Protozoa and digestive tract parameters of the impala.

Intestinal contents were collected from eight impala at three different localities during the winter hunting season (2005-2009), as well as from another 24 animals from a one-year trial at a game farm called Ditholo (2003-2004). Gas production, protozoa counts and several other physiological parameters were measured from both rumen and caecum or colon contents. Only higher ophryoscolecid and Isotrichidae species of protozoa were counted and identified. Ostracodinium gracile was present in all 32 impala. Eudiplodinium maggii was present in 31 animals and Eudiplodinium impalae and Epidinium (eit

Refractory barriers in the sand fly Phlebotomus papatasi (Diptera: Psychodidae) to infection with Leishmania panamensis.

The life cycle of Leishmania panamensis in Phlebotomus papatasi was studied to characterize barriers limiting parasite colonization, differentiation, migration, and attachment in an unnatural sand fly host. The insects were fed a suspension of L. panamensis-infected macrophages and human erythrocytes, and were examined up to 16 days post-infection by light and electron microscopy. Histologic examination of 401 flies showed the peritrophic membrane to be the first important barrier to parasite establishment in the gut lumen. In most flies, parasites were unable to escape from the closed peritro

Computational insights and impact of combinatorial peptidomimetics on immune escape SARS-CoV-2 variants.

BACKGROUND: The SARS-CoV-2 receptor-binding domain (RBD) undergoes frequent mutations, weakening current therapeutics. Despite its importance in viral entry, most inhibitors lack resilience to antigenic drift, emphasizing the need for broad-spectrum agents that maintain binding across variants while preserving host specificity. RESEARCH DESIGN AND METHODS: The authors employed computational mutagenesis to design h-ACE2-derived peptidomimetics incorporating nonstandard amino acids (NSAAs), targeting conserved RBD residues in SARS-CoV-2, Omicron, and JN.1. Approximately 10,000 NSAAs were screen…

Genomics and evolution of Pneumocystis species.

The genus Pneumocystis comprises highly diversified fungal species that cause severe pneumonia in individuals with a deficient immune system … hich in turn is determined by intrinsic features such as their self-fertilizing mating system, high host specificity, long generation times, and transmission mode. Essential key questions concerning their adaptation and speciation remain to be answered. The next cornerstone will consist in the establishment of a long-term culture system and genetic manipulation th …

Erhaia (Gastropoda: Rissooidea): phylogenetic relationships and the question of Paragonimus coevolution in Asia.

The human lung fluke Paragonimus is transmitted by gastropod taxa of two superfamilies: Ceritheoidea and Rissooidea. The question whether or … westermani complex have only been found in ceritheoidean snails. The implication is that there is a host specificity on the superfamily level. However, Asian freshwater species of the Ceritheoidea and Rissooidea usually are not sympatric and often prefer different habitats. It is therefore possible that ecological niche partitioning plays the primary role for Para …

Microevolutionary cophylogeny reflects host-symbiont population dynamics and human mitonuclear interactions.

Cophylogeny, the study of phylogenetic similarity between interacting organisms, provides insights into the specificity and shared evolutionary history of symbiosis. While the ecological drivers of cophylogeny have been investigated at the macroevolutionary scale, the influence of these processes on microevolution remains unclear. This is due, in part, to the fact that the ancestral relations between individuals within a sexually reproducing eukaryotic host species cannot be well represented with a single phylogenetic tree, since genetic distances between individuals change substantially acros

Plasmodium knowlesi tryptophan-rich antigen 40.1 binds human erythrocyte spectrin alpha.

Plasmodium infection induces extensive remodeling of host erythrocytes through parasite-exported proteins that interact with host cell components. Among these, tryptophan-rich antigens (TRAgs) are conserved across Plasmodium species and have been proposed as vaccine candidates, but their molecular functions remain largely unexplored. In this study, we characterized Plasmodium knowlesi tryptophan-rich antigen 40.1 (PkTRAg40.1) to investigate its localization, biochemical properties, host interactions, and immunogenicity. Recombinant PkTRAg40.1 showed no detectable binding to intact human erythr

Selective bacterial proteolysis in periodontal disease: Molecular mechanisms and therapeutic implications, a narrative review.

