Neuroimaging in the developmental disorders: the state of the science.

The developmental disorders of childhood autistic, developmental language, reading (dyslexia), and attention deficit-hyperactivity disorders … brain-behavior correlates in these disorders. The results of these neuropathologic, structural, and functional neuroimaging studies are presented and the reasons for the often divergent findings are discussed. As we approach the end of the Decade of the Brain, current neuroimaging techniques give us the technology for the first time to apply a fundamental cognitive appro …

Evaluating clinical and neuroimaging predictors for cognitive-behavioral therapy outcome in obsessive-compulsive disorder.

Cognitive-behavioral therapy (CBT) is the first-line treatment for obsessive-compulsive disorder (OCD), yet a significant number of patients do not achieve remission or substantial symptom relief. This study aims to enhance the prediction of CBT outcomes in OCD by integrating demographic, clinical, and neuroimaging data using machine learning (ML) models. We conduct a comprehensive analysis on a well-characterized clinical sample, employing a rigorous validation scheme to avoid data leakage, and comparing multiple ML algorithms to minimize bias. Out of four different ML models trained on demog

Mapping neural signatures of face processing in young children: an OPM-MEG study.

Facial expressions are fundamental to social communication, with emotional face processing developing throughout childhood. However, the neural mechanisms underlying this process in young children remain underexplored due to challenges in neuroimaging this population. Optically pumped magnetometers (OPMs), a wearable magnetoencephalography (MEG) technology, offer potential advantages for studying these responses early in life. This study investigated evoked responses and functional connectivity in 45 children (3-5 years) during an emotional face processing task using OPM-MEG. The M170 componen

Fronto-Parietal and Language Network Connectivity and Its Association With Gene Expression Profiles in Bipolar Disorder Before and After Treatment.

BACKGROUND: The resting-state functional connectivity (FC) patterns of the fronto-parietal network (FPN) and language network (LN) underlying bipolar disorder (BD) are obscure. This study aimed to uncover abnormal FC patterns of FPN and LN underlying BD and their evolution following treatment. METHODS: Imaging data at rest state and clinical variables were acquired from 82 patients with BD (with 43 finishing the follow-up) and 88 healthy controls (HCs). Seed-based FC analysis was performed, and correlations between FCs and clinical variables were investigated with whole-brain multiple regress

Differential Functional Connectivity Between Silent Reading and Resting-State fMRI and Their Relationships With Reading Performance in Children With and Without Dyslexia.

The clinical diagnostic parameters of dyslexia and the alterations it creates on brain connectivity continue to be the focus of current studies. In this study, where clinical neuropsychiatric tests and neuroimaging results are examined together, it is aimed to examine the functional connectivity differences that occur in the brain regions related to the reading pathway in children with dyslexia in a resting state and task-based manner and to compare them with healthy controls. Correlation analyses were performed to evaluate the correlation between the resting state and the reading task of the

Heterozygous truncating variants in SUFU cause congenital ocular motor apraxia.

PURPOSE: This study aimed to delineate the genetic basis of congenital ocular motor apraxia (COMA) in patients not otherwise classifiable. METHODS: We compiled clinical and neuroimaging data of individuals from six unrelated families with distinct clinical features of COMA who do not share common diagnostic characteristics of Joubert syndrome or other known genetic conditions associated with COMA. We used exome sequencing to identify pathogenic variants and functional studies in patient-derived fibroblasts. RESULTS: In 15 individuals, we detected familial as well as de novo heterozygous trun

Neuroimaging diagnosis in neurodegenerative diseases.

Dementia affects about 8% of people age 65 years and older. Identification of dementia is particularly difficult in its early phases when family members and physicians often incorrectly attribute the patient's symptoms to normal aging. The most frequently occurring ailments that are connected with neurodegeneration are: Alzheimer's disease, Parkinson's disease, Huntington's disease, amyotrophic lateral sclerosis, and multiple sclerosis. A variety of powerful techniques that have allowed visualization of organ structure and function with exact detail have been introduced in the last twenty-five

A DTI investigation of neural substrates supporting tool use.

Recent functional neuroimaging and brain lesion studies have implicated a network of left hemisphere regions in human tool use: 1) posterior middle temporal cortex involved in conceptual knowledge of tools, 2) posterior inferior parietal cortex for representations of learned tool use gestures, and 3) anterior inferior parietal cortex, along with posterior inferior frontal and ventral premotor cortices, involved in grasping and manipulating objects. Here, we use diffusion tensor imaging (DTI) to investigate the anatomical connections that support this putative network. DTI scans were acquired f…

Amblyopia: progress and promise of functional magnetic resonance imaging.

Amblyopia is a neurodevelopmental disorder characterized by functional deficits in the visual cortex. Functional magnetic resonance imaging (fMRI) is the most commonly used neuroimaging technique for investigating amblyopia. Herein, we systematically searched a PubMed database from inception to December 2021 to highlight the current progress and promises about fMRI technology in amblyopia; amblyopia's neural mechanism, the comparison of different types of amblyopia, and the evaluation of the therapeutic effect were explored. Study protocol for neuroimaging using 7 T MRI in the investigation of baricitinib for reduction of HIV in the CNS: a randomized placebo-controlled trial.

BACKGROUND: While antiretroviral therapy is successful in conferring peripheral human immunodeficiency virus type 1 (HIV-1) suppression, eradication of HIV-1 reservoirs in the central nervous system (CNS) is an immediate goal in the management of HIV-1 infection. The primary goal of this randomized and placebo-controlled Phase IIb trial is to evaluate the effects of baricitinib versus placebo on HIV-1 CNS persistence markers in a ten-week mechanistic study, with secondary endpoints evaluating the effects on peripheral blood, neuroimaging, and neuropsychological measures. Given the large scope

Effects of continuous positive airway pressure on cognitition and neuroimaging data in sleep apnea.

