Inhibition of MRP4 prevents and reverses pulmonary hypertension in mice.

Multidrug resistance-associated protein 4 (MRP4, also known as Abcc4) regulates intracellular levels of cAMP and cGMP in arterial SMCs. Here, we report our studies of the role of MRP4 in the development and progression of pulmonary arterial hypertension (PAH), a severe vascular disease characterized by chronically elevated pulmonary artery pressure and accompanied by remodeling of the small pulmonary arteries as a prelude to right heart failure and premature death. MRP4 expression was increased in pulmonary arteries from patients with idiopathic PAH as well as in WT mice exposed to hypoxic con

Subcellular localization and regulation of type-1C and type-5 phosphodiesterases.

We investigated the subcellular localization of PDE5 in in vitro human myometrial cells. We demonstrated for the first time that PDE5 is localized in discrete cytoplasmic foci and vesicular compartments corresponding to centrosomes. We also found that PDE5 intracellular localization is not cell- or species-specific, as it is conserved in different animal and human cells. PDE5 protein levels are strongly regulated by the mitotic activity of the smooth muscle cells (SMCs), as they were increased in quiescent, contractile myometrial cultures, and conditions in which proliferation was inhibited. Potentiation of slow component of delayed rectifier K(+) current by cGMP via two distinct mechanisms: inhibition of phosphodiesterase 3 and activation of protein kinase G.

1. Regulation of the slowly activating component of delayed rectifier K(+) current (I(Ks)) by intracellular guanosine 3'5' cyclic monophosphate (cGMP) was investigated in guinea-pig sino-atrial (SA) node cells using the whole-cell patch-clamp method. 2. When a cell was dialyzed with pipette solution containing 100 micro M cGMP, I(Ks) started to gradually increase and reached a maximum increase of a factor of 2.37 +/- 0.39 (n = 4) about 10-15 min after rupture of patch membrane. Atrial natriuretic peptide (ANP, 100 nM) also potentiated I(Ks), consistent with intracellular cGMP-induced enhanceme

Activation of the human, intermediate-conductance, Ca2+-activated K+ channel by methylxanthines.

This study demonstrated that the methylxanthines, theophylline, IBMX and caffeine, activate the human, intermediate-conductance, Ca2+-activated K+ channel (hIK) stably expressed in HEK-293 cells. Whole-cell voltage-clamp experiments showed that the hIK current increased reversibly and voltage independently after the addition of methylxanthines. In current-clamp experiments, theophylline dose-dependently hyperpolarised the cell membrane from a resting potential of -18 mV to -56 mV. The methylxanthines did not affect large-conductance (BK) or small-conductance (SK2), Ca2+-activated K+ channels,

Phosphodiesterase-2 inhibitor reverses post-traumatic stress induced fear memory deficits and behavioral changes via cAMP/cGMP pathway.

Phosphodiesterase 2 is one of the phosphodiesterase (PDEs) family members that regulate cyclic nucleotide (namely cAMP and cGMP) concentrations. The present study determined whether PDE2 inhibition could rescue post-traumatic stress disorder (PTSD)-like symptoms. Mice were subjected to single prolonged stress (SPS) and treated with selective PDE2 inhibitor Bay 60-7550 (0.3, 1, or 3 mg/kg, i.p.). The behavioral tests such as forced swimming, sucrose preference test, open field, elevated plus maze, and contextual fear paradigm were conducted to determine the effects of Bay 60-7550 on SPS-induced

Divergent Strategies in Innate Immune Evasion: A Comparative Review of Three Alphaherpesvirus Subfamily Members-PRV, HSV-1, and VZV.

Pseudorabies virus (PRV), an important member of the Alphaherpesvirinae subfamily, shares high evolutionary homology with other members of the same subfamily, including human herpes simplex virus (HSV) and varicella-zoster virus (VZV). However, significant differences exist among them in terms of host range, pathogenicity, and immune evasion strategies. This review systematically compares the roles of PRV, HSV type 1 (HSV-1), and VZV in modulating host innate immune responses, encompassing the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling, RIG-I-like receptor …

Deoxysphingolipids activate cGAS-STING and enhance antitumor immunity.

Deoxysphingolipids (deoxySLs) are a class of non-canonical sphingolipids that can adversely impact mitochondrial function. Mitochondrial DNA release resulting from mitochondrial disruption can activate the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway, and in turn, promote antitumor immunity. Here, we investigated whether inducing deoxySL accumulation in colon cancer cells drives immune-specific, antitumor effects. We show that depleting serine increased deoxySLs and enhanced immune cell infiltration in colon tumors, in parallel with suppressing tumor growth. Elev…

Inside Out: How Cellular Localisation Shapes cGAS Functions in Health and Disease.

The function of innate immune sensors is intricately shaped by their spatial distribution within cells. cGAS (cyclic GMP-AMP synthase), a key cytosolic DNA sensor, illustrates this principle through its unexpected localisation to diverse organelles-including the nucleus, micronuclei, mitochondria, and plasma membrane. In these compartments, cGAS assumes distinct regulatory states and executes specialised functions. For instance, chromatin-bound nuclear cGAS remains inactive under homeostasis but contributes to genome maintenance during genotoxic stress, whereas mitochondrial or micronuclear cG…

Hierarchical engineering of mesoporous polydopamine for "homologous targeting-cascade blasting" biomimetic phototheranostic nanotrident.

