Be Aware of Aggregators in the Search for Potential Human ecto-5'-Nucleotidase Inhibitors.

Promiscuous inhibition due to aggregate formation has been recognized as a major concern in drug discovery campaigns. Here, we report some aggregators identified in a virtual screening (VS) protocol to search for inhibitors of human ecto-5'-nucleotidase (ecto-5'-NT/CD73), a promising target for several diseases and pathophysiological events, including cancer, inflammation and autoimmune diseases. Four compounds (A, B, C and D), selected from the ZINC-11 database, showed IC50 values in the micromolar range, being at the same time computationally predicted as potential aggregators. To confirm if

The Prescription Pattern of Acetaminophen and Non-Steroidal Anti-Inflammatory Drugs in Patients with Liver Cirrhosis.

Analgesics, known to be hepatotoxic drugs, are frequently prescribed to patients with liver cirrhosis who are prone to drug-induced liver injury. No guidelines are available regarding the prescription of analgesics in these patients. Therefore, we aimed to evaluate the prescription pattern of most frequently used analgesics in patients with cirrhosis. We assessed the prescription pattern of acetaminophen and non-steroidal anti-inflammatory drugs (NSAIDs) in patients with liver cirrhosis registered in Health Insurance Review Assessment Service database between January 1, 2012 and December 31, 2

Angiotensin-I-converting enzyme inhibitory peptides: Chemical feature based pharmacophore generation.

A validated 3D pharmacophore model was generated for a series of ACE inhibitory peptides, which consisted of five features (two hydrophobic functions, two hydrogen bond acceptors, and a negative ionizable function). The built model was able to correctly predict the activity of known ACE inhibitors. The model was then used as query to search 3D databases of peptides. Three novel peptides (I, II and III) were synthesized and biologically evaluated in vitro. It appears that the in vitro activity of peptides I, II and III was consistent with their molecular modeling results. Our results provided c

[Regulatory role and anticarcinogenic properties of certain vitamins' active derivatives and vitamin-like substances].

Vitamins are low-molecular compounds consisting of an essential part of the human diet. After entering the human organism vitamins or their precursors can undergo chemical modification, changing their biological properties and regulatory activity. For many decades, vitamins were considered mainly as precursors of enzyme cofactors, and hypovitaminosis was interpreted as a deficiency of a particular metabolite resulting from enzyme's insufficient activity. However, with the development of molecular biology techniques and breakthrough in understanding of gene expression regulation and cell signal

An interspecies correlation estimation (ICE) model to predict acute toxicity of organic chemicals to the African clawed frog (Xenopus laevis) using rainbow trout (Oncorhynchus mykiss) data.

An interspecies correlation estimation (ICE) model is presented to predict the 96-h acute LC50 for tadpoles of the African clawed frog (Xenopus laevis) using juvenile rainbow trout (Oncorhynchus mykiss) 96-h LC50 values. Both species are standard laboratory test species with a long history in aquatic toxicology. While the rainbow trout is one of the fish species most commonly used for acute testing, the African clawed frog is primarily used in endocrine assays. However, acute toxicity information for X. laevis may be required for test concentration setting in the endocrine assays and, where re

QSAR meets ecology: Predictive framework for assessing pesticide toxicity against mayfly using consensus modelling.

The extensive use of pesticides in agriculture holds serious toxicological risks to aquatic and terrestrial ecosystems. Traditional toxicity assessment methods are costly, time-consuming, and ethically constrained due to animal testing requirements. In this context, quantitative structure-activity relationship (QSAR) offers a distinct benefit to forecast the toxicity of substances. The current study builds partial least squares (PLS)-based QSAR models for evaluating the detrimental impacts of chemicals against mayfly (Ephemera vulgata) species, following OECD rules and using LD50 as a specifie