Obstructive sleep apnea (OSA) has been associated with a broad range of neurocognitive difficulties. The current view is that the neurocognitive impairment in OSA is due to the adverse effects of sleep fragmentation and/or intermittent hypoxia. The overall picture of cognitive deficits in OSA is complex. On balance, there appears to be negative effects of OSA on cognition, most likely in the domains of attention/vigilance, verbal and visual delayed long-term memory, visuospatial/constructional abilities, and executive dysfunction. Continuous positive airway pressure (CPAP) is the most effectiv

Structural and Functional Brain Features in Takotsubo Syndrome.

Takotsubo syndrome is featured by transient left ventricular dysfunction typically triggered by emotional or physical stress. Growing evidence supports a pivotal role of brain-heart interactions in its pathophysiology. Structural neuroimaging studies have identified reduced cortical thickness and gray matter volume in limbic and autonomic regulatory regions, including the insula, cingulate cortex, and amygdala. Functional imaging studies, including 18F-fluorodeoxyglucose (FDG) PET and functional MRI, have demonstrated persistent amygdalar hyperactivity, altered limbic system activity, and wide

Supraspinal Mechanisms Underlying Ocular Pain.

Supraspinal mechanisms of pain are increasingly understood to underlie neuropathic ocular conditions previously thought to be exclusively peripheral in nature. Isolating individual causes of centralized chronic conditions and differentiating them is critical to understanding the mechanisms underlying neuropathic eye pain and ultimately its treatment. Though few functional imaging studies have focused on the eye as an end-organ for the transduction of noxious stimuli, the brain networks related to pain processing have been extensively studied with functional neuroimaging over the past 20 years.…

Movement Disorders Following Cerebrovascular Lesions in Cerebellar Circuits.

Cerebellar circuitry is important to controlling and modifying motor activity. It conducts the coordination and correction of errors in muscle contractions during active movements. Therefore, cerebrovascular lesions of the cerebellum or its pathways can cause diverse movement disorders, such as action tremor, Holmes' tremor, palatal tremor, asterixis, and dystonia. The pathophysiology of abnormal movements after stroke remains poorly understood. However, due to the current advances in functional neuroimaging, it has recently been described as changes in functional brain networks. This review d…

The neural correlates of impaired consciousness in coma and unresponsive states.

Over the last few years, functional neuroimaging studies have provided new insights into cerebral activity in subjects with severe brain damage leading to coma and other clinical states characterized by unresponsiveness. The present paper introduces the clinical picture of patients with impaired consciousness, and reviews the nosological criteria and functional neuroanatomical basis for brain death, coma, vegetative state, minimally conscious state, and the locked-in syndrome. Converging evidence suggests that disrupted activity in higher-order association areas, especially prefrontal and post…

A role for the cerebellum in motor speech planning: evidence from foreign accent syndrome.

A 3 year follow-up study was performed in a patient with foreign accent syndrome (FAS) as the sole cognitive manifestation of a left fronto-parietal stroke. The hypothesis of involvement of the right cerebellum in this motor speech planning disorder was investigated by means of functional neuroimaging (SPECT) and neurobehavioral assessments. Based on the close parallelism between the evolution of FAS symptoms and the perfusional changes in the right cerebellum, it is argued that FAS may result from a disruption of a close functional interplay between the supra- and infratentorial speech center…

A Prognostic Neonatal Neuroimaging Scale for Symptomatic Congenital Cytomegalovirus Infection.

BACKGROUND: Congenital cytomegalovirus (cCMV) can cause brain inflammation/destruction and teratogenic effects. The only validated neuroimaging prognostic categorization for symptomatic cCMV available is based on destructive lesions seen on computed tomography (CT). OBJECTIVE: The aim of this study was to establish the predictive ability of a comprehensive neonatal neuroimaging scale in symptomatic cCMV. METHODS: Twenty-six infants were studied by neonatal cranial ultrasound scans (US; n = 25), CT (n = 11) and magnetic resonance imaging (MRI; n = 9). A previously validated neuroimaging scale

Engaging Mood Brain Circuits with Psilocybin (EMBRACE): a study protocol for a randomized, placebo-controlled and delayed-start, neuroimaging trial in depression.

BACKGROUND: Major depressive disorder (MDD) is a leading cause of disability worldwide across domains of health and cognition, affecting overall quality of life. Approximately one third of individuals with depression do not fully respond to treatments (e.g., conventional antidepressants, psychotherapy) and alternative strategies are needed. Recent early phase trials suggest psilocybin may be a safe and efficacious intervention with rapid-acting antidepressant properties. Psilocybin is thought to exert therapeutic benefits by altering brain network connectivity and inducing neuroplastic changes

Train the brain with music (TBM): brain plasticity and cognitive benefits induced by musical training in elderly people in Germany and Switzerland, a study protocol for an RCT comparing musical instrumental practice to sensitization to music.

BACKGROUND: Recent data suggest that musical practice prevents age-related cognitive decline. But experimental evidence remains sparse and no concise information on the neurophysiological bases exists, although cognitive decline represents a major impediment to healthy aging. A challenge in the field of aging is developing training regimens that stimulate neuroplasticity and delay or reverse symptoms of cognitive and cerebral decline. To be successful, these regimens should be easily integrated in daily life and intrinsically motivating. This study combines for the first-time protocolled music
