Orexin excites GABAergic neurons of the arcuate nucleus by activating the sodium--calcium exchanger.

The neuropeptides orexins/hypocretins are essential for normal wakefulness and energy balance, and disruption of their function causes narcolepsy and obesity. Although much is known of the role of orexins in sleep/wake behavior, it remains unclear how they stimulate feeding and metabolism. One of the main targets of orexinergic neurons is the arcuate nucleus (ARC) of the hypothalamus, which plays a key role in feeding and energy homeostasis. By combining patch-clamp and RT-multiplex PCR analysis of individual neurons in mouse brain slices, we show that an electrophysiologically distinct subset

Effect of naloxone and antidepressants on hyperphagia produced by peptide YY.

Central injection of peptide YY (PYY) elicits a powerful feeding response with a short latency in satiated rats. Because of this effect, PYY has been implicated as a neurochemical signal in bulimia nervosa. Serotonin agonists and opioid antagonists induce anorectic effects upon feeding behavior in humans and animals. Therefore, to investigate a possible interaction between PYY-induced eating and these anorexigenic agents rats were given injections of either naloxone (100 micrograms/3 microliters, ICV, and 10 mg/kg, SC), fluoxetine (3-30 micrograms/3 microliters and 5-10 mg/kg, IP), or clomipra…

Reversal of phenylpropanolamine anorexia in rats by the alpha-1 receptor antagonist benoxathian.

Phenylpropanolamine (PPA) is a phenethylamine anorectic drug that exerts direct agonist effects predominantly on alpha-1 adrenergic receptor … agonist benoxathian (0, 2.5, 5.0 or 10.0 nmol) into the PVN was found to have no effect on baseline feeding behavior. Microinjection of 10.0 nmol benoxathian into the PVN completely reversed the anorexia induced by 2.5, 5.0 or 10.0 mg/kg PPA (IP), yet did not alter the hypodipsia produced by PPA. These data strongly suggest that PPA anorexia is mediated by an alph …

Behavioral, physiological, and endocrine responses of starlings to acute increases in density.

One potential stressor to vertebrates both in the wild and in captivity is the presence of numerous individuals in a confined space. To examine the effects of increased conspecific density in birds, we simultaneously measured cardiac, behavioral, and endocrine responses of European starlings (Sturnus vulgaris) to acute crowding. A cage containing a resident bird was outfitted with a trap door that allowed for the introduction of intruder birds (one, three, or five birds) without human interference. The resident bird was implanted with a subcutaneous heart rate (HR) transmitter, behavior was vi

Automated long-term two-photon imaging in head-fixed walking Drosophila.

BACKGROUND: The brain of Drosophila shows dynamics at multiple timescales, from the millisecond range of fast voltage or calcium transients to functional and structural changes occurring over multiple days. To relate such dynamics to behavior requires monitoring neural circuits across these multiple timescales in behaving animals. NEW METHOD: Here, we develop a technique for automated long-term two-photon imaging in fruit flies, during wakefulness and extended bouts of immobility, as typically observed during sleep, navigating in virtual reality over up to seven days. The method is enabled by

Breastfeeding Behavior in Brazilian Children with Congenital Zika Syndrome.

Congenital Zika Syndrome (CZS) is a condition that has emerged only recently, bringing together multiple changes, including significant changes in the stomatognathic system, which may compromise sucking behavior and consequently the breastfeeding practice. The aim of this study was to investigate the breastfeeding behaviors in children with CZS. A longitudinal study was carried out in two reference centers in Northeastern Brazil. The nonprobabilistic sample consisted of 79 children diagnosed with physical, neurological, and behavioral alterations compatible with CZS. Information regarding the …

Association between time budget of primiparous cows and their adaptation to an automatic milking system following parturition.

The objective of this study was to identify the differences in the transition period time budget of first-lactation cows that adapt quickly to a free-traffic automatic milking system (AMS) versus those that require more chronic fetching during the adaptation period following calving. A total of 22 primiparous Holstein cows were observed from ∼21 d before calving to 23 DIM, following their adaptation to an AMS. During the prepartum period, heifer time budgets were monitored for rumination, feeding activities, lying behavior, and standing behavior. These same behaviors were monitored in the post

The Impact of Lameness on Welfare of the Dairy Cow.

