Analyzing the toxicological effects of PET-MPs on male infertility: Insights from network toxicology, mendelian randomization, and transcriptomics.

Current research indicates that polyethylene terephthalate microplastics (PET-MPs) may significantly impair male reproductive function. This study aimed to investigate the potential molecular mechanisms underlying this impairment. Potential gene targets of PET-MPs were predicted via the SwissTargetPrediction database. GWAS summary statistics for male infertility were obtained from the GWAS Catalog, and eQTL data were acquired from the eQTLGen database. SMR analysis was subsequently performed to identify genes exhibiting significant causal associations with male infertility. The potential targe

Geographic patterns of antibiotic resistance in the human gut microbiome: insights from metagenome-assembled genomes across four Chinese provinces.

Next-generation metagenomic sequencing has substantially advanced our understanding of the human intestinal microbiome. Many commensal microbes in the human gut carry a wide array of antibiotic resistance genes (ARGs), collectively forming the gut-associated resistome. In this study, we investigated the gut resistome using metagenomic sequencing. We collected 119 fecal samples from individuals in four Chinese provinces: Yunnan, Guizhou, Sichuan, and Jiangsu. By constructing metagenome-assembled genomes (MAGs) and comprehensive gene catalogs, we aimed to characterize the microbial community str

The role of inflammatory cytokines and blood metabolites in osteoporosis: A Mendelian randomization study on pathways and therapeutic targets.

The precise mechanisms underlying the development of osteoporosis (OP) remain unclear, but evidence suggests that inflammatory cytokines and blood metabolites play a significant role. We aim to investigate, as a strictly genomic study based on genome-wide association studies (GWAS) summary statistics, the causal relationships between inflammatory cytokines, blood metabolites, and OP by using Mendelian randomization (MR) analysis and uncover potential therapeutic targets. We utilized inflammatory cytokines from a GWAS summary containing 8293 healthy participants, 1400 blood metabolites from GWA

Altered gut mycobiome and cross-kingdom microbial interactions in systemic lupus erythematosus.

BACKGROUND: Systemic lupus erythematosus (SLE) is a complex autoimmune disorder shaped by host genetics and environmental exposures, including the gut microbiota. While bacterial dysbiosis in SLE is well characterized, the role of the gut mycobiome and its cross-kingdom interactions remains largely unexplored. METHODS: Using fecal metagenomic sequencing from 117 SLE patients and 115 healthy controls (HCs), we established a non-redundant fungal genome catalog and revealed significant alterations in fungal composition, function, and cross-kingdom ecology. RESULTS: Fungal diversity was increased

Obesity reduces the urate-lowering efficacy among patients with primary gout: a prospective cohort study.

OBJECTIVES: Obesity affects 53% of gout patients, while its effect on urate-lowering therapy (ULT) is unclear. This study aimed to compare the response to febuxostat among different BMI catalogs of male gout patients. METHODS: A prospective study recruited 633 men with gout, classified by BMI into normal-weight, overweight and obese groups. Baseline age, disease duration and serum urate (SU) levels were matched simultaneously. All participants received febuxostat for 12 weeks, increasing the dosage from 20 to 40 mg daily. We compared the efficacy of ULT and the incidence of gout flares in thr

No Causal Relationship Between Helicobacter Pylori Infection and Rosacea: A 2-Sample Mendelian Randomization Study.

BACKGROUND: Previous studies have shown that patients with rosacea have a higher prevalence of Helicobacter pylori (H. pylori) infection. However, whether H. pylori infection contributes to the development of rosacea remains unclear, and no genetic association studies between the two have been conducted to date. Genome-wide association studies (GWAS) database is a public resource that stores and shares data that aims to identify genetic links to complex diseases, physiological traits, or drug responses. Mendelian randomization (MR) is an epidemiological approach to investigate the effect of th

The mediating role of circulating inflammatory cytokines in causal associations between plasma metabolites and asymptomatic bile duct and cholecyst calculus: A Mendelian randomization study.

Asymptomatic gallbladder and biliary tract calculus may make into symptomatic disease or bring anxiety for patients. The formation of gallstones was associated with genetic risk factors and metabolic abnormalities. Genome-wide association studies (GWAS) data of 1400 plasma metabolites (PMs) and 91 circulating inflammatory cytokines (CICs) were obtained from the GWAS catalog, while the GWAS data of calculus of gallbladder without cholecystitis and calculus of bile duct without cholangitis or cholecystitis were retrieved from the IEU OpenGWAS project. The causalities from PMs or CICs to asymptom

Neurobehavioral correlates of dopamine agonist-induced eye-blinking in the marmoset monkey.

