Role of the brain-stem reticular formation in tonic-clonic seizures: lesion and pharmacological studies.

Bilateral lesions of the pontine tegmentum involving the superior cerebellar peduncles and the nucleus reticularis pontis oralis have been shown to attenuate the tonic components of maximal seizures induced by electroshock, sound stimulation (audiogenic), or pentylenetetrazol, although having no effect on clonus in three separate seizure models. The pontine tegmental lesion also abolishes the clonus of minimal audiogenic seizures that have a motor pattern different from that of other clonic models, and are believed to originate in the brain stem. The preponderant suppression of tonus by the po…

Pathways for orofacial pain sensation in the trigeminal brain-stem nuclear complex of the Macaque monkey.

Eleven Macaque monkeys underwent a variety of lesions in the trigeminal afferent system; namely, tractotomy, rhizotomy, and radiofrequency destruction of various components of the bulbospinal trigeminal nuclear complex. Behavioral responses were evaluated before and after the lesions using a quantitative paradigm which measured lever-pressing in response to electrical stimulation of the dental pulp or facial skin, and by assessing adversive responses to facial cutaneous and intraoral pin-scratch. Thresholds for lever-pressing in response to cutaneous facial stimulation were elevated by tractot

Respiratory rhythm generation in the in vitro brain stem-spinal cord preparation of the neonatal rat.

An in vitro preparation was described for studying electrical activity of mammalian brain stem and spinal cord. The brain stem and the spinal cord were isolated from 0-4-day-old rats, placed in a bath and perfused with modified Krebs solution. Various reflex responses could be recorded from cranial nerves by stimulation of other cranial nerves. The preparation was viable for more than 7 h. Spontaneous periodic neural activity could be recorded from phrenic, hypoglossal and other spinal nerves. The periodic discharges of phrenic nerves are synchronized with those of ventral roots C4 and the upw…

Widespread cortical projections of the ventral tegmental area and of other brain stem structures in the cat.

Thirty-three cat brains with injections of horseradish peroxidase in various regions of the cerebral cortex were screened for afferent proje … e rat. Though our results might be attributed to species differences in the cortical innervation of brain stem structures, we prefer explanations which emphasize different densities in the distribution of brain stem afferents to the cortex, and/or which suggest different cortical targets of catecholaminergic and noncatecholaminergic neurons.

[The experimental study of the cerebral vasospasm--the micro-circulation on the hypothalamus and the brain stem (author's transl)].

The correlation between the cerebral vasospasm and the micro-circulation in the hypothalamus and brainstem was observed histologically and electronmicroscopically using the colloidal carbon infusion method through the right vertebral artery of dogs. The posterior communicating artery of the dog was ruptured with a fine needle, which resulted in subarachnoid hemorrhage. The colloidall infusion was done at 30 min., 48 hours, 1 week and 4 weeks after the subarachnoid hemorrhage. Results 1) The cerebral vasospasm was confirmed by vertebral angiography on all cases with 30 min., 48 hours, and 1 wee

The relationship between cyclic changes in thyroid function and behavior of cats with brain stem lesions.

Behavioral and thyroid-states were monitored in sixteen cats for 20 months. Six of the cats had stereotaxic lesions of the ventro-lateral parts of the brain stem; these six cats exhibited periodically an abnormal behavioral state that consisted of the tactile elicitability of 'grooming fragments'. The onsets of the abnormal behavioral state occurred at variable intervals after the surgery, appearing mostly in the fall of the year. The other ten cats were controls and never exhibited the abnormal behavioral state. The thyroid-state was monitored during the 20-month period by determining the per…

Orb2 RNA-binding activity promotes neural stem cell development and brain growth in Drosophila larvae.

Neurodevelopment requires precise translational control, the disruption of which is implicated in neurological disorders, developmental delays, and microcephaly. We identify a role for the Drosophila CPEB-family protein Orb2, a translational regulator, in controlling brain size in a dose-dependent manner. Loss of orb2 results in larval brain hypotrophy, whereas orb2 overexpression causes brain overgrowth. We demonstrate that orb2 is required for neural stem cell (NSC) development from embryonic through larval neurogenesis. Structure-function analysis reveals that Orb2 RNA-binding activity prom

Heat shock preconditioning potentiates the therapeutic efficacy of mesenchymal stem cells for traumatic brain injury.

Traumatic brain injury (TBI) is a serious disease characterized by a high incidence in young people and can lead to significant neurological impairment. Deterioration in cognitive function following TBI is often underpinned by a sustained neuroinflammatory state, which impairs the integrity of the blood-brain barrier (BBB) and accelerates neuronal loss. Mesenchymal stem cells (MSCs) have emerged as a promising therapeutic approach for TBI because of their regenerative and immunomodulatory properties. Here, we identified heat shock (HS) pretreatment as an effective method to potentiate the ther

Engineered neural stem cell vesicles activating Wnt promote blood-brain barrier repair after cerebral haemorrhage.

