Serum Interleukin-40 and Soluble CD40 Ligand as Complementary Biomarkers for Disease Activity in Multiple Sclerosis Patients.

OBJECTIVES: Multiple sclerosis (MS) is a complex autoimmune disease of the central nervous system for which reliable biomarkers of disease activity remain an unmet need. Interleukin-40 (IL-40) and soluble CD40 ligand (sCD40L) have been proposed to play roles in the pathogenesis of autoimmune diseases. This study aimed to evaluate serum levels of IL-40 and sCD40L as biomarkers of disease activity in MS patients, and to compare their diagnostic and monitoring performance between MS patients and healthy controls. METHODS: One hundred twenty MS patients were recruited from the Department of MS at

Association of status of acetylcholinesterase and ACHE gene 3' UTR variants (rs17228602, rs17228616) with drug addiction vulnerability in pakistani population.

Substance addiction is a chronic, relapsing mental disorder Characterized by compulsive drug seeking, and loss of control over drug intake and relapse after prolonged abstinence. Genetics has been shown to contribute towards an individual's vulnerability to addiction. Acetylecholine (ACh), a cholinergic neurotransmitter hydrolyzed by acetylcholinesterase (AChE), is an essential neurotransmitter and neuromodulator in central and peripheral nervous system and has regulatory influence on numerous neuronal functions including addiction. The present study was carried out to investigate the role of

Preliminary effects of progressive muscle relaxation on cigarette craving and withdrawal symptoms in experienced smokers in acute cigarette abstinence: a randomized controlled trial.

BACKGROUND: Cigarette craving usually occurs in conjunction with unpleasant feelings, including stress, as part of a withdrawal syndrome. Progressive muscle relaxation (PMR), a behavioral technique used to reduce stress by concentrating on achieving muscle relaxation, may reduce levels of cigarette craving and other substance-related negative feelings and withdrawal symptoms. METHODS: Demographic and cigarette use data were collected from 32 experienced smokers at the King Chulalongkorn Memorial Hospital, Bangkok, Thailand using the Semi-Structured Assessment for Drug Dependence and Alcoholis

Sympathetic nervous system mediated cardiovascular effects of cocaine are primarily due to a peripheral site of action of the drug.

Currently, augmentation of sympathetic nervous system function produced by cocaine is thought to be due primarily to stimulation of sympathetic centers in the brain (central effect) and to inhibition of catecholamine uptake into postganglionic sympathetic nerve terminals (peripheral effect). In this review of our work, we present the following evidence that cocaine-induced changes in cardiovascular function, particularly those that peak within 1 to 5 min after an i.v. bolus injection of the drug, are due to a peripheral effect of the drug: (1) In both dogs and cats, cocaine potentiates the tac

The psychology of perserverative and stereotyped behaviour.

Many forms of psychopathology in higher animals and humans include the production of maladaptive, repetitive behaviour. Behaviour which is both repetitive and excessive in amount can be described as stereotyped whereas behaviour which represents a restriction of behavioural possibilities without excessive production can be described as perseverative. Both types of repetition can result from pathology in the neural mechanisms which control either the production of motor output or the organisation of behaviour at a higher level. A number of forms of repetitive behaviour can be induced environmen

Gabapentinoids and Risk for Exacerbation of Chronic Obstructive Pulmonary Disease.

Rationale: Data on the effect of gabapentinoids on patients with chronic obstructive pulmonary disease (COPD) are sparse, although the U.S. Food and Drug Administration has issued a safety warning for these medications, particularly in individuals with respiratory risk factors. Objectives: To investigate whether gabapentinoid use is associated with increased COPD exacerbations requiring systemic corticosteroids. Methods: Using a nationwide administrative claims database, we conducted a retrospective cohort study using an active-comparator new-user design. We identified patients with COPD and n

Lack of CCR5 modifies glial phenotypes and population of the nigral dopaminergic neurons, but not MPTP-induced dopaminergic neurodegeneration.

Constitutive expression of C-C chemokine receptor (CCR) 5 has been detected in astrocytes, microglia and neurons, but its physiological roles in the central nervous system are obscure. The bidirectional interactions between neuron and glial cells through CCR5 and its ligands were thought to be crucial for maintaining normal neuronal activities. No study has described function of CCR5 in the dopaminergic neurodegeneration in Parkinson's disease. In order to examine effects of CCR5 on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced dopaminergic neurodegeneration, we employed CCR5 wil

Increased expression of cystine/glutamate antiporter in multiple sclerosis.

BACKGROUND: Glutamate excitotoxicity contributes to oligodendrocyte and tissue damage in multiple sclerosis (MS). Intriguingly, glutamate level in plasma and cerebrospinal fluid of MS patients is elevated, a feature which may be related to the pathophysiology of this disease. In addition to glutamate transporters, levels of extracellular glutamate are controlled by cystine/glutamate antiporter x(c)⁻, an exchanger that provides intracellular cystine for production of glutathione, the major cellular antioxidant. The objective of this study was to analyze the role of the system x(c)⁻ in glutamate

Differential nerve injury-induced expression of MHC class II in the mouse correlates to genetic variability in the type I promoter of C2ta.

