Impact of Deafness on the Lateralized Brain Responses to Letters and Digits: A Fast Periodic Visual Stimulation Exploratory Study in Deaf and Hearing Children.

Numbers and letters are culturally created symbols that acquire meaning through extensive training, significantly influencing brain function. The distinct hemispheric specialization of cortical regions for these categories has been hypothesized to relate to the co-activated brain networks: the left language regions for letters, and the right intra-parietal sulcus for numbers. However, the potential influence of deafness and sign language on hemispheric specialization for letters and numbers remains unclear. The present study aims to explore this issue by using a FPVS-EEG approach with an oddba…

[Hearing phenotype and genotypic characteristics in GSDME-related deafness pedigrees].

This retrospective study analyzed three GSDME-related deafness pedigrees diagnosed at the Chinese PLA General Hospital between January 2014 and December 2020. Through whole-exome sequencing and Sanger sequencing validation, all three pedigrees were found to carry splice-site variants causing exon 8 skipping. The patients presented with bilateral, symmetrical, and progressive sensorineural hearing loss, with an age of onset ranging from five to 25 years. The hearing loss initially affected high frequencies and gradually progressed to involve all frequencies. Significant phenotypic heterogeneity…

Spectrum of DNA Variants Underlying Deafness in an Ecuadorian Cohort.

The genetic etiology of hearing loss (HL) in Ecuador remains largely unexplored. This study investigates the spectrum of genetic variants associated with HL in a cohort of 66 Ecuadorian families using Exome or Genome sequencing (ES/GS). We identified pathogenic and likely pathogenic variants underlying HL in 27 families (41%). While variants were detected in 15 different genes, only GJB2 (in 12 families) and TMC1 (in 3 families) variants were identified in more than one family. The NM_004004.6 (GJB2):c.19 C > T (p.Gln7*) and NM_004004.6 (GJB2):c.35delG (p.Gly12Valfs*2) variants were more commo

Spatial hearing adaptation in congenital and acquired single-sided deafness.

INTRODUCTION: Spatial hearing depends on binaural integration of interaural time and level differences and monaural spectral cues. Single-sided deafness (SSD) disrupts this process, impairing localization and speech perception. Congenital (SSDc) and acquired (SSDa) SSD provide unique models for studying cortical adaptation, while cochlear implantation (CI) offers partial restoration of binaural hearing. This study examined differences in localization performance and response promptness across SSDc, SSDa, and SSD-CI listeners. METHODS: Thirty-one SSD listeners (9 SSDc, 11 SSDa, and 11 SSD-CI) …

Limited Potential to Reverse Deafness Caused by Mutation of Myo7a.

PURPOSE: MYO7A is involved in several forms of deafness in humans and mice, and in this study we aimed to investigate if the hearing loss could be reversed after its onset. METHODS: A knockdown allele of Myo7a in the mouse, Myo7atm1a, was characterised by recording ABR thresholds at ages from 4 weeks to 6 months old and measuring the amount of hair cell degeneration at 4 weeks old. Scanning electron microscopy was used to assess the condition of stereocilia bundles. A tamoxifen-inducible Flp recombinase was used to activate expression of Myo7a in Myo7atm1a/tm1a homozygotes at 4 weeks old by e…

A Case of Bilateral Sudden Deafness Caused by Wernicke Encephalopathy.

Wernicke encephalopathy, also known as thiamine deficiency, is characterized by a triad of symptoms: ophthalmoplegia, ataxia, and confusion. However, additional symptoms may manifest depending on the area affected by the lesion. Although multiple cranial neuropathies are possible, sudden onset bilateral hearing loss has been described in rare situations as the first manifestation of Wernicke encephalopathy. Here, we present a case report detailing the clinical experience of a patient diagnosed with Wernicke encephalopathy, whose initial presentation included sudden bilateral deafness. The pati…

Do Otologists and Other Otolaryngologists Manage Single-Sided Deafness Differently?

BACKGROUND: The aim of this study was to survey the knowledge and treatment management practices for single-sided deafness (SSD) among different subspecialties of otolaryngology. METHODS: A questionnaire was sent via Google Sheets to members of the Turkish and Egyptian Otorhinolaryngology Societies between December 2021 and February 2022. For the statistical analysis, the respondents were divided into 3 groups as otologists, non-otologists, and residents at the department of otolaryngology-head and neck department. RESULTS: There were no statistically significant differences between otologis…

A Multimodal Investigation of Listening Effort in Single-Sided Deafness.

PURPOSE: For patients with single-sided deafness (SSD), choosing between bone conduction devices (BCDs) and contralateral routing of signal hearing aids (CROS) is challenging due to mixed evidence on their benefits. The lack of clear guidelines complicates clinical decision making. This study explores whether realistic spatial listening measures can reveal a clinically valid benefit and if the optimal choice varies among patients. By assessing listening effort through objective and subjective measures, this research evaluates the efficacy of BCD and CROS, seeking to provide evidence-based reco…

AAV-mediated Gene Therapy for Hereditary Deafness: Progress and Perspectives.

Hereditary deafness is the most prevalent sensory deficit disorder, with over 100 identified deafness-related genes. Clinical treatment options are currently limited to external devices like hearing aids and cochlear implants. Gene therapy has shown promising results in various genetic disorders and has emerged as a potential treatment for hereditary deafness. It has successfully restored hearing function in >20 types of genetic deafness model mice and can almost completely cure patients with hereditary autosomal recessvie deafness 9 (DFNB9) caused by the OTOFERLIN (OTOF) mutation, thus servin…

Abnormal electrical brain responses to time deviance in beat deafness.

Humans have the spontaneous capacity to track the beat of music. Yet some individuals show marked difficulties. To investigate the neural correlates of this condition known as beat deafness, the cortical electric activity of ten beat-deaf adults, the largest cohort studied so far, as well as of 14 matched controls (Experiment 2), and 16 university students (Experiment 1) were examined. All were actively engaged in detecting anisochronous time-deviants in otherwise isochronous, metronome-like, sequences. As expected, participants with beat-deafness performed more poorly than controls; this beha…

Deafness gene screening based on a multilevel cascaded BPNN model.

Sudden sensorineural hearing loss is a common and frequently occurring condition in otolaryngology. Existing studies have shown that sudden sensorineural hearing loss is closely associated with mutations in genes for inherited deafness. To identify these genes associated with deafness, researchers have mostly used biological experiments, which are accurate but time-consuming and laborious. In this paper, we proposed a computational method based on machine learning to predict deafness-associated genes. The model is based on several basic backpropagation neural networks (BPNNs), which were casca…

Imeglimin-mediated glycemic control in maternally inherited deafness and diabetes.

Mitochondrial dysfunction causes maternally inherited deafness and diabetes (MIDD). Herein, we report improved glycemic control in a 47-year-old Japanese woman with MIDD using imeglimin without major adverse effects. Biochemical tests and metabolome analysis were performed before and after imeglimin administration. Blood glucose level fluctuations were determined. Sulfonylureas, dipeptidyl peptidase-4 inhibitors (DPP4is), and sodium glucose transporter-2 inhibitors (SGLT2i) were administered to evaluate the efficacy of their combination with imeglimin. Imeglimin decreased the HbA1c and ammonia…
