This systematic review examines the genetic and epigenetic factors associated with resting-state functional connectivity (RSFC) in healthy human adult brains across the lifespan, with a focus on genes associated with Alzheimer's disease (AD). There were 58 studies included. The key findings are: (i) genetic factors have a low to moderate contribution; (ii) the apolipoprotein E ε2/3/4 polymorphism was the most studied genetic variant, with the APOE-ε4 allele most consistently associated with deficits of the default mode network, but there were insufficient studies to determine the relationships

Alternating hemiplegia of childhood (AHC) is a rare neurodevelopmental disorder characterized by recurrent paroxysmal hemiplegic attacks that affect one or the other side of the body. Up to 74% of patients with AHC have a pathologic variant in the ATP1A3 gene. After the introduction of next-generation sequencing, intermediate cases and atypical cases have expanded the clinical spectrum of ATP1A3-related disorders. Herein, we report the first case of AHC in Korea. A 33-year-old man visited our hospital with recurrent hemiplegic and dystonic episode after his first birthday. He was completely no

BACKGROUND: Brain magnetic resonance imaging (MRI) and cognitive tests can identify heritable endophenotypes associated with an increased risk of developing stroke, dementia and Alzheimer's disease (AD). We conducted a genome-wide association (GWA) and linkage analysis exploring the genetic basis of these endophenotypes in a community-based sample.
METHODS: A total of 705 stroke- and dementia-free Framingham participants (age 62 +9 yrs, 50% male) who underwent volumetric brain MRI and cognitive testing (1999-2002) were genotyped. We used linear models adjusting for first degree relationships

BACKGROUND: Fluorescent reporter systems are useful for studying probiotic colonization and host-microbe interactions. However, their use in lactic acid bacteria is still limited by relatively weak fluorescence signals, insufficient expression stability, and limited resolution during in vivo imaging. In particular, efficient strain-specific tracking systems remain scarce.
METHODS: Here, we developed a red fluorescent reporter system for Lactobacillus casei T1 (L.c T1). Lactate dehydrogenase (LDH) promoters identified from the L.c T1 genome were compared with the constitutive P32 promoter to d

The facial anomalies associated with fetal alcohol syndrome (FAS), some of which are also present in individuals with less severe forms of the broader category of fetal alcohol spectrum disorders (FASD), are typically identified with the aid of linear distance measurements taken between facial landmarks. Digital facial imaging methods are increasingly being used in syndrome delineation. Distance measurements derived from stereo-photogrammetry and facial surface imaging have been used to study the FAS facial anomalies. Geometric morphometric methods capture the spatial arrangement between landm

Breast cancer is one of the leading causes of cancer-related death of women globally due to its genomic diversity and rapid metastasis. Because of their invasiveness, limited resolution, and patient variability, traditional diagnostic procedures like biopsies and mammography underscore the need for non-invasive, real-time imaging methods. Advancements in the near-infrared (NIR) spectral range (650-1700 nm) have greatly improved early detection, image-guided surgery, and targeted treatment for breast cancer. This is particularly true for fluorescent probes that can be activated by tumor-specifi

PURPOSE: Fragile X syndrome (FXS) is a common cause of intellectual disability and autism. FXS presents with abnormal facial features, which in pediatric patients are subtler than what is seen in adults. The three-dimensional (3D) facial images, which contain more stereoscopic and subtle information than two-dimensional (2D) photographs, are increasingly being used to classify genetic syndromes. Here, we used 3D facial images to describe facial features and construct a classification model, especially in male patients with FXS.
METHODS: We registered the 3D facial images of 40 Chinese boys wi

RATIONALE: Joubert syndrome (JS) is a rare genetic disorder that presents with various neurological symptoms, primarily involving central nervous system dysfunction. Considering the etiology of JS, peripheral nervous system abnormalities cannot be excluded; however, cases of JS accompanied by peripheral nervous system abnormalities have not yet been reported. Distinct radiological findings on brain magnetic resonance imaging were considered essential for the diagnosis of JS. However, recently, cases of JS with normal or nearly normal brain morphology have been reported. To date, there is no co

The human hippocampus can be subdivided into subfields with unique functional properties and differential vulnerability to disease or neuropsychiatric conditions. Identifying genes that confer susceptibility to such processes is an important goal in developing treatments. Recent advances in automatic subfield segmentation from magnetic resonance images make it possible to use these measures as phenotypes in large-scale genome-wide association studies. Such analyses are likely to rely largely on standard resolution (~1 mm isotropic) T1 -weighted images acquired on 3.0T scanners. Determining whe

