Rewiring viral immunity and autoimmunity through neuroimmune circuits.

Neuroimmune circuits are emerging as a critical layer of host defense. Recent studies show that sensory neurons shape interferon responses, myeloid dynamics, and T cell priming during viral infection, while viruses reprogram these circuits to drive immune pathology. These findings define a dynamic neuro-virus-immune axis with important therapeutic implications.

Candida albicans gastrointestinal colonization resistance: a host-microbiome balancing act.

While Candida albicans is a common, commensal yeast colonizing 50%-60% of humans, it has the potential to expand in the gastrointestinal tract and enter the blood stream resulting in invasive candidiasis. Invasive candidiasis carries a mortality approaching 50%, especially in the most vulnerable, immunocompromised population. Antibacterial use causes an increase in C. albicans gastrointestinal colonization, indicating that the colonic microbiota plays a major role in preventing an uncontrolled expansion, a phenomenon known as colonization resistance. Antibacterials, medications, diet, and co-m

Novel Strategies to Profile SARS-CoV-2 and Human Lung Proteome: Inflammatory Pathways in the Spotlight.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of COVID-19, has caused widespread morbidity and mortality worldwide. SARS-CoV-2 infection triggers innate and adaptive immune responses, but excessive cytokine release can drive hyperinflammation, acute respiratory distress syndrome and poor clinical outcomes. Although serological and molecular assays, such as ELISA and RT-qPCR, remain central to COVID-19 diagnostics, they have limited capacity to reveal host-pathogen interactions at the tissue level. Therefore, profiling the human lung proteome offers a powerfu…

Functional divergence and regulatory network of the type VI secretion system in Vibrio parahaemolyticus.

Vibrio parahaemolyticus is a significant marine pathogen causing gastroenteritis and wound infections in humans. Its pathogenicity is mediated by key virulence factors, including the type VI secretion system (T6SS). This review comprehensively synthesizes current knowledge on the functional divergence and regulatory networks of T6SSs in V. parahaemolyticus, with emphasis on T6SS1 and T6SS2. T6SS1, enriched in clinical isolates, is activated under high-salt and warm conditions and primarily facilitates antibacterial competition and adhesion to human epithelial cells in strains such as RIMD22106

Cryptococcus neoformans and Microglia In Vitro Interaction as a Tool to Study Central Nervous System Infection.

This chapter presents in vitro protocols for studying interactions between Cryptococcus neoformans and microglial cells. These phagocytes are a critical component of the central nervous system's immune response to C. neoformans, the most common fungal agent of meningoencephalitis worldwide. Given its severity and high incidence in immunocompromised patients, understanding the interaction of C. neoformans with central nervous system (CNS) cells could help advance treatment strategies. These protocols leverage microscopy and molecular biology techniques to elucidate the mechanisms of C. neoforma

Oncogenic viruses rewire the epigenome in human cancer.

Viruses contribute to approximately 15-20% of global cancer cases, yet the full spectrum of their oncogenic mechanisms continues to be uncovered. Beyond the classical roles of genome integration, chronic inflammation, and immune evasion, mounting evidence reveals that oncogenic viruses-including the human papillomavirus (HPV), Epstein-Barr virus (EBV), hepatitis B virus (HBV), hepatitis C virus (HCV), and Human T-cell leukemia virus type 1 (HTLV-1)-profoundly reshape the host epigenome to establish persistent infection and promote tumorigenesis. These viruses orchestrate widespread and durable

Sample Preparation for Global Metabolic Profiling of Vaccinia Virus-Infected Primary Human Foreskin Fibroblasts.

Vaccinia virus (VACV), the prototype member of the Poxviridae family, has played a crucial role in medicine as a key component in the development of smallpox vaccines, contributing to the eradication of this deadly disease. Beyond its historical significance, VACV continues to be pivotal in researching metabolic alterations induced by viral infections. Studies have revealed that VACV can impact pathways such as glycolysis, the tricarboxylic acid (TCA) cycle, and lipid metabolism in host cells, offering valuable insights into host-virus interactions and broader cellular metabolism. Dengue virus-host interactions: Structural and mechanistic insights for future therapeutic strategies.

Dengue pathogen, transmitted by mosquitoes, poses a growing threat as it is capable of inflicting severe illness in humans. Around 40% of the global population is currently affected by the virus, resulting in thousands of fatalities each year. The genetic blueprint of the virus comprises 10 proteins. Three proteins serve as structural components: the capsid (C), the precursor of the membrane protein (PrM/M), and the envelope protein (E). The other proteins serve as non-structural (NS) proteins, consisting of NS1, NS2A, NS2B, NS3, NS4A, NS4B, and NS5. The virus relies on these NS proteins to ex

Insight into the Life Cycle of Enterovirus-A71.

Human enterovirus 71 (EV-A71), a member of the Picornaviridae family, is predominantly associated with hand, foot, and mouth disease in infants and young children. Additionally, EV-A71 can cause severe neurological complications, including aseptic meningitis, brainstem encephalitis, and fatalities. The molecular mechanisms underlying these symptoms are complex and involve the viral tissue tropism, evasion from the host immune responses, induction of the programmed cell death, and cytokine storms. This review article delves into the EV-A71 life cycle, with a particular emphasis on recent advanc

Functional and mechanistic insights into the stealth protein full-length CpsY is conducive to understanding immune evasion mechanisms by Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb) is a crucial and destructive intracellular pathogen responsible for causing tuberculosis (TB), a disease of substantial morbidity and mortality. Mtb capsular polysaccharides can misdirect the host's immune response pathways, resulting in additional challenges in TB treatment. These capsule polysaccharides are biosynthesized by a series of stealth proteins including CpsY. Our prior investigations elucidated the structural and functional information of the central domain (aa 201-520) of CpsY within Mtb. However, within the host milieu, it is the full-length itera

Deciphering macrophage dynamics and immune evasion strategies by the intracellular bacteria.