BACKGROUND/OBJECTIVE: Periodontal diseases are characterized by complex interactions between the immune system, oral bacteria, host tissues, and various external and internal factors. A hallmark of disease progression is the selective degradation of host proteins by bacterial proteases. This review explores the molecular mechanisms of bacterial proteolysis, with a focus on the selectivity toward host proteins essential for oral homeostasis. METHODS: An extensive electronic search was conducted using PubMed, Scopus, and Web of Science, covering the last 20 years. Search terms included "degrada

Eng1 and Exg8 Are the Major β-Glucanases Secreted by the Fungal Pathogen Histoplasma capsulatum.

Fungal cell walls contain β-glucan polysaccharides that stimulate immune responses when recognized by host immune cells. The fungal pathogen Histoplasma capsulatum minimizes detection of β-glucan by host cells through at least two mechanisms: concealment of β-glucans beneath α-glucans and enzymatic removal of any exposed β-glucan polysaccharides by the secreted glucanase Eng1. Histoplasma yeasts also secrete the putative glucanase Exg8, which may serve a similar role as Eng1 in removing exposed β-glucans from the yeast cell surface. Here, we characterize the enzymatic specificity of the Eng1 a

The interaction of donor and host lymphoid cells in the pathogenesis of renal cortical destruction induced by a local graft versus host reaction.

The graft versus host reaction (GVHR), which results from the injection of parental strain spleen cells beneath the kidney capsule of F(1) hybrid rats, is transferable during its developmental phase into F(1) hybrid hosts isogeneic with the primary host, but not into secondary hosts of the parental (donor) strain. Furthermore, the GVHR propagates but rarely in secondary hybrid hosts which are allogeneic with respect to the primary hosts, but which are also genetically tolerant of donor-type cells. These findings indicate that the donor cells not only initiate the GVHR but also maintain it by v

The pathogenesis of influenza virus infections: the contributions of virus and host factors.

Influenza viruses cause acute respiratory inflammation in humans and symptoms such as high fever, body aches, and fatigue. Usually these symptoms improve after several days; however, the 2009 pandemic H1N1 influenza virus [influenza A(H1N1) 2009] is more pathogenic than seasonal influenza viruses and the pathogenicity of highly pathogenic H5N1 viruses is still higher. The 1918 influenza pandemic virus caused severe pneumonia, resulting in an estimated 50 million deaths worldwide. Several virulence factors have been identified in these virus strains, but host factors are also responsible for th

Comparison of historic and novel data reveals higher contemporary diversity of trematode metacercariae in freshwater fish.

Trematode metacercariae are the most abundant and frequently encountered helminths in freshwater fish. Yet, accurate species identification … reported Clinostomidae were absent. Fish of the Leuciscidae hosted the highest trematode diversity. Host specificity of metacercariae was generally low, with most species being euryxenous. At the microhabitat level, eyes harboured the highest number of species, while muscles showed the highest metacercarial density. Notably, we detected species first genetically c …

Reprogramming product selectivity and activity in promiscuous terpene synthases via substrate conformation engineering.

Terpenoids are structurally diverse natural products with broad applications, yet their biosynthesis is often constrained by the low catalytic efficiency and poor product selectivity of terpene synthases (TPSs). Simultaneously enhancing product specificity and catalytic performance remains a major challenge in TPS engineering. Here, we elucidate the regio- and stereoselectivity of a fungal germacrene A synthase, which catalyzes the 1,10-cyclization of farnesyl diphosphate to form a central sesquiterpene intermediate. The side-chain volume of a non-catalytic residue (T169) adjacent to the subst

Whole blood mRNA expression-based targets to discriminate active tuberculosis from latent infection and other pulmonary diseases.

Current diagnostic tests for tuberculosis (TB) are not able to predict reactivation disease progression from latent TB infection (LTBI). The main barrier to predicting reactivation disease is the lack of our understanding of host biomarkers associated with progression from latent infection to active disease. Here, we applied an immune-based gene expression profile by NanoString platform to identify whole blood markers that can distinguish active TB from other lung diseases (OPD), and that could be further evaluated as a reactivation TB predictor. Among 23 candidate genes that differentiated pa

Antigens of Onchocerca volvulus.

Most studies on immunologic responses to Onchocerca volvulus have employed extracts or antigens from related filarial parasites. Consequently, little is known about the nature of O. volvulus antigens. Potential antigen sources include in vitro cultures and physicochemical fractionation of O. volvulus extracts. IgE antibody responses to a wide range of antigens occur in patients with onchocerciasis, but there is little evidence of species specificity in serologic tests. Some potent allergens are released by microfilariae, but host serum proteins appear to contaminate the most reactive fractions
