Neurodevelopmental involvement in schizophrenia: the olfactory epithelium as an alternative model for research.

It has been suggested that disturbances during neurodevelopment may play a crucial role in the etiology of schizophrenia (SZ). This premise is supported by brain imaging, epidemiological, and pathological studies as well as the discovery of susceptibility genes for SZ that appear to be implicated in development of the central nervous system. Here, we discuss the limitations of the current methods and models for studying the neurodevelopmental implications in SZ. We agree with the proposal that the olfactory epithelium, in which neurodevelopment continues throughout life, might represent an alt

The roles of vitamin A in the prevention of neoplastic diseases: a critical assessment.

The role of vitamin A and its analogs (retinoids) in the prevention and therapy of neoplastic diseases is discussed. Epidemiological data showing a relationship between vitamin A deficiency and increased frequency of tumors in man and animals are examined and the experimental models demonstrating the protective action of retinoids in the respect of both spontaneous and induced neoplasias of animals are reviewed. Among the possible mechanisms responsible of the antineoplastic activity, the immunopotentiating effect is underlined. Some toxicological aspects of retinoids administration and the co

Mathematical Modeling for Scrub Typhus and Its Implications for Disease Control.

BACKGROUND: The incidence rate of scrub typhus has been increasing in the Republic of Korea. Previous studies have suggested that this trend may have resulted from the effects of climate change on the transmission dynamics among vectors and hosts, but a clear explanation of the process is still lacking. In this study, we applied mathematical models to explore the potential factors that influence the epidemiology of tsutsugamushi disease. METHODS: We developed mathematical models of ordinary differential equations including human, rodent and mite groups. Two models, including simple and comple

An approach to the epidemiology of childhood injuries.

Injury epidemiology is summarized and important concepts such as injury versus accident, the agent-host-environment model, level of severity, morbidity versus mortality, and definitions of rates are considered. The authors suggest steps needed to improve the understanding of injury epidemiology and to remove barriers to the acceptance of injury prevention as a recognized field of study and clinical practice.

Methods for reproducible comparison of strategies in stochastic modelling.

Stochastic simulations are often used to model real-world phenomena such as infectious disease dynamics. In this modelling, differing strate … d show when each of our three proposed methods ought to be used. We illustrate our methods with two epidemiological models: one simple model of a vaccine-preventable infection and one complex model of African sleeping sickness, which can be controlled through multiple interventions. We show how our Bernoulli hashing method works very well for simple models, and a variat …

Modeling environmental inhalant exposure in rheumatoid arthritis.

The mucosal origins hypothesis posits that environmental inhalant exposures, including cigarette smoke (CS) and crystalline silica (c-silica), trigger immune responses in the lung mucosa, an extra-articular site, which precede initiating events of rheumatoid arthritis (RA) pathogenesis in distant joints. Epidemiological data strongly associates these exposures with RA risk, especially in genetically susceptible individuals carrying HLA-DRB1 alleles, and with the production of autoantibodies such as anti-citrullinated peptide antibodies (ACPA) and rheumatoid factor (RF). However, establishing c

Influence of demographic changes on the impact of vaccination against varicella and herpes zoster in Germany - a mathematical modelling study.

BACKGROUND: Epidemiological studies suggest that reduced exposure to varicella might lead to an increased risk for herpes zoster (HZ). Reduction of exposure to varicella is a consequence of varicella vaccination but also of demographic changes. We analyzed how the combination of vaccination programs and demographic dynamics will affect the epidemiology of varicella and HZ in Germany over the next 50 years. METHODS: We used a deterministic dynamic compartmental model to assess the impact of different varicella and HZ vaccination strategies on varicella and HZ epidemiology in three demographic

On the risk of severe dengue during secondary infection: a systematic review coupled with mathematical modeling.

BACKGROUND & OBJECTIVES: The present study aimed to systematically quantify the well known risk of severe dengue during secondary infection in literature and to understand how epidemiological mechanisms of enhancement during the secondary infection influence the empirically estimated risk of severe dengue by means of mathematical modeling. METHODS: Two conditional risks of severe dengue, i.e. symptomatic illness and dengue hemorrhagic fever (DHF) or dengue shock syndrome (DSS), given secondary infection were explored based on systematically searched prospective studies. A two-strain epidemiol

Predicting outcome after acute and subacute stroke: development and validation of new prognostic models.

BACKGROUND AND PURPOSE: Statistical models to predict the outcome of patients with acute and subacute stroke could have several uses, but no adequate models exist. We therefore developed and validated new models. METHODS: Regression models to predict survival to 30 days after stroke and survival in a nondisabled state at 6 months were produced with the use of established guidelines on 530 patients from a stroke incidence study. Three models were produced for each outcome with progressively more detailed sets of predictor variables collected within 30 days of stroke onset. The models were exte

Evolution of the "fourth stage" of epidemiologic transition in people aged 80 years and over: population-based cohort study using electronic health records.

