Hormonal perturbations induced by multicategory endocrine-disrupting chemicals increase the risk of polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) is intricately linked to both reproductive and metabolic health, and its pathogenesis remains unclear. Previous studies suggest that exposure to endocrine disrupting chemicals (EDCs) may contribute to PCOS. However, there is still a lack of systematic researches and analyses on this topic. In this study, we investigated the association between PCOS and EDC exposure and established a mechanistic link between the two by analyzing hormone levels. The meta-analysis pointed out that the levels of EDCs in patients with PCOS were significantly higher than those in the

Transcriptomic- and metabolomic-based elucidation of macrophage inflammatory response in gestational bisphenol A (BPA)-induced cholesterol metabolic reprogramming.

Accumulating evidence reveals that bisphenol A (BPA) exposure triggered maternal lipid metabolism disorders and pregnancy complications, but the molecular regulatory networks driving metabolic reprogramming remain to be fully elucidated. This study employed an integrated multi-omics approach based on BPA-exposed animal and cell models to systematically explore the molecular mechanisms underlying gestational BPA-induced metabolic dysfunction and macrophage inflammatory activation. Untargeted metabolomics revealed elevated cholesterol abundance in gestational BPA-exposed mice, indicating that ge

Intake of Trans-Fat Diet With Prenatal Bisphenol A Exposure Induce Global DNA Methylation in Rats Offspring.

BACKGROUND: Bisphenol A (BPA) is a ubiquitous industrial compound found in an enormous variety of consumer goods such as plastics, epoxy resins, and thermal paper. Despite its widespread usage, the effects of BPA on the risk and development of metabolic syndrome are not fully understood. Prenatal exposure to BPA has been shown to disrupt the development of offspring's metabolically active tissues and increase their susceptibility to the adverse consequences of a trans-fat diet (TFD). METHODS: In this experiment, pregnant rats (n = 18) were randomly separated into three groups. These groups we

Bisphenol A in utero induced glutamate and D-serine metabolic dysregulation in the hippocampus of rats and primary cultured astrocytes.

Bisphenol A (BPA) is a widely used synthetic compound that could cause neurobehavioral abnormalities in mammals. Previous studies have suggested that NMDAR may be a potential target of BPA-induced neurotoxicity. However, the impact of exposure to BPA on glutamate (Glu) and D-serine (D-ser) metabolism (key regulators of NMDAR activation) has not been clarified yet. Pregnant SD rats were exposed to 0, 0.05, 0.5, 5 and 50 mg/kg/day BPA via oral gavage from gestational day (GD) 5 to GD 19, and primary cultured astrocytes (AS) from neonatal rats were exposed to 5, 10 and 20 μmol/L BPA, respectively

Integrative genetic, single-cell and mechanistic dissection identifies bisphenol a as a causal driver of preeclampsia via fibroblast-derived IL-6/STAT3 signaling.

In hypertensive disorders of pregnancy, particularly preeclampsia, epidemiological associations have increasingly implicated the endocrine disruptor Bisphenol A (BPA), but robust mechanistic evidence remains elusive. Building upon accumulating clinical concern and in vivo data, we pursued an integrative, multi-omic investigation to dissect whether BPA causally and directly contributes to placental maladaptation and vascular dysfunction. Two-sample Mendelian randomization analyses using large European-ancestry genome-wide association datasets demonstrated a significant, directionally concordant

Co-Exposure to Polystyrene Microplastics and Bisphenol A Contributes to the Formation of Liver Fibrosis in Mice through Inhibition of the BMAL1/E-Cad Signaling Pathway.

The food safety risks posed by exposure to polystyrene microplastics (PS-MPs) and bisphenol A (BPA) have become an issue worldwide. However, the toxic effects of PS-MPs and BPA coexposure on the mammalian liver remain elusive. In this study, we found that PS-MPs and BPA coexposure have synergistic toxic effects on AML12 cells and the mouse liver. Histopathological staining revealed excessive accumulation of the extracellular matrix in the coexposure liver. Co-exposure to PS-MPs and BPA downregulated Bmal1 and E-cad both in vitro and in vivo. Additionally, Bmal1-/- AML12 cells and liver-specifi

Unexpected Interacting Effects of Physical (Radiation) and Chemical (Bisphenol A) Treatments on Male Reproductive Functions in Mice.

For decades, numerous chemical pollutants have been described to interfere with endogenous hormone metabolism/signaling altering reproductive functions. Among these endocrine disrupting substances, Bisphenol A (BPA), a widely used compound, is known to negatively impact germ and somatic cells in the testis. Physical agents, such as ionizing radiation, were also described to perturb spermatogenesis. Despite the fact that we are constantly exposed to numerous environmental chemical and physical compounds, very few studies explore the impact of combined exposure to chemical and physical pollutant

Association between prenatal bisphenol a exposure and promoter hypermethylation of CAPS2, TNFRSF25, and HKR1 genes in cord blood.

In utero exposure to bisphenol A (BPA) in early stages of development has been reported to exert adverse health effects on offspring later in life. Epigenetic alterations, particularly DNA methylation, may be one plausible biological mechanism involved. We examined the association between maternal BPA exposure and DNA methylation in cord blood. We randomly selected 96 paired samples of maternal urine and infant cord blood collected from the Shanghai-Minhang Birth Cohort. BPA levels in maternal urine were measured using high-performance liquid chromatography (HPLC). Three cord blood samples wit