BACKGROUND: Molecular characteristics of retinoblastoma cannot be assessed before treatment because tumor biopsy is contraindicated. Photoreceptorness reflects photoreceptor-related gene expression and tumor differentiation. Non-invasive imaging biomarkers that capture this biology are therefore needed.
OBJECTIVE: To evaluate whether quantitative radiomics derived from pretreatment magnetic resonance imaging can predict loss of photoreceptorness in retinoblastoma and validate this approach in an independent cohort.
MATERIALS AND METHODS: In this retrospective multicenter study, patients with

BACKGROUND: Informative biomarkers play a vital role in guiding clinical decisions regarding management of cancers. We have previously demonstrated the potential of a deep convolutional neural network (CNN) for predicting cancer driver gene mutations from expert-curated histopathologic images in papillary thyroid carcinomas (PTCs). Recognizing the importance of whole slide image (WSI) analysis for clinical application, we aimed to develop an automated image preprocessing workflow that uses WSI inputs to categorize PTCs based on driver mutations.
METHODS: Histopathology slides from The Cancer

BACKGROUND: Lung neuroendocrine tumours (NETs, also known as carcinoids) are rapidly rising in incidence worldwide but have unknown aetiology and limited therapeutic options beyond surgery. The current WHO classification, based on mitotic count and presence or absence of necrosis, divides lung NETs into grade-1 typical, and grade-2 atypical tumours. This dichotomous classification however does not account for recently described molecular entities nor is it sufficient for clinical management.
METHODS: Here we conducted integrative multi-omic analyses on over 300 lung NETs including whole-genom

UNLABELLED: The P22 capsid is a T=7 icosahedrally symmetric protein shell with a portal protein dodecamer at one 5-fold vertex. Extending outwards from that vertex is a short tail, and putatively extending inwards is a 15-nm-long α-helical barrel formed by the C-terminal domains of portal protein subunits. In addition to the densely packed genome, the capsid contains three "ejection proteins" (E-proteins [gp7, gp16, and gp20]) destined to exit from the tightly sealed capsid during the process of DNA delivery into target cells. We estimated their copy numbers by quantitative SDS-PAGE as approxi

BACKGROUND: Chronic thromboembolic pulmonary hypertension (CTEPH) has traditionally been considered a long-term complication of pulmonary embolism. However, a substantial proportion of patients have no history of venous thromboembolism (VTE), suggesting underlying biological heterogeneity.
METHODS: We analyzed clinical and imaging characteristics of 712 patients with CTEPH stratified by prior VTE history. Whole-genome sequencing was performed in all patients and 2952 healthy controls to assess common variants. Bulk RNA sequencing of pulmonary endarterectomy tissues and plasma protein quantifi

BACKGROUND: [177Lu]Lu-PSMA radioligand therapy targets metastatic castration-resistant prostate cancer by delivering radiation to cells expressing prostate-specific membrane antigen (PSMA). While some patients show remarkable responses, up to 50% show little to no benefit, and disease progression inevitably occurs. Eligibility is mainly based on PSMA avidity by PET imaging, yet responses remain highly variable, highlighting a disconnect between PSMA expression and clinical efficacy. This study used PSMA-positive patient-derived xenografts (PDXs) from the Melbourne Urological Research Alliance

Despite substantial advances in genomic testing, many individuals with neurodevelopmental disorders remain without a molecular diagnosis, while others receive a genetic diagnosis that does not fully explain phenotypic variability, developmental trajectory or tissue-specific consequences. Artificial intelligence (AI)-assisted methods are increasingly used for phenotyping, variant prioritisation, splice prediction, protein modelling, DNA methylation episignature classification and multi-omic analysis. However, these approaches differ substantially in evidentiary status and are often applied as s

BACKGROUND: Predicting response to treatment and long-term disability in multiple sclerosis (MS) remains challenging. In other complex diseases, combining genetic risk variants has enabled the detection of relevant clinical endophenotypes associated with important outcomes, but this strategy has never been applied to MS.
METHODS: We applied unsupervised hierarchical clustering to genomic risk scores in a prospective Welsh MS cohort (n=1455) and replicated the findings in the postmortem Netherlands Brain Bank (NBB) MS (NBB-MS) cohort (n=272). Disease progression was assessed using survival ana

BACKGROUND: New treatments for transthyretin amyloidosis improve survival, but diagnosis remains challenging. Pathogenic or likely pathogenic (P/LP) variants in the transthyretin (TTR) gene are one cause of transthyretin amyloidosis, and genomic screening has been proposed to identify at-risk individuals. However, data on disease features and penetrance are lacking to inform the utility of such population-based genomic screening for TTR.
OBJECTIVES: This study characterized the prevalence of P/LP variants in TTR identified through exome sequencing and the burden of associated disease from ele
