Human disease agents exist within complex environments that have underappreciated effects on transmission, especially for parasites with multi-host life cycles. We examined the impact of multiple host and parasite species on transmission of the human parasite Schistosoma mansoni in Kenya. We show S. mansoni is impacted by cattle and wild vertebrates because of their role in supporting trematode parasites, the larvae of which have antagonistic interactions with S. mansoni in their shared Biomphalaria vector snails. We discovered the abundant cattle trematode, Calicophoron sukari, fails to devel

Lernaeenicus radiatus, a mesoparasitic pennellid copepod, has long been known in the northwest Atlantic with metamorphosed females infecting the muscle of marine fish. The study herein is the first to identify a definitive first host, black sea bass Centropristis striata, for L. radiatus supporting larval development to adults and sexual reproduction in the gills. This finding suggests a two-host life cycle for L. radiatus, with black sea bass as the first host. Heavy infections in the gill were associated with considerable pathology related to a unique and invasive attachment process that pen

Many studies have shown that coagulation systems play an important role in the defence against pathogens in invertebrates and vertebrates. In vertebrates, particularly in mammals, it has been established that the coagulation system participates in the entrapment of pathogens and activation of the early immune response. However, functional studies investigating the importance of the fish coagulation system in host defence against pathogens are scarce. In the present study, injection of turbot (Scopthalamus maximus) with the pathogenic ciliate Philasterides dicentrarchi led to the formation of m

Among different species of mites, Demodex folliculorum and Demodex brevis are the only 2 that affect the human eye. Because demodicosis is highly age-dependent and can be found in asymptomatic adults, the pathogenicity of these mites has long been debated. In this study, we summarize our research experience including our most recent study regarding Demodex infestation as a potential cause of ocular inflammatory diseases. Specifically, we describe the pathogenesis of demodicosis and then discuss the results of work investigating the associations and relationships between ocular demodicosis and

Demodex are cosmopolitan mites occurring in many species of mammals. Two morphologically distinct Demodex species parasitize on humans: Demodexfolliculorum (Simon, 1842) and Demodex brevis (Akbulatova, 1963). Until recently, these species have been regarded as commensals; now they are considered as parasitic organisms. In people, Demodex spp. can be found mainly in the sebaceous glands of the skin (mainly in the area of the forehead, cheeks, nose, behind ears and neck), in hearing aids, and on the scalp, covered with hair. Infection with demodex happens through direct contact with infected bed

Experimental infections of Egyptian Radix natalensis and French Galba truncatula with miracidia of Fasciola gigantica of sheep were carried out to determine the larval productivity of this parasite. Rediae and cercariae were thus counted in snails dissected at regular intervals from day 21 to day 49 postexposure (p.e.) at 24 degrees C, while cercarial shedding was studied in other two groups of snails after day 30 p.e. At day 49, the total number of free rediae and that of cercariae-containing rediae in R. natalensis (shell height, 10.0 mm) were 71.5 and 44, respectively, whereas mean values i

OBJECTIVES: To determine the natural crustacean host, the Paragonimus species, and to investigate the potential host-parasite relationship between Manipur Paragonimus and some of the laboratory animals.
METHODS: The laboratory animals such as puppies, albino rats and Swiss mice were infected orally with metacercariae isolated from the fresh water crabs, Potamiscus manipurensis. The fecal specimens of the experimentally infected animals were examined microscopically for Paragonimus eggs at regular intervals. The animals were autopsied on days 35 to approximately 328 after infection and the iso

Sea lice are ectoparasites of salmonids that have been associated with the recent decline in sea trout numbers in north-west Europe. Observed patterns of louse load distribution between sea trout in the seas surrounding the UK, Ireland and Norway and a simple model have been used to analyse the epidemiology of lice. Loads are aggregated and deviate strongly from the Poisson distribution, although less than is observed with many other parasites. The louse numbers on fish from offshore sites are slightly less variable than for fish from coastal sites with comparable mean loads. Analysis of louse

Pig faeces in which Ascaris suum eggs had been embryonating for 57 days were placed in buckets of soil containing either 30 or no earth-worms (Aporrectodea longa). When present, earthworms consumed the faeces and transported the eggs down into the soil, without inflicting any visible damage on the eggs. In later experiments 10 earthworms from the above experiment were fed to each of ten pigs, and another 40 earthworms were dissected. None of the 10 pigs became infected with A. suum through consumption of earthworms, and none of the dissected earthworms were found to contain A. suum larvae. Thi

