROS-pro

OBJECTIVE: This study aims to explore the function of circRIMS in cerebral ischemia/reperfusion (CIR) and its regulatory mechanism.
METHOD: The expression of the circRIMS was examined in GEO chip data and validated by qRT-PCR analysis. A middle cerebral artery occlusion/repression (MCAO/R) model was developed using C57BL/6J mice. Starbase and circinteractome were employed to identify the target miRNA and mRNA. The result was confirmed by dual-luciferase reporter assay, and biotinylated RNA-pulldown assay. The cell viability and apoptosis were confirmed through CCK-8 and flow cytometry assay.
