Engineering CAR-Macrophages With Advanced Delivery Systems for Tissue Repair.

Tissue injury and organ dysfunction remain major clinical challenges, as conventional therapies often fail to achieve functional regeneration. Chimeric antigen receptor (CAR) technology endows macrophages with the ability to specifically recognize and clear pathological targets, making CAR-macrophages (CAR-M) a promising tool in tissue engineering and regenerative medicine. However, the efficient, safe, and controllable engineering of CAR-M still depends on advanced chemical delivery systems. This review systematically summarizes five major platforms for CAR-M engineering, including viral vect…

In vivo engineering of CAR immune cells by nonviral nanoparticles.

Chimeric antigen receptor (CAR) immune cell therapy has revolutionized the treatment of hematologic malignancies, yet conventional ex vivo manufacturing remains costly, complex, and logistically demanding. Emerging in vivo engineering strategies, especially those based on nonviral nanoparticles (NPs), offer a transformative alternative by enabling direct programming of immune cells within the body. Recent advances in NPs-mediated delivery of CAR constructs, including mRNA, circular RNA (circRNA), self-amplifying RNA (saRNA), DNA and gene editing tools, have demonstrated promising potential for…

Pancreatic tissue engineering: towards a vascularized bioengineered cure for diabetes.

UNLABELLED: Type 1 diabetes results from autoimmune destruction of pancreatic β-cells, and while whole-organ pancreas and islet transplantation can restore endogenous insulin secretion, both are limited by donor scarcity, early inflammatory injury, hypoxia, and the need for lifelong immunosuppression. Pancreatic tissue engineering has therefore emerged as a strategy to generate scalable, transplantable constructs capable of reestablishing physiological glucose regulation. However, as native islets depend on a robust microvascular network for oxygen delivery, nutrients and coordinated hormone s…

Designing of porous scaffolds for tissue engineering and regenerative medicine.

The potential of the porous scaffolds lies in its dynamic architectural features, such as porosity, pore size & shape, interconnectivity, and spatial distribution which are highly essential in tissue engineering applications. The porous structure of the scaffolds influences the biological and physicochemical activities in the tissue regeneration process. The porosity also allows better distribution of nutrients and diffusion of biological fluids thereby dispersing the seeding material uniformly. Generally, damaged tissues can be regenerated on their own but many times due to unavoidable factor…

Engineering nonconventional yeasts to produce carboxylic acids: Advances and Prospects.

Due to their versatile carboxy group, carboxylic acids can be used as bio-based monomers for high-value-added industrial products with great market demand. In recent years, metabolic engineering of nonconventional yeasts to produce carboxylic acids from renewable raw materials is showing increasing promise. In this review, we first summarize the characteristics of nonconventional yeasts (e.g., Yarrowia lipolytica, Candida tropicalis), and then focus on metabolic engineering strategies for the synthesis of mono-, di- and tricarboxylic acids. Finally, the development trends of low-cost, green an…

Engineering inflammation-responsive proteins through nitric oxide-caged amino acids.

Nitric oxide is upregulated in inflammatory tissues but has not been used to directly control the activity of folded proteins. Here we report a protein engineering strategy that enables selective restoration of protein function in nitric oxide-rich environments. Protein activity is temporarily suppressed by site-specific substitution of a catalytically or structurally essential glutamate residue with a synthetic amino acid whose side chain is chemically masked. Exposure to nitric oxide triggers decaging of this residue, regenerating the native glutamate and restoring protein function. Using th…

Beyond rejection: engineering immune responses to redefine skin graft survival.

Skin grafting remains a fundamental reconstructive technique in reconstructive and dermatologic surgery for restoring skin integrity following burns, trauma, chronic ulcers, and excision of malignancies. However, a graft survival rate is determined by multiple factors and by a delicate balance between inflammatory and reparative immune system processes. Controlled inflammation promotes wound healing while excessive or prolonged immune activation has the potential to result in cytotoxicity and complement activation which will eventually lead to graft rejection. Early rejection is driven by anti

In vivo immune cell engineering from bench to clinical reality.

Adoptive immune cell therapies, exemplified by chimeric antigen receptor T cells, have transformed the treatment of hematological malignancies. However, their broader clinical application is limited by complex ex vivo manufacturing, high cost, and safety concerns. In vivo immune cell engineering has emerged as an alternative strategy that delivers genetic instructions directly to immune cells, thereby generating or modulating therapeutic immune cells within the body and reducing the reliance on individualized in vitro operations. These advances underscore the need for a systematic evaluation o…

Engineering suppressor tRNAs for effective treatment of Duchenne muscular dystrophy.

Duchenne muscular dystrophy (DMD) is a fatal disorder caused by loss of dystrophin, a protein essential for muscle cell integrity. To date, no therapeutic has restored full-length dystrophin. Suppressor transfer RNAs (sup-tRNAs) have the potential to treat the ∼15% of patients with DMD and nonsense mutations. We have evolved highly potent UAA sup-tRNAs through an extensive mutagenesis screening campaign, optimized our adeno-associated virus (AAV) expression vectors, and developed efficient RNA polymerase III promoters. We show that systemic delivery of these sup-tRNAs restores full-length dyst

Non-invasive ovulation tracking enables genetic engineering in wild rodents.

Many non-model rodent species are inaccessible to genetic engineering due to our limited understanding of their reproductive biology. Here, we present a low-cost, camera-based estrous-tracking technology that enables transgenesis in the white-footed mouse Peromyscus leucopus, a key reservoir for Lyme disease. We demonstrate the efficient generation of pregnant and pseudopregnant mice via timed ovulation, provide protocols for embryo generation, cultivation, microinjection, and transplantation as well as an accurate developmental timeline, and report the first engineered Peromyscus. The same te…

Current Collector Engineering for New Efficient Bioresorbable Sodium-Ion Batteries.

Developing bioresorbable energy sources that eliminate the need for secondary surgery for device removal remains a major challenge in resorbtronics. In this work, we present an alternative fabrication strategy for new bioresorbable quasi-solid Na-ion batteries using Mo or Mg thin films as current collectors. The assembled batteries were characterized using scanning electron microscopy, spectroscopic techniques including X-ray photoelectron spectroscopy, physicochemical, and electrochemical characterization techniques. Results demonstrate that the choice of current collector strongly influences

Advancing recombinant protein production in CHO cells through metabolic engineering.

Chinese hamster ovary (CHO) cells serve as the predominant platform for producing recombinant therapeutic proteins in biopharmaceutical manufacturing, the production capacity of which relies heavily on efficient protein synthesis, folding, and secretion pathways. However, during high-density and prolonged cultivation, these cells frequently encounter bottlenecks-including excessive lactate and ammonia accumulation, redox imbalance, and endoplasmic reticulum (ER) stress-which ultimately constrain both the yield and quality of target protein. To overcome these limitations, metabolic engineering …
