End-stage renal disease (ESRD) causes the buildup of metabolic waste in the body, requiring hemodialysis to replace the kidneys' filtering function. The main challenge in hemodialysis is the membrane's effectiveness, which is often limited by hemocompatibility issues. Therefore, improving membrane materials to enhance hemocompatibility and remove uremic toxins remains a key focus of research. In this study, we used modified natural polymers as materials for hemodialysis membranes. Our goal was to develop chitosan (Cs)-based membranes crosslinked with vanillin (Va) and blended with gelatin (Gel

Engineering catalytic microenvironments within hybrid materials is a critical strategy for enhancing enzyme stability and catalytic efficiency; however, precise regulation of intermediate transfer and interfacial catalytic coupling remains challenging. Here, we report a crosslinking-regulated, double-crystallized hierarchical hybrid nanoflowers system (D-Cu@c-GOx NFs) that establishes a nanoconfined biointerface for cooperative cascade biocatalysis. The system is constructed via in situ enzyme incorporation, followed by glutaraldehyde-mediated crosslinking and a second crystallization step, ge

Cross-linked lipoic acid nanoparticles (cLA) exhibit significant potential in oral nano-delivery due to their high stability and unique barrier penetration ability, which is mediated by the poly(disulfide) backbone. However, the strong negative charge on their surface (approximately -68 mV) generates strong electrostatic repulsion with the mucus layer and intestinal epithelial cells, limiting the trans-barrier transport efficiency of the nanoparticles. To address this issue, we constructed a cross-linked lipoic acid nanoparticle platform (cLAS) modified with zwitterionic sulfobetaine. This mod

Crosslinking is a key step in the production of stable all-natural polymeric scaffolds for tissue engineering, as it slows the degradation and increases the mechanical properties of the material. In this study, we investigate the crosslinking parameters of a natural, anisotropic scaffold produced from gelatine, chitosan and cellulose through a modified freeze drying protocol, with the goal of maintaining the porous architecture of the scaffold while improving its degradation and mechanical strength. Genipin and EDC were selected as the two crosslinking alternatives, while crosslinker concentra

This study reports a novel pH-tuned double-network hydrogel via an entirely physical crosslinking strategy. The hydrogel consists of a first network formed between ferulic acid and ε-polylysine via hydrogen bonding and electrostatic interaction, while a second polyvinyl alcohol network established through freeze-thaw cycles. The former network confers biological activities, while the combination of the two networks ensures mechanical strength. The hydrogels exhibit pH-tuned behavior, with significant differences in mechanical performance, swelling characteristics, and antioxidant/antibacterial

Conventional wound dressings often fail to integrate rapid hemostasis, antibacterial protection, and a pro-regenerative microenvironment, leading to persistent infection and delayed healing in complex wounds. Herein, we report a multifunctional self-adhesive hydrogel for integrated hemostatic, antibacterial, and regenerative wound management with a synergistic dual-crosslinked network. The hydrogel is composed of methylacrylamide-modified type I collagen (ColMA), o-nitrobenzene-modified hyaluronic acid (HANB), and methylacrylamide-modified chitosan (CSMA), and can rapidly form in situ under UV

In this study, a photocrosslinkable methacrylated gelatin (GelMA) hydrogel, functionalized with metal oxide nanoparticles, was developed to support dentin regeneration in direct pulp capping applications. Magnesium (MgO), silicon (SiO2), strontium (SrO), and zinc (ZnO) were incorporated into GelMA at concentrations of 0.1%, 0.2%, 0.3%, 0.5%, and 1% (v/v) to assess cytotoxicity in human stem cells from the apical papilla (SCAPs). Based on the results, MgO, SiO2, and SrO were selected for further functionalization of GelMA at reduced concentrations (0.1%, 0.075%, 0.05%, and 0.025%; v/v) to evalu

Current cell encapsulation methods face problems with large encapsulation volumes. In this study, we present a modular cell-surface conformal coating strategy using a bispecific DNA-enzyme conjugate to direct molecular assembly and crosslinking. The bispecific conjugate exhibited sustained retention on the cell membrane with a half-life on the order of hours. The conjugate catalyzed the formation of a 10 to 20 μm thick fibrin layer upon addition of fibrinogen, allowing the adsorption and deposition of alginate and poly-L-lysine crosslink the coating. The platform was compatible with spheroids

Gastrectomy is often associated with serious complications that lack effective treatments. Despite the widespread adoption in tissue engineering, the rapid degradation of small intestinal submucosa (SIS) compromised its performance in gastric tissue engineering. Thereby, genipin, a natural crosslinking agent, was employed to modify SIS, and the potential of genipin-crosslinked SIS (GP-SIS) in promoting the regeneration of full-thickness gastric defects was explored. The data on crosslinking efficiency demonstrated that genipin can efficiently fix SIS. GP-SIS exhibited high resistance against c

The effectiveness of state-of-the-art cross-linking strategies and mass spectrometry (MS) detection was explored in an important biological context, namely, the ubiquitin-proteasome system, which is responsible for most of the regulated protein degradation in eukaryotic cells. The locations of possible binding sites on the S. cerevisiae 19S proteasome regulatory particle for Lys48 linked polyubiquitin chains were examined using cross-linking strategies and MS based detection by comparing two types of cross-linkers: a (bis)-sulfosuccinimidyl suberate (BS3) and diethyl suberothioimidate (DEST).

