Surveillance of above- and below-ground mosquito breeding habitats in a rural midwestern community: baseline data for larvicidal control measures against West Nile Virus vectors.

BACKGROUND: Mosquitoes in the genus Culex are thought to play a major role as vectors in the transmission cycle of West Nile virus (WNV) and other arboviruses in the United States. Effective control of mosquitoes through larviciding and adulticiding is expensive for communities and should be guided by reliable surveillance data on the distribution of mosquito breeding habitats. However, few small to medium sized cities in rural areas of the midwestern United States have this type of baseline information available. OBJECTIVE: During the summer of 2004, we investigated the characteristics of Cu

Comparative Analysis of the Metabolic Profiles of Strains of Tribolium castaneum (Herbst) Adults with Different Levels of Phosphine Resistance Based on Direct Immersion Solid-Phase Microextraction and Gas Chromatography-Mass Spectrometry.

The management of phosphine (PH3) resistance in stored grain pests is an essential component of implementing timely and effective pest control strategies. The prevailing standard method for PH3 resistance testing involves the exposure of adult insects to a specific concentration over a fixed period. Although it is widely adopted, this method necessitates an extensive period for assay preparation and diagnosis. To address this issue, this study employed Direct Immersion Solid-Phase Microextraction (DI-SPME) coupled with Gas Chromatography-Mass Spectrometry (GC-MS) to compare and analyze the met

Parasitism by Cuscuta pentagona attenuates host plant defenses against insect herbivores.

Considerable research has examined plant responses to concurrent attack by herbivores and pathogens, but the effects of attack by parasitic plants, another important class of plant-feeding organisms, on plant defenses against other enemies has not been explored. We investigated how attack by the parasitic plant Cuscuta pentagona impacted tomato (Solanum lycopersicum) defenses against the chewing insect beet armyworm (Spodoptera exigua; BAW). In response to insect feeding, C. pentagona-infested (parasitized) tomato plants produced only one-third of the antiherbivore phytohormone jasmonic acid (

The Use of Disabled Insecticidal Proteins (DIPs) to Investigate the Interaction Between Aedes aegypti-Active Toxins from Bacillus thuringiensis.

A variety of pesticidal proteins derived from the bacterium Bacillus thuringiensis exhibit activity against the yellow fever mosquito Aedes aegypti and are used to control this insect vector. Several of these proteins, including Cry1Ca and Cry2Aa, additionally have activity against lepidopteran insects. Furthermore, the specificity of Cry2Aa has recently been shown to depend on domain I of the Cry protein, whereas it is generally recognized that domain II is the primary specificity-determining domain. This work has made use of disabled forms of three Cry proteins (Cry2Aa, Cry1Ca and Cry11Aa) a

Mating harassment may boost the effectiveness of the sterile insect technique for Aedes mosquitoes.

The sterile insect technique is based on the overflooding of a target population with released sterile males inducing sterility in the wild female population. It has proven to be effective against several insect pest species of agricultural and veterinary importance and is under development for Aedes mosquitoes. Here, we show that the release of sterile males at high sterile male to wild female ratios may also impact the target female population through mating harassment. Under laboratory conditions, male to female ratios above 50 to 1 reduce the longevity of female Aedes mosquitoes by reducin

CRISPR/Cas9-Mediated Mutagenesis of Abdominal-A and Ultrabithorax in the Asian Corn Borer, Ostrinia furnacalis.

(1) Background: Abdominal-A (Abd-A) and Ultrabithorax (Ubx) are homeotic genes that determine the identity and morphology of the thorax and abdomen in insects. The Asian corn borer, Ostrinia furnacalis (Guenée) (Lepidoptera: Pyralidae), is a devastating maize pest throughout Asia, the Western Pacific, and Australia. Building on previous knowledge, we hypothesized that the knockout of Abd-A and Ubx would disrupt the abdominal body planning in O. furnacalis. (2) Methods: CRISPR/Cas9-targeted mutagenesis was employed to decipher the functions of these homeotic genes. (3) Results: Knockout insects

Temperature Sensitivity of Wild-Type, Mutant and Genetic Sexing Strains of Ceratitis capitata.

Area-wide integrated pest management (AW-IPM) programmes with a sterile insect technique component (SIT) are used to control populations of insect pests worldwide, including the Mediterranean fruit fly, Ceratitis capitata. SIT consists of the mass rearing, radiation-induced sterilization, handling, and release of sterile insects over the target area. Although SIT can be performed by using both sterile males and females, male-only releases significantly increase the efficiency and cost-effectiveness of SIT applications. Male-only releases can be achieved by using genetic sexing strains (GSS). T

Brassinosteroids mediate susceptibility to brown planthopper by integrating with the salicylic acid and jasmonic acid pathways in rice.

