Unveiling Novel Lytic Bacteriophages as Natural Biocontrol Agents Against Multidrug-Resistant Escherichia coli: Isolation, Characterization, and In vitro Application.

The alarming findings presented in the latest WHO report on the global antimicrobial resistance crisis have redirected scientific attention toward phage-based approaches as a renewed line of defense against multidrug-resistant (MDR) bacteria. In this study, four bacteriophages infecting a MDR-Escherichia coli strain were isolated from water sources and subjected to detailed phenotypic and genomic characterization. All phages efficiently inhibited MDR-E. coli at MOIs of 0.1 and 0.01, showing high stability across a broad temperature (4–65 °C) and pH (4–10) range. TEM analysis revealed that all

Combating multidrug-resistant uropathogenic E. coli using lytic phages, enhancing antibiotic synergy and inhibiting biofilms.

The global emergence of multidrug-resistant Escherichia coli (MDR E. coli), driven by excessive antibiotic use and environmental persistence, poses a major threat to public health. Bacteriophages (phages) have garnered renewed interest as targeted biocontrol agents against such pathogens. In this study, we isolated and characterized two lytic phages, EC.W1-1 and EC.W15-3, specifically targeting diverse sequence types (STs) of MDR E. coli. Both phages belong to the family Straboviridae and genus Tequatrovirus. They demonstrated remarkable stability across pH 2-10 (4 h) and temperatures below 80

High functional P-glycoprotein activity is more often present in T-cell acute lymphoblastic leukaemic cells in adults than in children.

There is a distinct difference in prognosis between childhood versus adult acute lymphoblastic leukaemia (ALL). To define whether multidrug resistance (MDR) genes might contribute to this distinction, the expression and functional activity of P-glycoprotein (P-gp) and MDR associated proteins (MRP) were determined with RT-PCR (MDR-1, MRP1, MRP2, MRP3) and flow cytometry (P-gp and MRP). Patient samples were obtained from 36 children and 35 adults with de novo ALL. Of these patients, 38 showed a T-lineage and 33 showed a B-lineage immunophenotype. In the samples, large variability in P-gp activit

NSUN2-mediated m5C modification drives alternative splicing reprogramming and promotes multidrug resistance in anaplastic thyroid cancer through the NSUN2/SRSF6/UAP1 signaling axis.

Rationale: Anaplastic thyroid carcinoma (ATC) is an extraordinarily aggressive form of thyroid cancer, frequently presenting with locally advanced infiltration or distant metastases at the time of initial diagnosis, thus missing the optimal window for surgical intervention. Consequently, systemic chemotherapy and targeted therapies are vital for improving the prognosis of ATC. However, ATC exhibits significant resistance to conventional treatments, highlighting the need to elucidate the biological mechanisms underlying this drug resistance and identify novel therapeutic targets to overcome it.

Enhanced nuclear enrichment of chemotherapeutics by a biomimetic ZIF-8 nanosystem for multidrug-resistant cancer treatment.

The exclusion of chemo-drugs from their target sites, e.g. the nucleus, is a critical factor contributing to the failure of chemotherapy. Nanoparticle-based technology holds promise for specific delivery of chemo-drugs to nucleus. However, limited lysosomal escape, small size of nuclear pores and incapability of direct regulation of drug efflux-related transporters made great challenges for nanoparticles. Here, a zeolitic imidazolate framework-8 (ZIF-8)-based nanosystem was found able to disrupt lysosomes by the pH-responsiveness of ZIF-8 and mediate the nuclear entry of chemotherapeutics thro

Integrated analysis of global regulators and efflux genes associated with antimicrobial resistance reversal in multidrug resistant Klebsiella pneumoniae.

BACKGROUND: Multidrug-resistant Klebsiella pneumoniae poses a growing clinical challenge due to its ability to evade antibiotic treatment, particularly through the overexpression of efflux systems. Among these, the AcrAB-TolC pump is central to resistance against fluoroquinolones. While the global regulator's MarA, SoxS, and Rob are known modulators of efflux in Enterobacteriaceae, their functional relevance in clinical K. pneumoniae remains insufficiently defined and largely correlation-based. OBJECTIVE: This study was designed to (1) determine the transcriptional association between the glo

Silent Reservoirs: Antibiotic-Resistant Escherichia coli in Autochtonous Portuguese Laying Hens.

Antimicrobial Resistance (AMR) is a critical public health challenge requiring a coordinated One Health approach. Escherichia coli is a key indicator of AMR and fecal contamination, as well as a zoonotic pathogen transmissible from animals to humans, often through contaminated products like meat and eggs. This study assessed the presence of antibiotic-resistant E. coli and associated resistance genes in 248 cloacal/eggshell samples collected from four autochthonous Portuguese laying hen breeds (Preta Lusitânica, Amarela, Branca, and Pedrês Portuguesa) raised under low antibiotic exposure. A to

The use of subtractive proteomics and comparative genomics prioritizes novel therapeutic targets in Salmonella Infantis.

Salmonella enterica serovar Infantis has emerged as a globally relevant non-typhoidal Salmonella (NTS) serovar, due to its increasing prevalence in foodborne outbreaks, level of multidrug resistance (MDR), and persistence in poultry production systems. Despite extensive genomic surveillance, few studies have integrated genome-wide analyses with subtractive proteomics for therapeutic target discovery. Here, N = 144 S. Infantis genomes, from clinical and environmental sources were retrieved from the NCBI Pathogen Detection database and systematically analyzed. Virulence profiling revealed a cons