Two groups of ten hamsters received a primary challenge of five Opisthorchis viverrini metacercariae intragastrically (Groups I and III) and a third group was left untreated (Group II). 90 days after the primary challenge Groups I and II were given a further 50 metacercariae. Animals were killed 35 days after the secondary challenge and worm burdens and faecal egg counts evaluated. Compared with Group II animals, Group I showed a 24.88% reduction in mean liver worm burden, a difference which was significant at a level of P less than 0.01 Differences for faecal egg output/worm between Groups I

Intracellular pathogens have complex metabolic interactions with their host cells to ensure a steady supply of energy and anabolic building blocks for rapid growth. Here we use the obligate intracellular parasite Toxoplasma gondii to probe this interaction for isoprenoids, abundant lipidic compounds essential to many cellular processes including signaling, trafficking, energy metabolism, and protein translation. Synthesis of precursors for isoprenoids in Apicomplexa occurs in the apicoplast and is essential. To synthesize longer isoprenoids from these precursors, T. gondii expresses a bifuncti

The parasite Cryptosporidium modulates the host cell cytoskeleton and microvilli during invasion and development. Identification of effector molecules and their targets is essential for understanding parasite-host interactions and pathogenesis. Here, we examined seven mucin glycoproteins (MUC1-MUC7) located on chromosome 2 of Cryptosporidium parvum using gene tagging. These small proteins are expressed in different secretory organelles including dense granules (DG), small granules (SG), and the rhoptry (ROP), and all but one are exported to the cytoplasm or microvilli of infected cells. Mutati

Toxoplasma gondii is an intracellular parasite that can activate the NLRP1 inflammasome leading to macrophage pyroptosis in Lewis rats, but the underlying mechanism is not well understood. In this study, we performed a genome-wide CRISPR screen and identified the dense granule proteins GRA35, GRA42, and GRA43 as the Toxoplasma effectors mediating cell death in Lewis rat macrophages. GRA35 localizes on the parasitophorous vacuole membrane, where it interacts with the host E3 ubiquitin ligase ITCH. Inhibition of proteasome activity or ITCH knockout prevented pyroptosis in Toxoplasma-infected Lew

In many host-parasite systems, overdispersion in the distribution of macroparasites leads to parasite aggregation in the host population. This overdispersed distribution is often characterized by the negative binomial or by the power law. The aggregation is shown by a clustering of parasites in certain hosts, while other hosts have few or none. Here, I present a theory behind the overdispersion in complex spatiotemporal systems as well as a computational analysis for tracking the behavior of transmissible diseases with this kind of dynamics. I present a framework where heterogeneity and comple

Toxoplasma gondii is a ubiquitous eukaryotic pathogen responsible for toxoplasmosis in humans and animals. This parasite is an obligate intracellular pathogen and actively invades susceptible host cells, a process which is mediated by specific receptor-ligand interactions. Here, we have identified an unnatural 2,4-disulfated d-glucuronic acid (Di-S-GlcA), a hexuronic acid composed of heparin/heparan sulfate, as a potential carbohydrate ligand that can selectively bind to T. gondii parasites. More importantly, the gelatin conjugated Di-S-GlcA multivalent probe displayed strong inhibition of par

The ability of the malaria parasite Plasmodium falciparum to invade and replicate asexually within human red blood cells (RBCs) is central to its pathogenicity, accounting for hundreds of thousands of deaths each year. RBC invasion is a multi-step process involving several host-parasite interactions, yet the host factors acting during invasion remain underexplored, largely due to the intractability of mature enucleated RBCs. The transmembrane protein CD44 was identified as a host factor for P. falciparum invasion through a forward genetic screen using genetically modified RBCs derived from pri…

Parasitic infections are responsible for significant morbidity and mortality throughout the world. Management strategies rely primarily on antiparasitic drugs that have side effects and risk of drug resistance. Therefore, novel strategies are needed for treatment of parasitic infections. Host-directed therapy (HDT) is a viable alternative, which targets host pathways responsible for parasite invasion/survival/pathogenicity. Recent innovative combinations of genomics, proteomics and computational biology approaches have led to discovery of several host pathways that could be promising targets f

Hyperparasitism, in which a parasite exploits another parasite, represents a logical extension of parasite diversity and complexity within ecological networks. While common and ecologically significant in microbial systems, hyperparasitism among metazoan parasites appears comparatively rare. This discrepancy is intriguing given the high diversity and frequent co-occurrence of parasites within hosts. Here, we compile and review published cases of hyperparasitism involving metazoan parasites to assess their taxonomic distribution and ecological characteristics. We identify a wide range of hyperp
