The pathogenesis of the Treacher Collins syndrome (mandibulofacial dysostosis).

Numerous synonyms have been used to describe syndromes affecting structures derived from the first and second branchial arches. These conditions are most conveniently grouped into the asymmetrical anomalies of hemifacial microsomia and the symmetrical syndrome of mandibulofacial dysostosis. By examination of animal models of these conditions it can be demonstrated that the pathogenesis is distinct but different for each group. The characteristic facies of mandibulofacial dysostosis suggests a mechanism of malformation which operates early in embryogenesis, acting uniformly on parts which are d

In vitro culture of Echinococcus multilocularis and Echinococcus vogeli metacestodes: studies on the host-parasite interface.

The larval stage of Echinococcus multilocularis causes alveolar echinococcosis (AE) in various mammalians including humans, while Echinococcus vogeli larvae cause a related disease which is also occasionally found in man. Traditionally, Echinococcus metacestodes have been maintained in the laboratory by serial transplantation passages into susceptible animals such as mice or gerbils, enabling the parasite to proliferate asexually. These experimental animal models have been used extensively to investigate host-parasite interactions and to study immunological events occurring at the host-parasit

Sociability as a personality trait in animals: methods, causes and consequences.

Within animal populations there is variation among individuals in their tendency to be social, where more sociable individuals associate more with other individuals. Consistent inter-individual variation in 'sociability' is considered one of the major axes of personality variation in animals along with aggressiveness, activity, exploration and boldness. Not only is variation in sociability important in terms of animal personalities, but it holds particular significance for, and can be informed by, two other topics of major interest: social networks and collective behaviour. Further, knowledge

Finding a suitable coat: The ecology of the invasive deer ked (Lipoptena cervi (Linnaeus, 1758); Diptera: Hippoboscidae), an ectoparasite of large mammals in the Czech Republic.

Some parasitic diseases of wild animals are transmissible to humans and cause health problems. A widespread ectoparasite of vertebrates is the deer ked, Lipoptena cervi ((L.); Diptera: Hippoboscidae). We studied the interaction of this parasite with habitat-related, host-related, and temporal factors in the Czech Republic. We analysed observations of its abundance in hunted wild animals in the context of their environment. Our results confirmed that cervids are a preferred host species. Most of the other relationships between ked abundance and the studied factors were probably due to the olfac

Urinary Tract Infection in a Small Animal Model: Transurethral Catheterization of Male and Female Mice.

Urinary tract infections (UTI) are extremely common worldwide, incurring significant morbidity and healthcare-associated expenses. Small animal models, which accurately reflect disease establishment and progression, permit dissection of host-pathogen interactions and generation of immunity to infection. In mice, intravesical instillation of uropathogenic E. coli, the causative agent in more than 85% of community acquired UTI, recapitulates many of the stages of infection observed in humans. Until recently, however, UTI could only be modeled in female animals. This limitation has hindered the s

Rat as an animal model for Peyronie's disease research: a review of current methods and the peer-reviewed literature.

While the exact mechanism of Peyronie's disease (PD) remains an enigma, the pathophysiology of PD is considered to be multifactorial, with interactions of genetic predisposition, trauma, tissue inflammation and aberrant wound healing. A non-systematic review of the existing English language literature pertaining to the use of rodent models in the evaluation of PD was performed using the Medline database. Multiple free-text searches were performed on key words: animal models of PD, transforming growth factor β1 (TGF β1), tunical and/or corporal fibrosis, subtunical injection and penile myofibro

Inappropriate use of albino animals as models in research.

Sensory-neural, biochemical-metabolic, and physiological anomalies occur in albino mammals. There are ontogenic and biochemical parallels between the senses, peripheral nervous system, endocrine glands, metabolism, and melanin pigmentation. All albino mammals examined have abnormal optic systems. Many drugs cannot be adequately evaluated in an albino model because of melanin's ability to bind and interact with some chemicals. There is evidence that a general reduction in melanin pigment is correlated with a paucity of amino acids necessary for normal chemical function of the brain. There is a

Myocardial B cells have specific gene expression and predicted interactions in dilated cardiomyopathy and arrhythmogenic right ventricular cardiomyopathy.

INTRODUCTION: Growing evidence from animal models indicates that the myocardium hosts a population of B cells that play a role in the development of cardiomyopathy. However, there is minimal data on human myocardial B cells in the context of cardiomyopathy. METHODS: We integrated single-cell and single-nuclei datasets from 45 healthy human hearts, 70 hearts with dilated cardiomyopathy (DCM), and 8 hearts with arrhythmogenic right ventricular cardiomyopathy (ARVC). Interactions between B cells and other cell types were investigated using the CellChat Package. Differential gene expression analy

An animal model recapitulates human hepatic diseases associated with GATA6 mutations.

Heterozygotic GATA6 mutations are responsible for various congenital diseases in the heart, pancreas, liver, and other organs in humans. However, there is lack of an animal that can comprehensively model these diseases since GATA6 is essential for early embryogenesis. Here, we report the establishment of a gata6 knockout zebrafish which recapitulates most of the symptoms in patients with GATA6 mutations, including cardiac outflow tract defects, pancreatic hypoplasia/agenesis, gallbladder agenesis, and various liver diseases. Particularly in the liver, the zebrafish gata6 model exhibits the pau

Five decades of tick-man interaction in Denmark--an analysis.