Major histocompatibility complex (MHC) class II is of critical importance for the induction of immune responses. Levels of MHC class II in the nervous system are normally low, but expression is up-regulated in many disease conditions. In rat and human, variation in the MHC class II transactivator gene (C2ta) is associated with differential expression of MHC class II and susceptibility to autoimmune disease. Here we have characterized the response to facial nerve transection in 7 inbred mouse strains (C57BL/6J, DBA/2J, 129X1/SvJ, BALB/cJ, SJL/J, CBA/J, and NOD). The results demonstrate differen

Interoceptive autonomic regulation in typical development and autism spectrum disorder: A computational model integrating multiple physiological systems.

BACKGROUND: Interoceptive cardiovascular signals, including heart rate (HR) and blood pressure (BP), arise from coordinated sympathetic (SNS) and parasympathetic (PSNS) regulation and contribute to affective and cognitive processes. Although atypical autonomic nervous system (ANS) modulation has been reported in autism spectrum disorder (ASD), the dynamical structure underlying branch-specific coordination remains insufficiently characterized. OBJECTIVE: To estimate latent ANS regulatory structure in typically developing (TD) and ASD individuals using a computational modeling framework. METH

Cerebellar nuclei output pathways regulate motor and non-motor behaviors through forebrain circuits.

The adult cerebellum is involved in modulating most, if not all, behaviors via its direct or indirect projections throughout the central nervous system. Its involvement in higher order behaviors is most prominent in human, where the lateral cerebellum (hemispheres) has expanded along with areas in the cerebral cortex that regulate language, emotions and cognition. An important aspect of wiring of the brain, is how the circuitry is established during normal development and activity, compared to how it is established when a developmental defect occurs, such as an injury that kills cells or a gen

Prolonged performance-related neuroendocrine activation and perseverative cognition in low- and high-anxious university music students.

Music performances are social-evaluative situations that can elicit marked short-term neuroendocrine activation and anxious thoughts especially in musicians suffering from music performance anxiety (MPA). The temporal patterns of neuroendocrine activity and concert-related worry and rumination (perseverative cognition, PC) days before and after a concert in low- and high-anxious musicians are unknown. The first goal of the present study was to investigate the prolonged effects of a solo music performance and the effects of trait MPA on salivary cortisol (sC), alpha-amylase (sAA), and concert-r

Plastic changes along auditory pathway during salicylate-induced ototoxicity: Hyperactivity and CF shifts.

High dose of salicylate, the active ingredient in aspirin, has long been known to induce transient hearing loss, tinnitus and hyperacusis making it a powerful experimental tool. These salicylate-induced perceptual disturbances are associated with a massive reduction in the neural output of the cochlea. Paradoxically, the diminished neural output of the cochlea is accompanied by a dramatic increase in sound-evoked activity in the auditory cortex (AC) and several other parts of the central nervous system. Exactly where the increase in neural activity begins and builds up along the central audito

Over-excitation of tyrosine hydroxylase neurons in the paraventricular nucleus of the hypothalamus drives a neuroimmune positive feedback loop in rheumatoid arthritis.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent inflammation and pain, leading to a significant decline in patients' quality of life. Recent studies suggest that the central nervous system (CNS) may also contribute to disease progression by modulating autonomic and immune functions. However, the precise mechanisms underlying CNS involvement remain unclear. OBJECTIVE: This study aims to investigate the alterations in the functional state of tyrosine hydroxylase (TH) within the paraventricular nucleus (PVN) in RA and to explore their potential r

[Studies on the effects of constant exercise around the ventilatory threshold using spectral analysis of EMG and ECG-RR interval variability].

The effects constant exercise at three levels around the ventilatory threshold (VT) were investigated by spectral analysis of EMG and RR interval variability of ECG. The subjects were 11 healthy male students (ranging from 20 to 25 years old). Surface EMG from the knee extensor muscular (m. vastus medialis) and the RR interval of the ECG were recorded by bicycle ergometer during the three levels of exercise which were 20% less than the VT level (-20% VT), the VT level and 20% more than the VT level (+20 VT). Power spectral analysis of EMG was performed to obtain the power spectral density (PSD

Canine leishmaniosis and peripheral neuropathy: a lesson from the neurologist.

BACKGROUND: Canine leishmaniosis (CanL), a sand fly-borne zoonotic disease caused by Leishmania infantum, is potentially lethal in dogs. A similar or slightly higher quantity of antigens over antibodies promotes the formation of soluble circulating immune complexes (sCIC), which are deposited in the capillary wall, causing the inflammatory cascade responsible for clinical manifestations. Nervous system involvement during CanL is rarely reported in both humans and dogs, and the exact underlying process involving the peripheral nervous system (PNS) is still debated in both species. METHODS: Two