Cell microencapsulation technologies allow non-autologous implantation of therapeutic cells for sustained drug delivery purposes. The perm-selective membrane of these systems provides resistance to rupture, stablishes the upper molecular weight limit in bidirectional diffusion of molecules, and affects biocompatibility. Thus, despite being a decisive factor to succeed in terms of biosafety and therapeutic efficacy, little progress has been made in its optimization so far. Here we show that, compared to other usually used coating designs, genipin-crosslinked double poly-L-lysine (GDP) membranes

Cost-effective and eco-friendly extraction of gallium is a technical challenge. Gallium-containing anions can be effectively adsorbed by Zn2+, quaternary ammonium and polyamine. Polyamine can be grafted into adsorbent by formaldehyde and the crosslinking agent of epichlorohydrin. Zn2+/quaternary ammonium/polyamine lignin was constructed for adsorption of Ga(III) through embellishing of Zn2+, quaternary ammonium and polyamine and crosslinking of epichlorohydrin. The adsorbent was characterized by SEM, ICP-MS, BET, BJH, LDM, FTIR and XRD. The effects of pH, initial gallium concentration, adsorpt

The Pyrrolo[2,1-c][1,4]benzodiazepine dimer, SJG-136, selectively induces DNA interstrand crosslinks (ICL) between guanine residues and exhibits potent in vitro cytotoxicity. SJG-136 demonstrated significant in vivo anti-tumor activity in rodents. Clinical trials showed interesting but incomplete responses. Pharmacokinetic and pharmacodynamic profiles of the molecule displayed high interpatient variability highlighting the need for biomarker-driven patient selection. Accurate detection and assessment of ICL in patients' samples is one such approach. Here we report a modified SJG-136 harboring

Calcific aortic valve disease is an inexorably degenerative pathology and a common cause of morbidity and mortality in heart disease. Clinically, it has no effective medical therapy but can be managed with artificial heart valve replacement, particularly using bioprosthetic heart valves (BHV), entailing high biocompatibility. Current BHV mainly derives from glutaraldehyde (GA) cross-linked pericardium, however, residual aldehyde groups may inevitably remain during cross-link, potentially binding with calcium and inducing in situ calcification after implantation, thus leading to valve re-failur

Bioprinting a resilient yet optically transparent corneal tissue substitute remains a challenge. In this study we introduce an innovative methodology aimed at bolstering the mechanical and optical attributes of silk fibroin (SF) hydrogels, pivotal for the progression of cornea tissue engineering. We devised a unique eosin Y-based photoinitiator system to instigate di-tyrosine linkages within highly concentrated pristine SF solutions under green light exposure. This pioneering technique resulted in SF hydrogels fortified by dityrosine covalent bonds, preserving exceptional transparency and soft

Selective scleral crosslinking has been proposed as a novel treatment to increase scleral stiffness to counteract biomechanical changes associated with glaucoma and high myopia. Scleral stiffening has been shown by transpupillary peripapillary scleral photocrosslinking in rats, where the photosensitizer, methylene blue (MB), was injected retrobulbarly and red light initiated crosslinking reactions with collagen. Here, we adapted a computational model previously developed to model this treatment in rat eyes to additionally model MB photocrosslinking in minipigs and humans. Increased tissue leng

PURPOSE: Corneal crosslinking (CXL) procedures are the treatment of choice in halting progressive corneal ectasia and preserving visual acuity due to keratoconus. Pulsed crosslinking (P-CXL) was developed using intermittent pulsing ultraviolet (UV) light to mitigate the depletion of oxygen levels that occurs with continuous UV exposure in standard crosslinking protocols (C-CXL). This study aimed to explore the use of P-CXL in the treatment of keratoconus and determine whether the availability of oxygen in P-CXL carries superior efficacy outcomes as an alternative to C-CXL modalities.

Exposure to reactive oxygen species (ROS) can induce DNA-protein crosslinks (DPCs), unusually bulky DNA lesions that block replication and transcription and play a role in aging, cancer, cardiovascular disease, and neurodegenerative disorders. Repair of DPCs depends on the coordinated efforts of proteases and DNA repair enzymes to cleave the protein component of the lesion to smaller DNA-peptide crosslinks which can be processed by tyrosyl-DNA phosphodiesterases 1 and 2, nucleotide excision and homologous recombination repair pathways. DNA-dependent metalloprotease SPRTN plays a role in DPC re

Skin aging, a complex and inevitable biological process, results in wrinkles, dermal laxity, and skin cancer, profoundly influencing appearance and overall health. Collagen serves as the fundamental element of the dermal matrix; nevertheless, collagen is susceptible to enzymatic degradation within the body. Crosslinking is employed to enhance the physicochemical properties of collagen. However, conventional crosslinking agents may harbor potential issues such as cytotoxicity and calcification risks, constraining their application in the biomedical field. Therefore, we have for the first time d

Covalent adaptable crosslinks, such as the alkyl-hydrazone, endow hydrogels with unique viscoelastic properties applicable to cell delivery and bioink systems. However, the alkyl-hydrazone crosslink lacks stability in biologically relevant environments. Furthermore, when formed with biopolymers such as hyaluronic acid (HA), low molecular weight polymers (<60 kDa), or low polymer content (<2 wt%) hydrogels are typically employed as entanglements reduce injectability. Here, a high molecular weight (>60 kDa) HA alkyl-hydrazone crosslinked hydrogel is modified with benzaldehyde-poly(ethylene glyco