Macrophages are the constituents of the innate immune system that play an important role in the control of infections, phagocytosis, tissue surveillance, and homeostasis. Macrophages display multiple strategies to cope with the invading microbes and prevent the spread of infections. On the other hand, many intracellular pathogens have evolved strategies to circumvent the macrophage-mediated immune responses and even thrive inside macrophages for their survival and replication. The intracellular bacteria possess a specific secretory system that secretes molecules that help regulate the host's i

Transient receptor potential channels in viral infectious diseases: Biological characteristics and regulatory mechanisms.

BACKGROUND: Viral infectious diseases have long posed a challenge to humanity. In recent decades, transient receptor potential (TRP) channels have emerged as newly investigated cation channels. Increasing evidence suggests that TRP channel-mediated Ca2+ homeostasis disruptions, along with associated pathological changes, are critical factors in the onset and progression of viral infectious diseases. However, the precise roles and mechanisms of TRP channels in these diseases remain to be systematically elucidated. AIM OF REVIEW: The aim of this review is to systematically summarize recent adva

Candida albicans natural diversity: a resource to dissect fungal commensalism and pathogenesis.

Candida albicans is a ubiquitous fungus of humans. It is not only a component of the oral and intestinal microbiota of most healthy adults but also a major cause of mucosal disorders and life-threatening disseminated infections. Until recently, research on the biology and pathogenesis of the fungus was largely based on a single clinical isolate. We review investigations that have started to dissect a diverse set of C. albicans strains. Using different approaches to leverage the species' phenotypic and/or genetic diversity, these studies illuminate the wide range of interactions between fungus

In silico agent-based modeling approach to characterize multiple in vitro tuberculosis infection models.

In vitro models of Mycobacterium tuberculosis (Mtb) infection are a valuable tool for examining host-pathogen interactions and screening drugs. With the development of more complex in vitro models, there is a need for tools to help analyze and integrate data from these models. To this end, we introduce an agent-based model (ABM) representation of the interactions between immune cells and bacteria in an in vitro setting. This in silico model was used to simulate both traditional and spheroid cell culture models by changing the movement rules and initial spatial layout of the cells in accordance…

Characterization of Exosomes Released from Mycobacterium abscessus-Infected Macrophages.

Extracellular vesicles (EVs), such as exosomes, play a critical role in cell-to-cell communication and regulating cellular processes in recipient cells. Non-tuberculous mycobacteria (NTM), such as Mycobacterium abscessus, are a group of environmental bacteria that can cause severe lung infections in populations with pre-existing lung conditions, such as cystic fibrosis (CF) and chronic obstructive pulmonary disease (COPD). There is limited knowledge of the engagement of EVs in the host-pathogen interactions in the context of NTM infections. In this study, we found that M. abscessus infection i…

Flagellin-modulated inflammasome pathways characterize the human alveolar macrophage response to Burkholderia pseudomallei, a lung-tropic pathogen.

Melioidosis is an emerging tropical infection caused by inhalation, inoculation, or ingestion of the flagellated, facultatively intracellular pathogen Burkholderia pseudomallei. The melioidosis case fatality rate is often high, and pneumonia, the most common presentation, doubles the risk of death. The alveolar macrophage is a sentinel pulmonary host defense cell, but the human alveolar macrophage in B. pseudomallei infection has never been studied. The objective of this study was to investigate the host-pathogen interaction of B. pseudomallei infection with the human alveolar macrophage and t

Immunomodulation by Enteric Viruses.

Enteric viruses display intricate adaptations to the host mucosal immune system to successfully reproduce in the gastrointestinal tract and cause maladies ranging from gastroenteritis to life-threatening disease upon extraintestinal dissemination. However, many viral infections are asymptomatic, and their presence in the gut is associated with an altered immune landscape that can be beneficial or adverse in certain contexts. Genetic variation in the host and environmental factors including the bacterial microbiota influence how the immune system responds to infections in a remarkably viral str

Native-PAGE analysis of protein aggregation upon viral infection in mouse macrophages.

Upon viral infection, several proteins in the innate signaling pathway form aggregates, which in turn promote the activation of innate antiviral immune response. In this protocol, we use herpes simplex virus type 1 (HSV-1) to infect mouse peritoneal macrophages, and show how to detect the aggregation of TBK1 upon viral infection. The protocol is adaptable for other proteins and other viruses. For complete details on the use and execution of this profile, please refer to Yan et al. (2021).

PML Alternative Splice Products Differentially Regulate HAdV Productive Infection.

Promyelocytic leukemia nuclear bodies (PML-NBs) were considered to maintain antiviral capacity, as these spherical complexes are antagonized by viruses. Actual work provides evidence, that PML-NB-associated factors might also be beneficial for distinct viral processes indicating why genomes and replication centers of nuclear replicating viruses are often found juxtaposed to PML-NBs. Several early HAdV proteins target PML-NBs, such as E4orf3 that promotes redistribution into track-like structures. PML-associated dependency factors that enhance viral gene expression, such as Sp100A remain in the