BACKGROUND: In the "fourth stage" of epidemiological transition, the distribution of non-communicable diseases is expected to shift to more advanced ages, but age-specific changes beyond 80 years of age have not been reported. METHODS: This study aimed to evaluate demographic and health transitions in a population aged 80 years and over in the United Kingdom from 1990 to 2014, using primary care electronic health records. Epidemiological analysis of chronic morbidities and age-related impairments included a cohort of 299,495 participants, with stratified sampling by five-year age group up to

The perfect food and the filth disease: milk-borne typhoid and epidemiological practice in late Victorian Britain.

This article explores the initial set of epidemiological investigations in Victorian Britain that linked typhoid fever to milk from dairy cattle. Because Victorian epidemiologists first recognized the milk-borne route in outbreaks of typhoid fever, these investigations served as a model for later studies of milk-borne scarlet fever, diphtheria, and perhaps tuberculosis. By focusing on epidemiological practices conducted by Medical Inspectors at the Medical Department of the Local Government Board and Medical Officers of Health, I show that Victorian epidemiology was committed to field-based, o

Regression-based modeling of pairwise genomic linkage data identifies risk factors for healthcare-associated pathogen transmission: application to carbapenem-resistant Klebsiella pneumoniae transmission in a long-term care facility.

Pathogen whole-genome sequencing (WGS) has significant potential for improving healthcare-associated infection (HAI) outcomes. However, methods for integrating WGS with epidemiologic data to quantify risks for pathogen spread remain underdeveloped. To identify analytic strategies for conducting WGS-based HAI surveillance in high-burden settings, we modeled patient- and facility-level transmission risks of carbapenem-resistant Klebsiella pneumoniae (CRKP) in a long-term acute care hospital (LTACH). Using rectal surveillance data collected over 1 year, we fit three pairwise regression models wit

A review of mechanistic models of viral dynamics in bat reservoirs for zoonotic disease.

The emergence of SARS-CoV-2, a coronavirus with suspected bat origins, highlights a critical need for heightened understanding of the mechanisms by which bats maintain potentially zoonotic viruses at the population level and transmit these pathogens across species. We review mechanistic models, which test hypotheses of the transmission dynamics that underpin viral maintenance in bat systems. A search of the literature identified only twenty-five mechanistic models of bat-virus systems published to date, derived from twenty-three original studies. Most models focused on rabies and related lyssa

[Epidemiological surveillance: review of data from the Febrile Unit of the Muñiz Hospital 2020-2024].

The appearance of epidemiological events that threaten the health system marked the importance of responses that go beyond the dissemination of information and since the COVID-19 pandemic, the Government of the City of Buenos Aires (Argentina) proposed an epidemiological surveillance model in devices called Febrile Units (UF). At the Muñiz Hospital, this unit provided epidemiological surveillance strategies in the pandemic and the different epidemics and outbreaks, still continuing its operation today. The objective of this work is to evaluate the implementation of the Febril Unit as an epidem

Sustainable Coexistence with Infectious Diseases: A Behavioral Feedback Model Driven by Resource Accessibility under Static-Dynamic Optimal Control.

This study develops a behavior-disease feedback model grounded in imitation dynamics to analyze optimal long-term epidemic coexistence strategies from both static and dynamic perspectives. Analytical findings reveal that the stability of the system depends on both the basic reproduction number and the behavioral threshold, with the latter and risk attention jointly determining optimal static intervention intensity. Data-driven analyses of influenza (Shanxi) and COVID-19 (Shanghai) show that adaptive public behavior can drive recurrent epidemic waves. In dynamic optimization simulations involvi

Markovian Approach for Exploring Competitive Diseases with Heterogeneity-Evidence from COVID-19 and Influenza in China.

Due to the complex interactions between multiple infectious diseases, the spreading of diseases in human bodies can vary when people are exposed to multiple sources of infection at the same time. Typically, there is heterogeneity in individuals' responses to diseases, and the transmission routes of different diseases also vary. Therefore, this paper proposes an SIS disease spreading model with individual heterogeneity and transmission route heterogeneity under the simultaneous action of two competitive infectious diseases. We derive the theoretical epidemic spreading threshold using quenched m

Estimation of prediction error for survival models.

When statistical models are used to predict the values of unobserved random variables, loss functions are often used to quantify the accuracy of a prediction. The expected loss over some specified set of occasions is called the prediction error. This paper considers the estimation of prediction error when regression models are used to predict survival times and discusses the use of these estimates. Extending the previous work, we consider both point and confidence interval estimations of prediction error, and allow for variable selection and model misspecification. Different estimators are com
