Effect of aerobic and anaerobic atmosphere on the detection of micro-organisms from blood cultures.

Blood from patients with suspected bacteraemia was simultaneously cultured in two Vacutainer blood culture bottles containing 45 ml of supplemented peptone broth (Becton Dickinson), five ml of blood in each bottle. The cultures were incubated at 37 degrees C for seven days. One of the bottles was vented and the other unvented. A total of 3958 blood culture sets were processed during the 20-month period of the study. Altogether 438 (11%) of the blood culture sets were positive. Of the 438 isolates, 317 (72%) were detected in both bottles, 61 (14%) in the vented bottle and 60 (14%) in the unvent

A Frog-Derived Cathelicidin Peptide with Dual Antimicrobial and Immunomodulatory Activities Effectively Ameliorates Staphylococcus aureus-Induced Peritonitis in Mice.

As antimicrobial resistance poses an increasing threat to public health, it is urgent to develop new antimicrobial agents. In this paper, we identify a novel 30-residue peptide (Nv-CATH, NCNFLCKVKQRLRSVSSTSHIGMAIPRPRG) from the skin of the frog Nanorana ventripunctata, which belongs to the cathelicidin family. Nv-CATH exhibited broad-spectrum antimicrobial activity against Gram-positive and Gram-negative bacteria. Nv-CATH significantly protected mice from lethal infections caused by Staphylococcus aureus. Furthermore, the peptide suppressed excessive and harmful inflammatory responses by repre…

Organoiodine (III)-mediated synthesis of 3-aryl/heteroaryl-5,7-dimethyl-1,2,4-triazolo[4,3-c]pyrimidines as antibacterial agents.

Synthesis of 12 new 3-aryl/heteroaryl-5,7-dimethyl-1,2,4-triazolo[4,3-c]pyrimidines (3a-l) has been accomplished by the oxidation of pyrimidinylhydrazones (2a-l) of various aryl/heteroaryl aldehydes using 1.1equiv. of iodobenzene diacetate (IBD) in dichloromethane. All the compounds 3a-l tested in vitro for their antibacterial activity against two Gram-positive bacteria namely, Bacillus subtilis, Bacillus stearothermophilus and two Gram-negative bacteria Pseudomonas putida, Escherichia coli. Two compounds, namely 3-(2,4-dichlorophenyl)-5,7-dimethyl-1,2,4-triazolo [4,3-c]pyrimidine (3j) and 3-(…

Recent trends in clinical isolates from paranasal sinusitis.

Trends in the detection of causative pathogens and changes in bacterial counts in patients with sinusitis treated between January 1989 and December 1993 were investigated. In adult patients with chronic sinusitis, Staphylococcus aureus (S. aureus), coagulase negative staphylococci (CNS), Streptococcus pneumoniae (S. pneumoniae), Corynebacterium sp., Haemophilus influenzae (H. influenzae), and Moraxella catarrhalis were often isolated while Pseudomonas aeruginosa (P. aeruginosa) and anaerobic bacteria were detected in 2.4% and 5.3% of patients, respectively. The bacteria isolated from adult pat

Azithromycin and clarithromycin.

Azithromycin and clarithromycin are alternatives to conventional macrolides in the routine treatment of many dermatologic, upper respiratory, and lower respiratory tract infections. In this role as alternative therapy, they are better tolerated, less toxic, and more convenient to take, although at a greater cost to the patient. This dosing convenience is an important consideration for the clinician; as shown by Nelson, patient compliance ranges from 95% with once-daily dosing to 58% with four-times-a-day dosing. Thus, less frequent dosing with both drugs as well as the shorter course of therap

In vitro antimicrobial activity of cefodizime, a third generation cephalosporin.

Cefodizime is one of the new broad-spectrum cephalosporins. It is an aminothiazolyl iminomethoxy cephalosporin which is metabolically stable and has a prolonged serum half life. Cefodizime was primarily active against gram-negative bacilli and at the concentration of 0.5 mg/L, it inhibited 90 per cent of Enterobacteriaceae. P. mirabilis was the most susceptible species tested (MIC90 of 0.02 mg/L). E.coli, K.pneumoniae, Salmonella spp., Shigella spp. and M. morganii were also very sensitive to cefodizime, with the MIC90 of 0.25-0.5 mg/L. Cefodizime, however, was not active against most gram-neg

Enumeration of bacteria in clinically significant blood cultures in neutropenic and non-neutropenic patients using a pour plate method.