The tick Ixodes ricinus (L.) is a known vector of several zoonotic diseases such as Tick Borne Encephalitis (TBE), Lyme borreliosis and ehrlichiosis. The interaction between humans and ticks are at the very core of our understanding of the epidemiology of tick borne zoonoses, but only few experimental studies have been performed. Hence our understanding of the epidemiology of tick-borne disease has to rest on the assumption that the potential tick activity, which is reflected in tick samples from the vegetation or samples from animals, corresponds with tick-human interaction. Observations whic

Micro- and nanoplastics influences in Parkinson's disease: lessons from human stem cell models.

Neuroinflammatory contributions play a critical role in Parkinson's disease onset and progression. Key drivers of neuroinflammation include glial cell reactivity, cytokine signaling, protein aggregation, and mitochondrial dysfunction. Although animal models have been extensively used to investigate the mechanisms, their translational relevance is limited because neuroinflammation in humans is typically chronic, heterogeneous, and sustained over years, whereas in rodents is often acute, transient, and resolves within days to weeks. This paper highlights the utility of human stem cell-derived mo

Molecular pathogenesis of necrotizing fasciitis.

Necrotizing fasciitis, also known as the flesh-eating disease, is a severe invasive infection associated with very high rates of human morbidity and mortality. It is most commonly caused by group A Streptococcus(GAS), a versatile human pathogen that causes diseases ranging in severity from uncomplicated pharyngitis (or strep throat) to life-threatening infections such as necrotizing fasciitis. Herein, we review recent discoveries bearing on the molecular pathogenesis of GAS necrotizing fasciitis. Importantly, the integration of new technologies and the development of human-relevant animal mode

Leishmania major infection in humanized mice induces systemic infection and provokes a nonprotective human immune response.

BACKGROUND: Leishmania (L.) species are the causative agent of leishmaniasis. Due to the lack of efficient vaccine candidates, drug therapies are the only option to deal with cutaneous leishmaniasis. Unfortunately, chemotherapeutic interventions show high toxicity in addition to an increased risk of dissemination of drug-resistant parasites. An appropriate laboratory animal based model is still missing which allows testing of new drug strategies in the context of human immune cells in vivo. METHODOLOGY/PRINCIPAL FINDINGS: Humanized mice were infected subcutaneously with stationary phase proma

Traumatic injuries detected at slaughter in cattle: impact of production system and season on animal welfare and meat condemnation in Sweden.

The purpose of ante- and post-mortem inspections at slaughterhouses is to ensure that meat and other relevant food products of animal origin are safe for human consumption. However, these inspections can also be useful for detecting animal health and welfare issues. In cattle, traumatic injuries from on-farm incidents, transport or handling at the slaughterhouse are indications of both reduced animal welfare and increased risk of food waste, ultimately resulting in economic losses for both farmers and slaughterhouses. This observational study aimed to investigate the prevalence and seasonal va

Immune cells in uteroplacental tissues throughout pregnancy: a brief review.

In a brief introduction, this review states why the presence of immune cells at the interface poses problems for an immunologist (Medawar paradigm). Different types of placentation are then discussed, and the various interactions with leukocytes, the extreme being with the equids where a certain degree of 'attack' is often seen. The limits of animal models when dealing with the human situation are emphasized. It is then stated why the various phases of pregnancy are different, and an analysis made of the cellular movements at the implantation, peri-implantation, immediate post-implantation and

The influence of crude cobra venom on electrolyte levels and Na+, K+ ATPase activity in erythrocytes liver and heart cells.

The present study focuses on the in vitro interaction between snake venom and electrolytes and Na+, K+-stimulated ATPase system in number of tissues from humans and animals. The venom produced a change in the resting cellular sodium and potassium levels in all the tissues tested. When compared, only human cancerous tissue showed apparent fall in sodium and a rise in potassium, while no significant change occurred in normal. The effect of snake venom (25 microg/ml) on sodium pump activity, a transport enzyme, was assessed in normal tissues. Human erythrocyte membrane pump was significantly inhi

Modeling Alzheimer's disease in mouse without mutant protein overexpression: cooperative and independent effects of Aβ and tau.

BACKGROUND: Alzheimer's disease (AD), the most common cause of dementia in the elderly, has two pathological hallmarks: Aβ plaques and aggregation of hyperphosphorylated tau (p-tau). Aβ is a cleavage product of Amyloid Precursor Protein (APP). Presenilin 1 (PS1) and presenilin 2 (PS2) are the catalytic subunit of γ-secretase, which cleaves APP and mediates Aβ production. Genetic mutations in APP, PSEN1 or PSEN2 can lead to early onset of familial AD (FAD). Although mutations in the tau encoding gene MAPT leads to a subtype of frontotemporal dementia and these mutations have been used to model

Interaction of Bordetella pertussis with human respiratory mucosa in vitro.

The human respiratory tract pathogen Bordetella pertussis is the major cause of whooping cough in infants and young children, and also causes chronic cough in adults. B. pertussis infection damages ciliated epithelium in the respiratory tract. However, the interaction of the bacterium with the respiratory mucosa is poorly understood, and previous studies have either utilized animal tissue which may not be appropriate, or isolated cell systems which lack the complexity of the respiratory mucosa. We have studied the interaction of B. pertussis strain BP536 with human nasal turbinate tissue in an

Generating Free-floating Normal Human Epithelial-Fibroblast Spheroid Co-Cultures.

Three-dimensional (3D) culture systems have become increasingly popular due to their ability to reproduce natural cell properties and architectures, thus mimicking tissue-like structures in vitro. Among these models, the culture of spherical cell aggregates (so-called spheroids) embedded in a semi-solid extracellular matrix (ECM) represents an advanced near-in vivo cell culture model, as it allows for different functional cell states as a result of cell-cell and cell-ECM interactions, as well as oxygen and nutrient gradients. Because spheroids are technically less demanding and relatively inex
