Once considered as a "metabolic waste," lactate is now recognized as a major fuel for tricarboxylic acid (TCA) cycle. Our metabolic flux ana … glucose to TCA cycle. MCT1 deficiency increases the expression of mitochondrial proteins, augments cell respiration rate, and elevates mitochondrial activity in the muscle. Mechanistically, the protein level of PGC-1α, a master regulator of mitochondrial biogenesis, is elevated upon loss of MCT1 via increases in cellular NAD+ level and SIRT1 activity. Collectivel …

Iron metabolism is pivotal for cell fitness in the mammalian host; however, its role in group 3 innate lymphoid cells (ILC3s) is unknown. Here we show that transferrin receptor CD71 (encoded by Tfrc)-mediated iron metabolism cell-intrinsically controls ILC3 proliferation and host protection against Citrobacter rodentium infection and metabolically affects mitochondrial respiration by switching of oxidative phosphorylation toward glycolysis. Iron deprivation or Tfrc ablation in ILC3s reduces the expression and/or activity of the aryl hydrocarbon receptor (Ahr), a key ILC3 regulator. Genetic abl

Cystatin B (CSTB) acts as an inhibitor of cysteine proteases of the cathepsin family and loss-of-function mutations result in human brain diseases with a genotype-phenotype correlation. In the most severe case, CSTB-deficiency disrupts brain development, and yet the molecular basis of this mechanism is missing. Here, we establish CSTB as a regulator of chromatin structure during neural stem cell renewal and differentiation. Murine neural precursor cells (NPCs) undergo transient proteolytic cleavage of the N-terminal histone H3 tail by cathepsins B and L upon induction of differentiation into n

Chemical and biological limitations in bioactive compound design based on natural product (NP) structure can be overcome by the combination of NP-derived fragments in unprecedented arrangements to afford "pseudo-natural products" (pseudo-NPs). A new pseudo-NP design principle is described, i.e., the combination of NP-fragments by transformations that are not part of current biosynthesis pathways. A collection of indofulvin pseudo-NPs is obtained from 2-hydroxyethyl-indoles and ketones derived from the fragment-sized NP griseofulvin by means of an iso-oxa-Pictet-Spengler reaction. How metabolic status controls the fates of different types of leukemia cells remains elusive. Using a SoNar-transgenic mouse line, we demonstrated that B cell acute lymphoblastic leukemia (B-ALL) cells had a preference in using oxidative phosphorylation. B-ALL cells with a low SoNar ratio (SoNar-low) had enhanced mitochondrial respiration capacity, mainly resided in the vascular niche, and were enriched with more functional leukemia-initiating cells than that of SoNar-high cells in a murine B-ALL model. The SoNar-low cells were more resistant to cytosine arabinoside (Ara-C) treatment. cyclic a

The tuberous sclerosis complex (TSC) 1/2 is a negative regulator of the nutrient-sensing kinase mechanistic target of rapamycin complex (mTORC1), and its function is generally associated with tumor suppression. Nevertheless, biallelic loss of function of TSC1 or TSC2 is rarely found in malignant tumors. Here, we show that TSC1/2 is highly expressed in Burkitt's lymphoma cell lines and patient samples of human Burkitt's lymphoma, a prototypical MYC-driven cancer. Mechanistically, we show that MYC induces TSC1 expression by transcriptional activation of the TSC1 promoter and repression of miR-15

Mitochondria generate ATP and building blocks for cell growth and regeneration, using pyruvate as the main substrate. Here we introduce PyronicSF, a user-friendly GFP-based sensor of improved dynamic range that enables real-time subcellular quantitation of mitochondrial pyruvate transport, concentration and flux. We report that cultured mouse astrocytes maintain mitochondrial pyruvate in the low micromolar range, below cytosolic pyruvate, which means that the mitochondrial pyruvate carrier MPC is poised to exert ultrasensitive control on the balance between respiration and anaplerosis/gluconeo

In the presence of glucose, Saccharomyces cerevisiae prefers glycolysis followed by fermentation to generate ATP. One adaptation to this fermentative lifestyle is the retrograde pathway (RTG), which induces the transcription of genes, including several Krebs cycle genes, to increase pools of alpha-ketoglutarate. We previously found that both petite and RTG mutants are highly sensitive to canavanine, a toxic analog of arginine, indicating that residual respiration and the RTG pathway are required to maintain arginine levels in the cell. Yet, from an amino acid biosynthesis standpoint, it is unc

Alzheimer's disease (AD) is a devastating neurodegenerative disorder characterized by memory loss and a decline in cognitive function. Hallmarks of AD include an age-dependent accumulation of toxic amyloid beta (Aβ) 42 in the brain, energy dyshomeostasis caused by mitochondrial dysfunction, and iron overload. However, the role of iron overload and mitochondrial dysfunction in AD pathology is unknown and their precise relationship with Aβ 42 toxicity in AD pathology is unclear. C. elegans provide a powerful model system to untangle and clarify these relationships. In this study, we quantify the

