The ICGN mouse strain is a unique model for naturally occurring nephrotic syndrome. In the present study, we examined the onset of the clinical manifestation of nephrotic syndrome and determined the sequence of intraglomerular events associated with progression of nephrotic conditions. Laboratory analysis revealed that homozygous (nep/nep) mice showed urinary albumin excretion during the suckling stage, rapidly leading to hypoalbuminemia accompanied by body growth failure. Renal pathology demonstrated that an initial intraglomerular event in the nephrotic mice was observed 3 weeks after birth

This paper describes the cellular and tissue distribution of P-glycoprotein (P-GP) (mdr1 gene product), the role of P-GP in vivo and immunodiagnosis of multi-drug-resistant cancers. We mainly used MRK 16 monoclonal antibody (MAb) reactive with P-GP. P-GP was found to be expressed very strongly in the adrenal cortex of adults and strongly in the renal tubules of the kidney, capillary blood vessels of the brain, and also in placenta. Interestingly, P-GP was not distributed in fetal and neonatal adrenals, and thus may be closely related to adrenal maturation. A high level of P-GP expression was a

OBJECTIVE: Dyslipidemia is common in people with HIV (PWH) and linked to cardiometabolic disease risk. Subcutaneous adipose tissue (SAT) regulates lipid storage and release, but how SAT cellular composition might influence circulating lipids in PWH on contemporary antiretroviral therapy (ART) is not well defined.
DESIGN: Cross-sectional, observational cohort of PWH on long-term contemporary ART with virologic suppression.
METHODS: We performed untargeted fasting plasma lipidomic profiling on 127 individuals with a range of metabolic fitness (nondiabetes, prediabetes, diabetes). Adjusted logi

INTRODUCTION: Streptococcus pyogenes (S. pyogenes) can cause streptococcal toxic shock syndrome (STSS), especially in immunocompromised individuals. Early diagnosis is the key to treat STSS effectively. This study presents a fatal case of STSS caused by the emerging M1(UK) S. pyogenes lineage in a patient with type 2 diabetes mellitus (T2DM), alongside a comprehensive literature review. CASE PRESENTATION: A 65-year-old man with T2DM presented with altered consciousness, necrotizing soft tissue injury and was diagnosed with STSS. Bacterial cultures were obtained from right lower limb exudate, r

Patients with cervical spinal cord injuries (CSCI) experience hypotension mainly caused by attenuated total peripheral resistance. Some physicians and nurses believe that this peripheral blood pressure (PBP) decrease in patients with CSCI results in the formation of pressure injuries. We hypothesized that the measurement of PBP, such as arteriole and capillary pressure, could be a simple and efficient means of detecting skin and deep tissue damage in patients with CSCI. However, PBP in the skin has not been measured in patients with CSCI. The purpose of this study was to investigate PBP in pat

BACKGROUND/AIM: Oral squamous cell carcinoma (OSCC) is characterized by early metastasis, clinical resistance and poor prognosis. Recently, we showed that aggressive OSCC cells co-express endothelial cell markers and can form tube-like structures, known as vasculogenic mimicry (VM), a process associated with poor prognosis in head and neck cancers. Given the limited success of current antiangiogenic therapy in treating OSCC, this study sought to explore the efficiency of these drugs in targeting an ex vivo model of VM.
MATERIALS AND METHODS: OSCC cell lines from the tongue and floor of the mo

Antibiotic susceptibility testing is vital to tackle the emergence and spread of antimicrobial resistance. Inexpensive digital CMOS cameras can be converted into portable digital microscopes using 3D printed x-y-z stages. Microscopic examination of bacterial motility can rapidly detect the response of microbes to antibiotics to determine susceptibility. Here, we present a new simple microdevice-miniature microscope cell measurement system for multiplexed antibiotic susceptibility testing. The microdevice is made using melt-extruded plastic film strips containing ten parallel 0.2 mm diameter mi

Cerebral blood flow is conveyed by vascular resistance arteries and downstream parenchymal arterioles. Steady-state vascular resistance to blood flow increases with decreasing diameter from arteries to arterioles that ultimately feed into capillaries. Due to their smaller size and location in the parenchyma, arterioles have been relatively understudied and with less reproducibility in findings than surface pial arteries. Regardless, arteriolar endothelial cell structure and function-integral to the physiology and etiology of chronic degenerative diseases-requires extensive investigation. In pa

BACKGROUND: Coinfection with malaria and typhoid fever is a major public health issue in developing countries. In endemic areas, including Ethiopia, people are at risk of acquiring both malaria and typhoid fever at the same time. Therefore, this study aimed to determine the magnitude of malaria-typhoid fever coinfection in febrile patients attending hospital at Southern Ethiopia.
METHODS: A hospital-based cross-sectional study was carried out on 416 febrile patients attending Arba Minch General Hospital from 1st October to 30th December 2021. The data was collected using a pretested structure

