Ramp acceleration and hydraulic conductivity (L(p)) of living capillaries.

Living mesenteric capillaries with either an intact or disrupted glycocalyx were challenged with ramp change in shear stress (Deltatau). Animals (Rana pipiens) were divided randomly into two experimental groups, and two true capillaries (TC) per animal were investigated. The modified Landis technique was combined with intravital microscopy to view individual TC and assess hydraulic conductivity (L(p)), an index of capillary function. Median L(p) was 3.2 x 10(-7) for control and 11.8 x 10(-7) cm s(-1) cm H(2)O(-1) after mild, brief (1 min) pronase treatment (P<0.0001). Analysis by stimulus comp

Broad Tuning of Paper Microfluidic Properties by Covalent Surface Modification for Precise Flow Control and Sensing.

In an effort to innovate on-site sensing platforms for a wide range of analytes in different matrices, microfluidic paper-based devices (μPADs) are promising candidates to bring the lab to the sample, as they allow passive, capillary-action-driven flow. Their use, however, is somewhat limited by the fact that the integration of advanced functionality and flow control is difficult. Although recent progress in this area has led to the development of on/off-valving and timing control of flow by changing the chemical and physical properties of paper, precise control over flow in paper microfluidic

Glutaric Acid Affects Pericyte Contractility and Migration: Possible Implications for GA-I Pathogenesis.

Glutaric acidemia I (GA-I) is an inherited neurometabolic childhood disease characterized by bilateral striatal neurodegeneration upon brain accumulation of millimolar concentrations of glutaric acid (GA) and related metabolites. Vascular dysfunction, including abnormal cerebral blood flow and blood-brain barrier damage, is an early pathological feature in GA-I, although the affected cellular targets and underlying mechanisms remain unknown. In the present study, we have assessed the effects of GA on capillary pericyte contractility in cerebral cortical slices and pericyte cultures, as well as

CIRP contributes to multiple organ damage in acute pancreatitis by increasing endothelial permeability.

Acute pancreatitis can lead to systemic inflammation and multiple organ damage. Increased endothelial permeability is a hallmark of systemic inflammation. Several studies have demonstrated that cold-inducible RNA-binding protein (CIRP) functions as a proinflammatory factor in various diseases. However, its role in endothelial barrier dysfunction during acute pancreatitis remains unknown. To study this, acute pancreatitis was induced by two hourly intraperitoneal injections of 4.0 g/kg L-arginine in wild-type (WT) or CIRP knockout mice. Our results showed that CIRP levels in the pancreas, small

The blood-ocular barriers.

The introduction of the concept of blood-ocular barriers in the ophthalmic literature is briefly reviewed. Two main blood-ocular barriers are proposed: the blood-aqueous barrier and the blood-retinal barrier. The blood-aqueous barrier is formed by an epithelial barrier located in the nonpigmented layer of the ciliary epithelium and in the posterior iridial epithelium, and by the endothelium of the iridial vessels. Both these layers have tight junctions of the "leaky" type. The pereability of the blood-aqueous barrier shows a significant degree of pressure-dependent diffusion associated with tr

Assessment of Blood-Brain Barrier Permeability in a Cerebral Ischemia-Reperfusion Model in Rats; A Pilot Study.

Animal models are an important tool for studying ischemic mechanisms of stroke. Among them, the middle cerebral artery occlusion (MCAO) model via the intraluminal suture method in rodents is closest to human ischemic stroke. It is a model of transient occlusion followed by reperfusion, thus representing cerebral ischemia-reperfusion model that simulates patients with vascular occlusion and timely recanalization. Although reperfusion is very beneficial for the possibility of preserving brain functions after ischemia, it also brings a great risk in the form of brain edema, which can cause the de

Arterial Pulsations cannot Drive Intramural Periarterial Drainage: Significance for Aβ Drainage.

Alzheimer's Disease (AD) is the most common form of dementia and to date there is no cure or efficient prophylaxis. The cognitive decline correlates with the accumulation of amyloid-β (Aβ) in the walls of capillaries and arteries. Our group has demonstrated that interstitial fluid and Aβ are eliminated from the brain along the basement membranes of capillaries and arteries, the intramural periarterial drainage (IPAD) pathway. With advancing age and arteriosclerosis, the stiffness of arterial walls, this pathway fails in its function and Aβ accumulates in the walls of arteries. In this study we

A Novel Three-Dimensional Human Peritubular Microvascular System.

Human kidney peritubular capillaries are particularly susceptible to injury, resulting in dysregulated angiogenesis, capillary rarefaction and regression, and progressive loss of kidney function. However, little is known about the structure and function of human kidney microvasculature. Here, we isolated, purified, and characterized human kidney peritubular microvascular endothelial cells (HKMECs) and reconstituted a three-dimensional human kidney microvasculature in a flow-directed microphysiologic system. By combining epithelial cell depletion and cell culture in media with high concentratio

Anti-endothelial cell antibodies in nephropathia epidemica and other viral diseases.