The increasing rise of multidrug-resistant bacteria necessitates an urgent need for the discovery of novel antibacterial agents. Natural products have long been a source for identifying and isolating novel antibacterial agents. Anacardic acids (AAs), a phenolic lipid isolated from solventextracted cashew nutshell liquid (CNSL) of Anacardium occidentale (Family Anacardiaceae), have garnered potential attention for their potent antibacterial properties. Besides Anacardium occidentale, different analogues of AAs have also been isolated from various natural sources. These natural and structurally

BACKGROUND: Microphthalmia-associated transcription factor (MITF) is a master regulator of mast cell (MC) differentiation and effector function and is essential for anaphylaxis (severe allergic reactions). Although MITF has been linked to mitochondrial regulation through pyruvate dehydrogenase, whether it broadly controls mitochondrial fitness and metabolic programs that exacerbate MC activation remains unclear.
OBJECTIVE: To determine how MITF regulates MC mitochondrial function and metabolic fitness beyond pyruvate dehydrogenase control, and to define its contribution to heightened MC activ

Mitochondria of seeds face the challenge of remaining quiescent over long periods and rapidly resuming their respiratory function once environmental conditions become favorable. Despite the fundamental importance of mitochondrial functions for germination, the composition and functionality of seed mitochondria remain poorly understood. Previous work has suggested that dry seeds contain immature promitochondria largely devoid of cristae or respiratory complexes and that key mitochondrial functions need to be re-established by rebuilding a functional proteome to support germination. Here, we exa

There is an increasing recognition that mitochondrial dysfunction is associated with autism spectrum disorders. However, little attention has been given to the etiology of mitochondrial dysfunction and how mitochondrial abnormalities might interact with other physiological disturbances such as oxidative stress. Reserve capacity is a measure of the ability of the mitochondria to respond to physiological stress. In this study, we demonstrate, for the first time, that lymphoblastoid cell lines (LCLs) derived from children with autistic disorder (AD) have an abnormal mitochondrial reserve capacity

Porin is the most abundant outer membrane (OM) protein of mitochondria. It forms the aqueous channel on the mitochondrial OM and mediates major metabolite flux between mitochondria and cytosol. Mitochondrial porin in Trypanosoma brucei, a unicellular parasitic protozoan and the causative agent of African trypanosomiasis, possesses a beta-barrel structure similar to the bacterial OM porin OmpA. T. brucei porin (TbPorin) is present as a monomer as well as an oligomer on the mitochondrial OM, and its expression is developmentally regulated. In spite of its distinct structure, the TbPorin function

The tumor microenvironment imposes diverse metabolic challenges to cancer cells. Overcoming these challenges is essential for survival, proliferation, and dissemination. However, how cancer cells cope with the harsh environment and how the different coexisting stresses affect the tumor in vivo are unknown. Recently, Groessl and colleagues published their findings in Science showing that acidosis outweighs all other stresses and plays a major role in adaptation to them. Mechanistically, acidosis inhibits the ERK-dynamin-related protein 1 (DRP1) pathway, resulting in mitochondria elongation, whi

Invariant natural killer T (iNKT) cells are a subset of innate-like T lymphocytes that play important roles in immune responses. The mechanisms underlying the stage-specific development and differentiation of iNKT effector subsets are not fully elucidated. Here, using Zbtb16-Cre-driven conditional knockout mice, we found that conditional deletion of the E3 ligase Von Hippel-Lindau (VHL) resulted in impaired terminal maturation of iNKT cells and attenuated lineage specification of the iNKT2 and iNKT17 subsets. VHL deficiency disrupted iNKT cell-mediated tumor surveillance, thereby promoting B16

Many bipolar disorder (BD) patients are non-responsive to lithium. The mechanisms underlying lithium (non-)responsiveness are largely unknown. By using gene-set enrichment analysis methods, we found that core clock gene-sets are significantly associated with lithium response. Among the top hits was BHLHE41, a modulator of the molecular clock and homeostatic sleep. Since BHLHE41 and its paralog BHLHE40 are functionally redundant, we assessed chronic lithium response in double-knockout mutant mice (DKO). We demonstrated that DKOs are non-responsive to lithium's effect in various behavioral tasks

Iron is an essential biometal, critical in processes that include oxygen transport, mitochondrial respiration, and cell signaling. Iron dyshomeostasis is linked with hyperglycemia and associated metabolic disorders, but the underlying mechanisms are poorly understood. To investigate these mechanisms, we conducted a 4-week study on mice given glucose-supplemented water. The supplementation induced metabolic shifts in the liver towards triglyceride synthesis. We tracked iron trafficking by analyzing liver and serum markers of iron metabolism alongside iron speciation analysis as determined by li

Regulatory T cells (Tregs) are a key determinant for the immunosuppressive and premetastatic niche for cancer progression after surgery resection. However, the precise mechanisms regulating Tregs function during surgical stress-facilitated cancer metastasis remain unknown. This study aims to unravel the mechanisms and explore potential strategies for preventing surgical stress-induced metastasis by targeting NEDD8. Using a surgical stress mouse model, we found that surgical stress results in the increased expression of NEDD8 in Tregs. NEDD8 depletion abrogates postoperative lung metastasis of
