A guinea pig model of Legionnaires' disease was produced by intratracheal inoculation of Legionella pneumophila. The bacterial inoculum given reproducibly resulted in a 95% fatality rate in untreated animals. These animals had lung histopathologic and bacteriologic findings almost identical to those found in human Legionnaires' disease. Administration of cefoxitin and gentamicin, which inhibited the infecting strain in low antibiotic concentration in vitro, was no more effective than saline; this occurred despite adequate serum and lung levels of the drugs. The mode of action of histamine and its analogues on the secretion of amylase from guinea-pig parotid gland was studied in vitro using chopped parotid tissue. Histamine, 4-methylhistamine and 2-(2-pyridyl)ethylamine dose-dependently stimulated the secretion of amylase in the presence of 3-isobutyl-1-methylxanthine. The responses of the guinea-pig ileum myenteric plexus-longitudinal muscle preparation to gamma-aminobutyric acid (GABA) and several analogs were examined in direct and electrically stimulated preparations. GABA, muscimol and 3-aminopropane sulfonic acid (3-APS), but not baclofen, produced a transient, concentration-dependent contraction followed by relaxation. A reproducible population of Legionella pneumophila was obtained in late exponential growth phase and inoculated into the trachea of young animals. Guinea pigs were restrained in a smoking machine and were exposed to the smoke of eight cigarettes a day, five days a week for 12 to 15 weeks. One group of animals inhaled filtered smoke from which most of the tar and nicotine had been removed. Heart weight of the smoking animals was found to be significantly increased in both the filtered and unfiltered groups. Transplacental transmission of cytomegalovirus (CMV) was demonstrated in guinea pigs, but transmission only occurred during acute primary maternal infection.