Eye-blinking has been used to catalog dopaminergic receptor subtype activation in several mammalian species. In this study, the dissimilar effects of directly-acting D1 and D2 agonists and an indirectly-acting non-selective agonist (SKF-82958, quinelorane, cocaine, respectively) on eye-blinking were confirmed in marmosets. Subsequently, functional magnetic resonance imaging (fMRI) was used to examine their effects on functional connectivity (FC) between the dopamine-rich putamen and other brain regions. Results indicate that SKF-82958 produced dose-dependent increases in blinking, with the hig

Linking coronary artery disease to neurodegenerative diseases through systems genetics.

Coronary artery disease (CAD) is still a leading cause of death worldwide despite the extensive research and the considerable progresses made through the years. As other cardiovascular diseases, CAD is the result of the complex interaction between genetic variants and environmental factors. Currently identified genetic loci associated to CAD revealed the contribution of multiple molecular pathways to its pathogenesis, suggesting the need for a systemic approach to understand the role of genetic determinants. In this study we wanted to investigate how GWAS variants associated to CAD interact wi

Underfunding of German university-based high-performance medicine exemplified by the treatment of varices in cirrhosis.

Facing increasing economization in the health care sector, clinicians have to adapt not only to the ever-growing economic challenges, but also to a patient-oriented health care. Treatment costs are the most important variable for optimizing success when facing scarce human resources, increasing material- and infrastructure costs in general, as well as low revenue flexibility due to flat rates per case in Germany, the so-called Diagnosis-Related Groups (DRG). University hospitals treat many patients with particularly serious illnesses. Therefore, their share of complex and expensive treatments,

Widespread transposable element dysregulation in human aging brains with Alzheimer's disease.

INTRODUCTION: Transposable element (TE) dysregulation is associated with neuroinflammation in Alzheimer's disease (AD) brains. Yet, TE quantitative trait loci (teQTL) have not been well characterized in human aged brains with AD. METHODS: We leveraged large-scale bulk and single-cell RNA sequencing, whole-genome sequencing (WGS), and xQTL from three human AD brain biobanks to characterize TE expression dysregulation and experimentally validate AD-associated TEs using CRISPR interference (CRISPRi) assays in human induced pluripotent stem cell (iPSC)-derived neurons. RESULTS: We identified 26,

Novel odd-chain cyclopropane fatty acids: detection in a mammalian lipidome and uptake by hepatosplanchnic tissues.

Microbe-produced molecules (xenometabolites) found in foods or produced by gut microbiota are increasingly implicated in microbe-microbe and microbe-host communication. Xenolipids, in particular, are a class of metabolites for which the full catalog remains to be elaborated in mammalian systems. We and others have observed that cis-3,4-methylene-heptanoylcarnitine is a lipid derivative that is one of the most abundant medium-chain acylcarnitines in human blood, hypothesized to be a product of incomplete β-oxidation of one or more "odd-chain" long-chain cyclopropane fatty acids (CpFAs). We dedu

Exploring genetic associations between immune cells and hypertensive disorder of pregnancy using Mendelian randomization.

BACKGROUND: Observational epidemiological studies suggested that immunological dysregulation and inflammation play a significant role in the placental and renal dysfunction that leads to maternal hypertension. The immunophenotypes' possible causalities with hypertensive disease of pregnancy remain ambiguous. We performed two-sample Mendelian randomization (MR) analyses to comprehensively investigate the causal effect of immunophenotypes on hypertensive disorder of pregnancy (HDP). METHODS: The large-scale genome-wide association studies (GWASs) data on immunological traits was taken from publ

The impact of neurocognitive and psychiatric disorders on the risk of idiopathic normal pressure hydrocephalus: A bidirectional Mendelian randomization study.

BACKGROUND: Neurocognitive and psychiatric disorders have been proved that they can comorbid more often with idiopathic normal pressure hydrocephalus (iNPH) than general population. However, the potential causal association between these disorders and iNPH has not been assessed. Thus, our study aims to investigate the causal relationship between them based on a bidirectional Mendelian randomization (MR) analysis. METHODS: Random effects of the inverse variance weighted (IVW) method were conducted to obtain the causal association among the neurocognitive disorders, psychiatric disorders, and i
