Plant defense activators for sustainable crop protection: a mechanism-based perspective on their regulatory positioning.

The environmental and ecological costs of chemical pesticide use have intensified the search for alternative crop protection strategies. Among these, plant defense activators (PDAs) represent a distinct class of compounds that enhance plant resistance through activation or priming of endogenous defense pathways, rather than through direct toxicity to target organisms. However, current regulatory frameworks often classify such agents as plant protection products or equivalent regulatory categories, irrespective of their non-biocidal, host-mediated mode of action. In this review, we focus on a s

Design and engineering of photorespiratory bypasses in plants.

Photorespiration is an essential metabolic process in C3 plants, yet it imposes significant carbon losses of up to 30% or more. Synthetic biology has recently enabled the engineering of diverse photorespiratory bypasses to overcome this limitation. In this review, we categorize these bypasses into three major types based on glycolate carbon retention and CO2 release. The first are chloroplast-localized carbon-releasing bypasses, which shift CO2 release from mitochondria to chloroplasts, thereby establishing a localized CO2-concentrating mechanism around Rubisco. The second are carbon-neutral b

Epigenetic-epitranscriptomic crosstalk through TaHAG1-TaNSUN2 coordinates thermotolerance in wheat.

High temperature is a primary abiotic stress that severely constrains crop productivity. Deciphering the regulatory pathways underlying heat responses is essential for breeding heat-tolerant crops with stable yields. Although both epigenetic and epitranscriptomic regulations are involved in plant heat adaptation, their mechanistic interplay remains unclear. Here, integrated epigenomic (H3K9Ac/H3K14Ac) and transcriptomic profiling under heat stress identifies the mRNA m⁵C methyltransferase TaNSUN2 as a key regulator of thermotolerance in wheat. We demonstrate that TaNSUN2 is transcriptionally a

Nanobiochar Mitigates Photosynthetic Impairment in Rice Caused by Antibiotic Cocontamination: A Mechanistic Elucidation of Dual-Site Photoinhibition and Metabolic Reprogramming.

Antibiotic cocontamination poses a severe but poorly understood threat to crop photosynthesis. This study employed an integrated multiscale framework to uncover the phytotoxicity of tetracycline (TC) and ciprofloxacin (CIP) on rice seedlings and evaluate the mitigation potential of nanobiochar (nBC). Coexposure to TC and CIP amplified antibiotic residues in both root and shoot, exacerbating growth inhibition and photosynthetic pigment depletion, with the net photosynthetic rate plummeting to 1.44 μmol CO2 m-2 s-1. Mechanically, chlorophyll fluorescence and transmission electron microscopy reve

Pilot deployment of beta carotene-enriched rice (Golden Rice) in the Philippines.

Golden Rice, registered as Malusog Rice in the Philippines, is a biofortified rice containing beta carotene in its grains to address vitamin A deficiency (VAD), a prevalent public health problem. It can provide 30-50% of the estimated average requirement for vitamin A. Its commercial approval in July 2021 led to a pilot-scale deployment in selected high VAD prevalent areas. From 2022 wet cropping season to 2023 wet cropping season, it was cultivated on 202 hectares across 24 provinces in the country, demonstrating significant field performance comparable to that of other rice varieties. It gai

Cyclin C and EB1c cooperatively regulate plant height in eggplant via internode elongation.

Plant height is an important agronomic characteristic that influences crop productivity and overall plant structure. Elucidating the molecular mechanisms underlying stem elongation in eggplant (Solanum melongena L.) is essential for advancing crop improvement. In this study, we identified a semi-dwarf eggplant mutant, eggplant dwarf4 (edf4), generated through Ethyl methanesulfonate (EMS) mutagenesis. Genetic mapping and functional analyses revealed that a point mutation in SmCycC, which encodes a homolog of Cyclin C, is responsible for the dwarf phenotype. The edf4 mutant exhibited 28.67% (p <

Foliar application of plant-derived peptides decreases the severity of leaf rust (Puccinia triticina) infection in bread wheat (Triticum aestivum L.).

BACKGROUND: Screening and developing novel antifungal agents with minimal environmental impact are needed to maintain and increase crop production, which is constantly threatened by various pathogens. Small peptides with antimicrobial and antifungal activities have been known to play an important role in plant defense both at the pathogen level by suppressing its growth and proliferation as well as at the host level through activation or priming of the plant's immune system for a faster, more robust response against fungi. Rust fungi (Pucciniales) are plant pathogens that can infect key crops …

Transcriptomic and metabolomic approaches elucidate the systemic response of wheat plants under waterlogging.

Extreme weather conditions lead to significant imbalances in crop productivity, which in turn affect food security. Flooding events cause serious problems for many crop species such as wheat. Although metabolic readjustments under flooding are important for plant regeneration, underlying processes remain poorly understood. Here, we investigated the systemic response of wheat to waterlogging using metabolomics and transcriptomics. A 12 d exposure to excess water triggered nutritional imbalances and disruption of metabolite synthesis and translocation, reflected by reductions in plant biomass an

Efficacy of Hydroponically Cultivated Saffron in the Preservation of Retinal Pigment Epithelium.

Saffron treatment is a broad-spectrum therapy used for several retinal diseases, and its effectiveness depends on a particular molecular composition (REPRON® saffron). Its production requires specific crops and procedures that, together with low yields, make this spice expensive. To reduce costs, the use of hydroponic crops is gradually increasing. In this study, we tested the protective properties of a hydroponic saffron (sH) batch in models of retinal pigmented epithelium (RPE) degeneration. ARPE-19 cells were pretreated with 40 µg/mL saffron and exposed to different types of damage: excess

Phenotypic characterization of the wheat temperature-sensitive leaf color mutant and physical mapping of mutant gene by reduced-representation sequencing.