Increased capillary permeability is a central feature of the severe forms of haemorrhagic fever with renal syndrome (HFRS) and occurs also, though less frequently, in nephropathia epidemica (NE), one of the milder forms of this syndrome, caused by Puumala virus. We therefore searched for antiendothelial cell antibodies (AECA) in patients with NE and in those with other presumed or serologically proven acute viral illnesses. By enzyme immunoassay, using human umbilical vein endothelial cells (HUVEC) as the substrate, IgG class AECA were detected significantly more frequently in patients with NE…

Impact of interleukin-6 on drug transporters and permeability in the hCMEC/D3 blood-brain barrier model.

The blood-brain barrier (BBB) is a highly selective membrane composed predominantly of brain capillary endothelial cells expressing drug efflux transporters that prevent substrates from accessing the brain. Inflammation is associated with central nervous system diseases and can impair BBB permeability via several mechanisms, including altered transporter and cell junction expression. This can modify the brain's exposure to drugs. However, comprehensive genomic analysis of the impact of interleukin (IL)-6, which plays a key role in the inflammatory response, on the BBB is lacking. In the presen

Inflammation is an amplifier of lung congestion by high lv filling pressure in hemodialysis patients: a longitudinal study.

INTRODUCTION: Since inflammation alters vascular permeability, including vascular permeability in the lung, we hypothesized that it can be an amplifier of lung congestion in a category of patients at high risk for pulmonary oedema like end stage kidney disease (ESKD) patients. OBJECTIVE AND METHODS: We investigated the effect modification by systemic inflammation (serum CRP) on the relationship between a surrogate of the filling pressure of the LV [left atrial volume indexed to the body surface area (LAVI)] and lung water in a series of 220 ESKD patients. Lung water was quantified by the numb

An experimental study on the pathogenesis of acute haemorrhagic enteropathy--significance of congestion and endotoxaemia.

To clarify the pathogenesis of acute haemorrhagic enteropathy, the present study was undertaken using the ileum of rats. The ligation of marginal veins of the ileum or the injection of endotoxin to the intact rats induced only slight mucosal haemorrhage. On the other hand, the injection of endotoxin into rats with ligation of the marginal veins led to severe mucosal haemorrhage. The severe mucosal haemorrhage was markedly inhibited by suppression of the production of bradykinin. These experimental results suggest that some cases of acute haemorrhagic enteropathy may be induced by the co-existe

Lead-induced abnormalities in blood-brain barrier permeability in experimental chronic toxicity.

The aim of this paper was to determine whether prolonged drinking of lead acetate-containing water by adult rats, which imitates environmental exposure to lead (Pb), affects some morphological and biochemical properties of rat brain microvessels. We noted a significant increase of lead level in capillaries and synaptosomes obtained from brains of rats under chronic toxicity conditions. Intravenously injected horseradish peroxidase (HRP) was used to evaluate the functional state of the blood-brain barrier (BBB). The results indicate that, systematically administered at low doses, lead induces B

Cochlear immunoglobulin in the C3H/lpr mouse model for autoimmune hearing loss.

Immunoglobulin staining was conducted in cochlear tissue from the C3H/lpr autoimmune strain mouse to better understand the local immune processes underlying autoimmune inner ear disease. This mouse is a model for spontaneous systemic lupus erythematosus with coincident elevated cochlear thresholds. Cochleas were examined from C3H/lpr mice at 2 months of age, before disease onset, and at 8 months of age, when systemic disease and hearing loss are manifested. Sections of these cochleas, along with cochlear sections from age-matched C3H/HeJ nonautoimmune controls, were immunocytochemically staine

[Heroin-induced pulmonary edema].

Based on a case story and having examined the literature, we describe the incidence, symptoms, course, complications and treatment of the kind of pulmonary oedema that may arise in connection with acute heroin intoxication. A possible pathogenesis is also discussed. Heroin-induced pulmonary oedema is rather frequent and the mortality is high. It differs from cardiogenic pulmonary oedema at essential points. It is most likely due to an increased permeability of the lung capillaries. However, it is still unclarified whether this is caused by a toxic or an allergic reaction, or by hypoxia. The tr

Neurotensin Regulates Primate Ovulation Via Multiple Neurotensin Receptors.

Neurotensin (NTS), a small neuropeptide, was recently established as a key paracrine mediator of ovulation. NTS mRNA is highly expressed by granulosa cells in response to the luteinizing hormone surge, and multiple NTS receptors are expressed by cells of the ovulatory follicle. To identify the role of NTS receptors NTSR1 and SORT1 in ovulation in vivo, the dominant follicle of cynomolgus macaques (Macaca fascicularis) was injected with either vehicle control, the general NTS receptor antagonist SR142948, the NTSR1-selective antagonist SR48692, or the SORT1-selective antagonist AF38469. hCG was