The blood-brain barrier (BBB), a highly specialized neurovascular structure indispensable for preserving cerebral homeostasis, exhibits significant impairment across diverse neurological pathologies; however, its therapeutic targeting persists as a formidable challenge due to the inherent complexity of its multicellular architecture and dynamic regulatory networks. Although the Wnt/β-catenin signalling pathway orchestrates the development and maintenance of the BBB, the clinical translation of Wnt-based interventions remains elusive. We fabricated functionalized extracellular vesicles derived

Mesenchymal stromal/stem cell-derived extracellular vesicles in brain disorders: mechanisms of repair and recovery.

Mesenchymal stem/stromal cell-derived small extracellular vesicles (MSC-sEVs) have emerged as promising cell-free therapeutics for central nervous system (CNS) disorders including stroke, traumatic brain injury (TBI), dementia, and multiple sclerosis (MS). MSC-sEVs offer advantages of low immunogenicity, ease of storage, and ability to cross the blood-brain barrier. This review provides a comprehensive analysis of the mechanisms by which MSC-sEVs have been reported to promote neural repair and recovery in preclinical models, through two convergent categories of action. First, MSC-sEVs exert di

Chronic haloperidol exposure impairs neurodevelopment via Notch1 signaling in human stem cell-derived brain organoids.

Haloperidol is a typical antipsychotic used to treat schizophrenia and induces dopamine D2 receptor antagonism. Long-term use of haloperidol can reduce brain size in animals and humans; however, the underlying mechanism of this effect remains unclear. Notch1 signaling regulates the development and function of the nervous system by balancing stem cell proliferation and differentiation. Therefore, we investigated the effects of long-term exposure to haloperidol on human-derived brain organoids, which served as sophisticated in vitro models of human brain development. Long-term exposure to halope

Cellular Stem Cell Therapy for Treating Traumatic Brain Injury: Strategies for Enhancement of Therapeutic Efficacy.

Traumatic brain injury (TBI) influences a considerable population globally. TBI notably impacts both fatalities and disabilities worldwide. The mortality related to TBI is a significant concern in public health, affecting persons across various age groups and demographic profiles. More research and preventative interventions are required to alleviate TBIs' effects and optimize patient outcomes. Stem cell (SC) treatment exhibits promise as a viable strategy for addressing TBI due to its capacity to possibly restore or regenerate the compromised cells within the central nervous system. Additiona

Brain-Derived Neurotrophic Factor Levels in Mesenchymal Stem Cell Culture Fluid Under Different Culture Conditions.

OBJECTIVE: Mesenchymal stem cells are used in various fields, such as cellular therapy, regeneration, or tissue engineering. It has been shown that they exhibit many protective factors and also work as a modulating chief within the region in which they are administered. There are studies on both the therapeutic and neuroprotective effects of brain-derived neurotrophic factor. Also, there are many studies on the improvement of culture conditions for in vitro reproduction of mesenchymal stem cells, which can be obtained from many sources in various bodies, such as adipose tissue and Wharton's je

Interleukin-33 improves the neurogenesis of neural stem cells in perinatal brain after hypoxia-ischemia.

Perinatal hypoxia-ischemia (HI) insult is an important cause of neonatal encephalopathy, and the effective therapeutic approaches are currently limited. Interleukin (IL)-33 acts as a member of the IL-1 superfamily and has been shown to be neuroprotective following experimental neonatal HI and adult stroke. Here, we explore the effect of IL-33 and its specific receptor ST2 axis on endogenous neurogenesis in neonatal brain after HI. ST2 was found on the surface of NSCs, and the expression of ST2 was further enhanced after HI challenge. Delivery of IL-33 obviously repopulated the size of NSC pool

Intravascular ultrasound guided vertebral artery stenting after basilar artery thrombectomy for acute brain stem stroke.

A 64-year-old lady presented as right vertebral artery occlusion and brain stem stroke (Figure 1A). Emergent thrombectomy opened the artery, but it re-occluded 10 minutes later (Figure 1B,C). Intravascular ultrasound showed heavy plaque burden and guided a balloon-expandable stenting successfully (Figure 1D-F).

Induced Pluripotent Stem Cell (iPSC)-Derived Endothelial Cells to Study Bacterial-Brain Endothelial Cell Interactions.

Bacterial meningitis is a serious infection of the central nervous system (CNS) that occurs when blood-borne bacteria are able to exit the cerebral vasculature and cause inflammation. The blood-brain barrier (BBB) and the meningeal blood-CSF barrier (mBCSFB) are composed of highly specialized brain endothelial cells (BECs) that possess unique phenotypes when compared to their peripheral endothelial counterparts. To cause meningitis, bacterial pathogens must be able to interact and penetrate these specialized BECs to gain access to the CNS. In vitro models have been employed to study bacterial-