The activation of the cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway represents a promising strategy for eliciting host immune responses to eradicate tumors. However, the clinical application of STING agonists is severely hindered by the tumor immunosuppressive microenvironment (TIME) and non-specific delivery. Herein, a biomimetic phototheranostic nanotrident (mPDXZ@M) is meticulously designed through hierarchical engineering of mesoporous polydopamine (mPDA). Upon near-infrared laser irradiation, mPDA-mediated photothermal therapy (PTT) evokes robust cell apopt…

Targeting cGAS-STING-macrophage axis: a novel therapeutic horizon for renal fibrosis.

Renal fibrosis, a pivotal pathological hallmark of chronic kidney disease (CKD), arises from persistent inflammatory responses and extracellular matrix (ECM) deposition. Emerging evidence indicates the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) signaling pathway has exerted a crucial influence on the progression of renal fibrosis. This pathway exacerbates the release of inflammatory factors and ECM deposition through reprograming the polarization state of macrophages, thereby driving renal fibrosis. This review delineates the regulatory role of the cGAS-STING signaling…

Attenuating AAV-triggered innate immunity in the adult mouse nervous system via cGAS-STING pathway inhibition.

While adeno-associated virus (AAV)-mediated gene delivery has emerged as a promising therapeutic modality for neurological disorders, dose-dependent immune responses remain a critical barrier to clinical translation. Here we reveal the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway as a key mediator of innate immune activation following intracranial AAV administration. Through comparative analyses in genetic and pharmacological intervention models, we demonstrate that STING signaling mediates key neuroinflammatory sequelae including glia reactivation, cytotoxic T c…

SIRT4 regulates antiviral and autoimmune responses by promoting cGAS-mediated signaling pathways.

Cyclic guanosine monophosphate (GMP)-AMP synthase (cGAS) is a critical cytosolic DNA sensor, whose activity can be regulated by acetylation. Here, we show that nicotinamide adenine dinucleotide (NAD+)-dependent lysine deacetylase SIRT4 interacts with cGAS and positively regulates innate immune responses triggered by DNA viruses or cytoplasmic DNA. Overexpression of SIRT4 inhibits HSV-1 infection, whereas knockdown of SIRT4 has the opposite effect. Deficiency of SIRT4, or treatment with a SIRT4 inhibitor, impairs antiviral innate immune signaling in response to DNA viruses or cytoplasmic DNA, b

Targeting the cGAS-STING pathway in ophthalmology: from innate immunity to therapeutic innovation.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is a key mediator of the innate immune response to cytosolic DNA, with growing implications beyond traditional host defense mechanisms. Recent research has linked the abnormal activation of this pathway to a wide range of ocular diseases, such as age-related macular degeneration, diabetic retinopathy, corneal injuries, glaucoma, and ocular tumors. This review examines the multifaceted roles of cGAS-STING signaling in ocular inflammation, tissue degeneration, fibrosis, angiogenesis, and immune surveillance. We empha…

Inhibiting cyclin D1-CDK6 suppresses senescence-associated inflammatory gene expression and age-related functional decline.

Cellular senescence contributes to aging and age-related diseases by driving chronic inflammation through the senescence-associated secretor … tion. This leads to the formation of cytoplasmic chromatin fragments that activate pro-inflammatory cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) signaling. In aged mouse livers, senescent hepatocytes show increased Ccnd1 expression. Hepatocyte-specific Ccnd1 knockout or treatment with the clinical grade CDK4/6 inhibitor palbociclib r …

Comparative innate immune responses across major RNA and DNA viral infections: Mechanisms, immunopathology, and therapeutic perspectives.

Innate immunity is the first defense against viral infections, limiting viral replication and initiating adaptive immunity. Viral pathogens are recognized by pattern recognition receptors (PRRs), including Toll-like receptors (TLRs), (RIG-I-like receptors) RLRs, and the (cyclic GMP-AMP synthase) cGAS-STING pathway, which trigger interferon production and antiviral responses. This review compares innate immune responses to major RNA viruses (influenza, SARS-CoV-2, HIV) and DNA viruses (HSV, HBV, CMV). While these viruses activate similar pathways, they differ in interferon dynamics, inflammator…

cGAS-STING Signaling in Central Nervous System Diseases: Neuroinflammatory Mechanisms and Immune Regulation.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway has emerged as a critical cytosolic DNA-sensing mechanism that orchestrates innate immune activation in response to cellular stress. In the central nervous system (CNS), this pathway demonstrates highly context-specific and cell-type-dependent functions, ranging from promoting neuroinflammation in neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD), to modulating immune surveillance and therapeutic responsiveness in glioma. This review systematically delineates the molecular mec…

A Bimodal MRI-Traceable Nanozyme with Immune-Activated Capability for Immunotherapy of Lung Metastases.

The therapeutic efficacy of cancer is greatly hampered by hypoxia, overexpressed glutathione (GSH) in the tumor microenvironment (TME), and the high propensity for tumor metastasis. Developing advanced diagnostic and therapeutic strategies capable of destroying tumors holds great promise. Herein, multifunctional manganese-based nanoparticles (FMBI NPs) with multiple nanozyme activities and GSH-depleting capabilities were designed to enhance cancer therapy by remodeling the TME. The release of Mn ions from FMBI NPs under TME conditions can modulate the tumor stroma, activate the cyclic GMP-AMP …