The five freedoms offer a framework for discussion of lameness and its impact on the welfare of cows. Altered feeding behavior is a cause of reduced body condition, smaller digital cushion, and lameness. Providing a comfortable environment is critical to recovery and welfare. Pain associated with injury or disease of feet or legs is manifested by lameness. Pain management is an important part of therapy. In cases of severe lameness, euthanasia may be preferred. Lameness interferes with an animal's ability to exhibit natural behaviors by altering lying time, social interaction, ovarian activity…

Low levels of chicken body louse (Menacanthus stramineus) infestations affect chicken welfare in a cage-free housing system.

BACKGROUND: The chicken body louse is an obligate ectoparasite of domestic chickens. Chicken body lice feed on feathers, and infestation with this louse is linked to decreases in egg production, hen weight, and feed conversion efficiency. However, it is unknown how chicken body lice impact egg-laying chickens in cage-free environments. Welfare and behavior metrics were collected from flocks of egg-laying chickens either infested with chicken body lice or left uninfested. METHODS: In two trials, two flocks of cage-free commercial egg-laying chickens were infested with chicken body lice or main

Hypothalamic neuronal histamine regulates feeding circadian rhythm in rats.

To clarify involvement of hypothalamic neuronal histamine in feeding circadian rhythm, we analyzed rat behavioral patterns using chemical probes which affect endogenous histaminergic activity. Sustained infusion of alpha-fluoromethylhistidine (FMH), a specific suicide inhibitor of a histamine-synthesizing enzyme, into the rat third cerebral ventricle disrupted light-dark cycles of feeding, drinking, and ambulatory behavior. Food and water intake and ambulatory activity during the 12-h light period increased, and those during the 12-h dark period decreased after the infusion. The ratio of the l

Exogenous estradiol does not regulate daily metabolic rhythms underlying diet-induced obesity in male mice.

Male mice fed high-fat diet become obese, but female mice are resistant to diet-induced weight gain. We previously found that circulating estradiol in females protects their daily rhythms from disruption by high-fat feeding to prevent diet-induced obesity. The goal of this study was to determine the effects of estradiol on daily metabolic rhythms in male mice. Male C57BL/6J mice were treated with estradiol and fed high-fat diet for 2 weeks. We measured the effects of high-fat diet feeding on daily rhythms of eating behavior and locomotor activity, and on the phases, or timing, of circadian rhy

Chronic activity-based anorexia alters food intake microstructure in a time-dependent manner in female rats.

Persistently disturbed eating behavior is a well-known feature of the severe eating disorder anorexia nervosa (AN) and contributes to high relapse rates. However, the underlying changes in eating microstructure are not well understood. This study employed the widely-used activity-based anorexia (ABA) model to examine feeding microstructure during acute and chronic starvation using an automated food intake monitoring system. After a 10-day acclimatization period, female adolescent Sprague-Dawley rats were assigned to the ad libitum (AL; ad libitum food, no wheel access), activity control (AC; a

Monitoring Multiple Behaviors in Beef Calves Raised in Cow-Calf Contact Systems Using a Machine Learning Approach.

The monitoring of pre-weaned calf behavior is crucial for ensuring health, welfare, and optimal growth. This study aimed to develop and validate a machine learning-based technique for the simultaneous monitoring of multiple behaviors in pre-weaned beef calves within a cow-calf contact (CCC) system using collar-mounted sensors integrating accelerometers and gyroscopes. Three complementary models were developed to classify feeding-related behaviors (natural suckling, feeding, rumination, and others), postural states (lying and standing), and coughing events. Sensor data, including tri-axial acce

Involvement of the histaminergic system in leptin-induced suppression of food intake.

The ob gene product leptin is secreted from white adipose tissue, and may regulate food intake by acting on the hypothalamus in the central nervous system. But the mechanism of this effect is still unclear. The central histaminergic system has been suggested to participate in the control of various physiological functions, particularly in feeding behavior, as it mediates anorectic signals like leptin. Thus, we hypothesized that the central histaminergic system is a target for leptin in its control of feeding. To prove this, we first examined the effect of i.p. administration of alpha-fluoromet…

Mechanical Behavior and Surface Properties of Silicone and PVC Gastric Tubes Under Controlled pH Conditions.

In this study, the mechanical behavior, wettability, and surface free energy (SFE) of gastric feeding tubes made of silicone and poly(vinyl chloride) (PVC) were investigated under controlled pH conditions and varying exposure times. Wettability was evaluated using contact angle measurements, and SFE was determined using the Owens-Wendt-Rabel-Kaelble (OWRK), Wu, and van Oss-Chaudhury-Good (vOCG) approaches. Mechanical behavior was analyzed by static tensile testing and stopper pull-off tests. The results showed distinct differences between the materials. PVC tended to exhibit a more stable mech