A 3-year review of clinically significant positive blood cultures was undertaken to assess any differences in the blood bacterial count between haematological neutropenic and other non-neutropenic patients. The pour-plate method was used. In Gram-positive infections the pour plate contained colonies in 61% of haematological patients and in 41% of others. In Gram-negative infection the figures were 54% and 25% respectively. The mean numbers of bacteria per ml of blood were increased in haematological patients compared with the others for both groups of organisms.

Lipopolysaccharide Evaluation in Peritoneal Dialysis Patients with Peritonitis.

BACKGROUND: Lipopolysaccharide (LPS), also known as endotoxin, is cell wall component of Gram-negative (GN) bacteria, which may contribute to the progression of a local infection to sepsis. Previous studies demonstrate that LBP is detectable in peritoneal effluents of peritoneal dialysis (PD) patients and it is significantly elevated in PD patients with peritonitis caused by both GN and Gram-positive (GP) bacteria. AIM: The aim of this study was to evaluate LPS levels in PD patients; in particular, we investigated different LPS levels in the context of GP and GN peritonitis. MATERIAL AND MET

[Rational use of antibiotics in mucoviscidosis pediatric cases with an account of local microbiological monitoring].

The results of local microbiological monitoring of bronchial secretion in 482 children with mucoviscidosis observed within 2000-2004 in the Republican Centre of Mucoviscidosis are presented. The results provided development of recommendations for rational use of antibiotics in the treatment of infectious processes in pediatric patients with mucoviscidosis. Since the emergence of MRSA in such patients is low, it is recommended to use antistaphylococcal betalactams (oxacillin, cefazolin, amoxycillin/clavulanate) for the treatment of infections due to S. aureus. For the treatment of infections du

Cefdinir: a review of its use in the management of mild-to-moderate bacterial infections.

Cefdinir (Omnicef) is an oral third-generation cephalosporin with good in vitro activity against many pathogens commonly causative in community-acquired infections. The drug provides good coverage against Haemophilus influenzae, Moraxella catarrhalis and penicillin-susceptible Streptococcus pneumoniae, the most common respiratory tract pathogens. Cefdinir is stable to hydrolysis by commonly occurring plasmid-mediated beta-lactamases and retains good activity against beta-lactamase-producing strains of H. influenzae and M. catarrhalis. The drug distributes into various tissues (e.g. sinus and t

Gram Typing Bacteria Panels in Whole Blood Using a Biphasic Duplex-Loop-Mediated Isothermal Amplification Assay.

Timely identification of bacteria in bloodstream infections is critical for guiding appropriate antibiotic treatment. However, current clinical workflows entail blood culture (1-5 days), followed by Gram staining, PCR, and antibiotic susceptibility testing. These steps delay actionable results, often leading clinicians to prescribe broad-spectrum antibiotics without results from the above tests, contributing to the rising threat of antimicrobial resistance. Specifically, rapid information of even presence of Gram-positive and/or Gram-negative bacteria would help clinicians choose a specific an…

Gram differentiation of bacteria by acoustic-enhanced flow cytometry.

Introduction. Recently, flow cytometry has gained the attention of clinical microbiologists for its ability to characterize bacterial species. This article shows how acoustic-enhanced flow cytometry, combined with the fluorescent dye SYTO 9, can differentiate between Gram-positive and Gram-negative bacteria.Gap Statement. SYTO 9 is a cell membrane-permeable dye with a high affinity for nucleic acids in both living and non-living prokaryotic and eukaryotic cells and has been used as a counterstain to discriminate between live and dead cells in combination with other dyes. However, the consisten…

Evaluating the potential of vancomycin-modified magnetic beads as a tool for sample preparation in diagnostic assays.

Vancomycin-functionalized micro- or nanoparticles are frequently used for isolation and enrichment of bacteria from various samples. Theoretically, only Gram-positive organisms should adhere to the functionalized surfaces as vancomycin is an antibiotic targeting a peptidoglycan precursor in the cell wall, which in Gram-negative bacteria is shielded by the outer cell membrane. In the literature, however, it is often reported that Gram-negative bacteria also bind efficiently to the vancomycin-modified particles. The goal of our study was to identify the underlying cause for these different findi…

Antimicrobial Properties of Capsaicin: Available Data and Future Research Perspectives.

Capsaicin is a phytochemical derived from plants of the genus Capsicum and subject of intensive phytochemical research due to its numerous physiological and therapeutical effects, including its important antimicrobial properties. Depending on the concentration and the strain of the bacterium, capsaicin can exert either bacteriostatic or even bactericidal effects against a wide range of both Gram-positive and Gram-negative bacteria, while in certain cases it can reduce their pathogenicity by a variety of mechanisms such as mitigating the release of toxins or inhibiting biofilm formation. Likewi…