Few available leaf color mutants in crops have greatly limited the understanding of photosynthesis mechanisms, leading to few accomplishments in crop yield improvement via enhanced photosynthetic efficiency. Here, a noticeable albino mutant, CN19M06, was identified. A comparison between CN19M06 and the wild type CN19 at different temperatures showed that the albino mutant was temperature-sensitive and produced leaves with a decreased chlorophyll content at temperatures below 10 °C. Genetic analysis suggested that the albinism was controlled by one recessive nuclear gene named TSCA1, which was

Crop protection practices and risks associated with infectious tropical parasitic diseases.

Two recent literature reviews have shown that: i) agroecological crop protection (ACP) practices generally reduce risks of viral zoonoses, unlike conventional (agrochemical-based) practices which tend to increase them; ii) substitution-based crop protection (CP) practices (mainly biocontrol-based) could result in fewer health risks from bacterial infectious diseases. Here, we present an analysis of the scientific literature to determine to what extent the conclusions regarding viruses or bacteria can be extended to infectious diseases caused by protozoan or helminthic parasites. This analysis

Magnesium Limitation Leads to Transcriptional Down-Tuning of Auxin Synthesis, Transport, and Signaling in the Tomato Root.

Magnesium (Mg) deficiency is becoming a widespread limiting factor for crop production. How crops adapt to Mg limitation remains largely unclear at the molecular level. Using hydroponic-cultured tomato seedlings, we found that total Mg2+ content significantly decreased by ∼80% under Mg limitation while K+ and Ca2+ concentrations increased. Phylogenetic analysis suggested that Mg transporters (MRS2/MGTs) constitute a previously uncharacterized 3-clade tree in planta with two rounds of asymmetric duplications, providing evolutionary evidence for further molecular investigation. In adaptation to …

β-N-Oxalyl-l-α,β-diaminopropionic Acid (β-ODAP) Content in Lathyrus sativus: The Integration of Nitrogen and Sulfur Metabolism through β-Cyanoalanine Synthase.

Grass pea (Lathyrus sativus L.) is an important legume crop grown mainly in South Asia and Sub-Saharan Africa. This underutilized legume can withstand harsh environmental conditions including drought and flooding. During drought-induced famines, this protein-rich legume serves as a food source for poor farmers when other crops fail under harsh environmental conditions; however, its use is limited because of the presence of an endogenous neurotoxic nonprotein amino acid β-N-oxalyl-l-α,β-diaminopropionic acid (β-ODAP). Long-term consumption of Lathyrus and β-ODAP is linked to lathyrism, which is

Engineered Rhizobacteria Exploit Bacterial Vesicles for Cross-Kingdom RNAi and Soilborne Disease Control.

The escalating severity of antifungal resistance necessitates novel crop protection strategies. RNAi is promising but faces challenges including environmental instability of dsRNA and poor cellular delivery efficiency. In this study, we engineered the cotton rhizobacterium Pantoea agglomerans YC4 to deliver dsRNA into Verticillium dahliae. Small RNA-seq revealed that bacterial outer membrane vesicles (OMVs) selectively enrich specific endogenous sRNAs compared with intracellular compartments, with a congruent sRNA enrichment pattern observed in fungal cells, thus confirming OMVs as the primary

Defense Mechanisms of Cotton Fusarium and Verticillium Wilt and Comparison of Pathogenic Response in Cotton and Humans.

Cotton is an important economic crop. Fusarium and Verticillium are the primary pathogenic fungi that threaten both the quality and sustainable production of cotton. As an opportunistic pathogen, Fusarium causes various human diseases, including fungal keratitis, which is the most common. Therefore, there is an urgent need to study and clarify the resistance mechanisms of cotton and humans toward Fusarium in order to mitigate, or eliminate, its harm. Herein, we first discuss the resistance and susceptibility mechanisms of cotton to Fusarium and Verticillium wilt and classify associated genes b

An Improved Leaf Protoplast System for Highly Efficient Transient Expression in Switchgrass (Panicum virgatum L.).

As a robust perennial C4-type monocot plant and a native species to North America, switchgrass (Panicum virgatum) has been evaluated and designated as a strong candidate bioenergy crop by the U.S. DOE. Although genetic modifications of switchgrass have been used to successfully reduce the recalcitrance of switchgrass biomass for biofuel production, the generation of transgenic switchgrass is still a slow and laborious process. A transient protoplast system can provide an excellent platform to accelerate the selection of genes-of-interest for tailoring switchgrass biomass. However, partially du

Epiphytic pink-pigmented methylotrophic bacteria enhance germination and seedling growth of wheat (Triticum aestivum) by producing phytohormone.

Methylotrophic bacteria were isolated from the phyllosphere of different crop plants such as sugarcane, pigeonpea, mustard, potato and radish. The methylotrophic isolates were differentiated based on growth characteristics and colony morphology on methanol supplemented ammonium mineral salts medium. Amplification of the mxaF gene helped in the identification of the methylotrophic isolates as belonging to the genus Methylobacterium. Cell-free culture filtrates of these strains enhanced seed germination of wheat (Triticum aestivum) with highest values of 98.3% observed using Methylobacterium sp.

Development of on-farm AMF inoculum production for sustainable agriculture in Senegal.

The integration of endomycorrhizal fungi into agricultural practices as inoculum offers the potential to improve plant productivity while reducing reliance on expensive chemical fertilizers, which are not only economically costly but also detrimental to the environment. Mycorrhizal fungi play a crucial role in facilitating plant access to essential mineral elements (such as Phosphorus, Potassium, etc.) and water, particularly in soils characterized by arid and semi-arid conditions. where these resources are often limited. Despite the obvious advantages, the development of arbuscular mycorrhiza