Large conduit arteries and the microcirculation participate in the mechanisms of elevation of blood pressure (BP). Large vessels play roles predominantly in older subjects, with stiffening progressing after middle age leading to increases in systolic BP found in most humans with aging. Systolic BP elevation and increased pulsatility penetrate deeper into the distal vasculature, leading to microcirculatory injury, remodelling, and associated endothelial dysfunction. The result is target organ damage in the heart, brain, and kidney. In younger individuals genetically predisposed to high BP, incr

Medical intractability, i.e. the absence of any response to anti-epileptic drug (AED) therapy, is an unresolved problem in many patients with epilepsy. Mechanisms of intractability are not well understood, but may include alterations of pharmacological targets and poor penetration of AEDs into the brain because of increased expression of multiple drug-resistance proteins, such as P-glycoprotein (Pgp; ABCB1), capable of active brain extrusion of various drugs, including AEDs. Increased expression of Pgp has been reported in brain tissue of patients with refractory epilepsy, but there is a lack

We successfully used argon plasma coagulation (APC) to treat two cases of dialysis patients with hemorrhagic gastric angiodysplasia. Gastric angiodysplasia is recognized as an important cause of gastrointestinal bleeding. Angiodysplastic lesion confined to the gastric antrum was first described in 1953 and named gastric antral vascular ectasia (GAVE). The condition is characterized as submucosal capillary dilatation and fibromuscular hyperplasia. The typical finding of GAVE is the so-called watermelon stomach, attributable to vasodilatation. In case 1, a 69-year-old man was introduced continuo

BACKGROUND: Vasculogenic mimicry (VM) and epithelial-mesenchymal transition (EMT) contribute to cervical cancer invasiveness and treatment resistance, yet the upstream regulators coordinating these plasticity programs remain incompletely defined. Ephrin receptor B2 (EPHB2) promotes malignant phenotypes in multiple tumor types; however, its functional role and mechanistic basis in cervical cancer, particularly in VM-associated plasticity, remain unclear.
METHODS: EPHB2 dysregulation was screened in public transcriptomic cohorts (GSE168244, tumor vs. normal; GSE138080, precancer vs. normal) and

BACKGROUND: High-intensity focused ultrasound (HIFU) is a noninvasive therapy that makes entire coagulative necrosis of a tumor in deep tissue through the intact skin. There are many reports about the HIFU's efficacy in the treatment of patients with breast cancer, but randomized clinical trials are rare which emphasize on the systematic assessment of histological changes in the ablated tumor vascularities, while clinical trials utilizing bevacizumab and other anti-angiogenic drugs in breast cancer have not demonstrated overall survival benefit. The purpose of this study is to evaluate the dam

BACKGROUND: Single doses of exogenous ketone salts (KS) transiently increase circulating beta-hydroxybutyrate (BHB) (∼1 mM; 1-2 h) regardless of starting levels of ketosis; however, no studies have explored how sustained use of KS influences measures of ketonemia and glycemia.
OBJECTIVES: To determine the response to a hypocaloric, well-formulated ketogenic diet (KD), with and without the inclusion of two daily racemic KS doses (6 g R-BHB + 6 g S-BHB per serving) on 1) daily fasting capillary R-BHB and glucose (R-BHB/GLUfast), 2) bi-weekly 13 h diurnal BHB and glucose (R-BHB/GLUdiur), 3) thre

Controversy persists regarding whether the efficacy of closed instrumental mitral commissurotomy compares well enough with that of open commissurotomy to warrant its continued use. The purpose of this study was to compare the results of operation as determined by catheterization studies in 63 patients with pure, severe, and noncalcified mitral stenosis. The patients were randomly assigned to one of two groups: thirty-two patients were operated on by the closed technique (group I) and 31 by the open technique (group II). All patients underwent left-sided and right-sided catheterization before a

Membrane distillation (MD) is a promising thermally driven desalination technology due to its high rejection of non-volatile contaminants and its ability to operate under relatively mild conditions. However, membrane wetting remains a persistent challenge that hinders its distillation efficiency and long-term application. This review presents a mechanism-based perspective on MD wetting by linking capillary pressure imbalance, surface-tension reduction, surfactant adsorption, scaling, and colloid-driven interfacial interactions to pore invasion and performance deterioration. The influence of ke

Biomimetic anisotropy is defined as intentionally engineered, nature-inspired directional differences in structure, chemistry, roughness, stiffness, or pore architecture. These directional differences lower transport resistance in one direction relative to the opposite direction, which results in rectified transport. In this review, anisotropy design is synthesized across surfaces, porous materials, and soft systems, with transport considered for droplets, low-surface-tension liquids, particles, and soft objects. Biological inspirations are summarized first, and the design lessons that can be