Improved knowledge of the interactions between plants and insects will facilitate better insect control in crops. Brassinosteroids (BRs) play a vital role in plant growth, developmental processes, and responses to pathogen infection, but the role of BRs in interactions between plants and insects remain largely unknown. In this study, we characterized a negative role of BRs in rice defense against brown planthopper (BPH, Nilaparvata lugens) and examined its underlying mechanisms. We found that BPH infestation suppressed the BR pathway while successively activating the salicylic acid (SA) and ja…

Integration of endocrine signals that regulate insect ecdysis.

The extremely large number of insects and members of allied groups alive today suggests that molting--shedding of an old cuticle--may be one of the most commonly performed behaviors on our planet. Removal of an old cuticle in insects is associated with stereotyped, species-specific patterns of behavior referred to as ecdysis. It has been recognized for decades that the initiation of ecdysis is under hormonal control, but until recently many of the key peptides that regulate ecdysis were unknown. The report in 1996 of a new ecdysis-triggering hormone (ETH) sparked an era of significant advances

Direct effects of recombinant nuclear polyhedrosis viruses on selected nontarget organisms.

A limitation to effective field use of naturally occurring nuclear polyhedrosis viruses (NPVs) is the slow rate at which they kill their host. In making NPVs a more attractive pest management tool, this problem has been addressed by modifying NPVs genetically to express insecticidal proteins resulting in substantial increases in their speed of action. One concern associated with these recombinant NPVs, however, is their effects on nontarget insects associated with pests targeted for control by applications of NPVs. Our studies evaluated the direct effects of wild-type Autographa californica NP

A YOLO11-MSC-Based Method for Small Pest Detection in Insect Pest Monitoring Lamp Images.

To address the challenges of small pest target sizes, dense distributions, strong background interference, and missed detection of small objects in insect pest monitoring lamp images, this study proposes YOLO11-MSC, a lightweight agricultural pest detection model based on an improved YOLO11. First, an Object-Centric Adaptive Slicing (OCAS) strategy is designed to construct the Insect35_OCAS dataset, which increases the relative proportion of small pest targets in the input images while reducing redundant background interference. Second, the Multi-path Nonlinear Star Aggregation (MNS) module an

Regulation of sterol nutrition in the fall armyworm, Spodoptera frugiperda.

Plant sterols constitute the sole source of sterol nutrition for insect herbivores, yet the sterol content of plants is complex and variable, and insect sterol nutrient regulation remains to be elucidated. In this study, we investigated cholesterol regulation inSpodoptera frugiperda using a geometric framework for nutrition. In the choice experiment, we provided larvae with eight food pairings. The insects did not adjust their feeding on the pair of complementary diets to achieve a specific cholesterol intake target. In the no-choice experiment, seven foods with different cholesterol concentra

The interaction between the Dbf4 ortholog Chiffon and Gcn5 is conserved in Dipteran insect species.

Chiffon is the sole Drosophila ortholog of Dbf4, the regulatory subunit for the cell-cycle kinase Cdc7 that initiates DNA replication. In Drosophila, the chiffon gene encodes two polypeptides with independent activities. Chiffon-A contains the conserved Dbf4 motifs and interacts with Cdc7 to form the Dbf4-dependent Kinase (DDK) complex, which is essential for a specialized form of DNA replication. In contrast, Chiffon-B binds the histone acetyltransferase Gcn5 to form the Chiffon histone acetyltransferase (CHAT) complex, which is necessary for histone H3 acetylation and viability. Survey of infection and determination of the transmission vector of Onchocerca fasciata in camels (Camelus bactrianus) in Inner Mongolia, China.

Onchocerciasis in camels is caused by adult Onchocerca spp. and results in great economic losses to the camel industry. However, only a few studies on Onchocerca have been conducted, especially regarding the intermediate host and vector(s). In the present study, 192 camels were examined from December and January during 2013 and 2016, and the filarial larvae suspected to be Onchocerca spp. were further identified. Furthermore, aquatic dipteran insects in the living environment of camels were collected from May to September between 2013 and 2017 and dissected. Eventually, onchocercal lesions wer

Identification and Evaluation of Reference Genes for Quantitative PCR Normalization in Alligator Weed Flea Beetle (Coleoptera: Chrysomelidae).

Stably expressed reference genes are critical internal standards for the quantification of gene transcription levels using quantitative real-time PCR. Housekeeping genes are commonly used as reference genes but their expressions were variable depending on experimental conditions in many insect species studied. Here we report the identification and evaluation of 10 housekeeping genes in alligator weed flea beetle, Agasicles hygrophila Selman & Vogt (Coleoptera: Chrysomelidae), a biocontrol agent of alligator weed. The 10 housekeeping genes are: beta-actin (Actin), ribosomal protein L13A (PRL13a

Natural products in crop protection.

The tremendous increase in crop yields associated with the 'green' revolution has been possible in part by the discovery and utilization of chemicals for pest control. However, concerns over the potential impact of pesticides on human health and the environment has led to the introduction of new pesticide registration procedures, such as the Food Quality Protection Act in the United States. These new regulations have reduced the number of synthetic pesticides available in agriculture. Therefore, the current paradigm of relying almost exclusively on chemicals for pest control may need to be rec
