Bumetanide Derivatives AqB007 and AqB011 Selectively Block the Aquaporin-1 Ion Channel Conductance and Slow Cancer Cell Migration.

Aquaporins (AQPs) in the major intrinsic family of proteins mediate fluxes of water and other small solutes across cell membranes. AQP1 is a water channel, and under permissive conditions, a nonselective cation channel gated by cGMP. In addition to mediating fluid transport, AQP1 expression facilitates rapid cell migration in cell types including colon cancers and glioblastoma. Work here defines new pharmacological derivatives of bumetanide that selectively inhibit the ion channel, but not the water channel, activity of AQP1. Human AQP1 was analyzed in the Xenopus laevis oocyte expression syst

Acid-sensing ion channel 2 (ASIC2) is selectively localized in the cilia of the non-sensory olfactory epithelium of adult zebrafish.

Ionic channels play key roles in the sensory cells, such as transducing specific stimuli into electrical signals. The acid-sensing ion channel (ASIC) family is voltage-insensitive, amiloride-sensitive, proton-gated cation channels involved in several sensory functions. ASIC2, in particular, has a dual function as mechano- and chemo-sensor. In this study, we explored the possible role of zebrafish ASIC2 in olfaction. RT-PCR, Western blot, chromogenic in situ hybridization and immunohistochemistry, as well as ultrastructural analysis, were performed on the olfactory rosette of adult zebrafish. A…

Acid-sensing ion channel 1 and nitric oxide synthase are in adjacent layers in the wall of rat and human cerebral arteries.

Extracellular acidification activates a family of proteins known as acid-sensing ion channels (ASICs). One ASIC subtype, ASIC type 1 (ASIC1), may play an important role in synaptic plasticity, memory, fear conditioning and ischemic brain injury. ASIC1 is found primarily in neurons, but one report showed its expression in isolated mouse cerebrovascular cells. In this study, we sought to determine if ASIC1 is present in intact rat and human major cerebral arteries. A potential physiological significance of such a finding is suggested by studies showing that nitric oxide (NO), which acts as a pow…

Diminished vision in healthy aging is associated with increased retinal L-type voltage gated calcium channel ion influx.

Extensive evidence implicates an increase in hippocampal L-type voltage-gated calcium channel (L-VGCC) expression, and ion influx through these channels, in age-related cognitive declines. Here, we ask if this "calcium hypothesis" applies to the neuroretina: Is increased influx via L-VGCCs related to the well-documented but poorly-understood vision declines in healthy aging? In Long-Evans rats we find a significant age-related increase in ion flux through retinal L-VGCCs in vivo (manganese-enhanced MRI (MEMRI)) that are longitudinally linked with progressive vision declines (optokinetic tracki

Effects of the potassium ion channel modulators BMS-204352 Maxipost and its R-enantiomer on salicylate-induced tinnitus in rats.

Currently, there are no effective pharmacological therapies for chronic tinnitus despite a number of efforts from clinical studies and more recently, studies in animals using compounds to enhance endogenous inhibition or reduce central hyperactivity. The purpose of the current study was to evaluate the therapeutic efficacy of a novel anxiolytic with potassium channel activity in suppressing salicylate induced tinnitus in animals. Kv7 potassium channels are present in the peripheral and central auditory system where they are believed to modulate neural activity. Maxipost, a compound which atten

Neuroprotective effect of acid-sensing ion channel inhibitor psalmotoxin-1 after hypoxia-ischemia in newborn piglet striatum.

Na+,Ca2+-permeable acid-sensing ion channel 1a (ASIC1a) is involved in the pathophysiologic process of adult focal brain ischemia. However, little is known about its role in the pathogenesis of global cerebral ischemia or newborn hypoxia-ischemia (H-I). Here, using a newborn piglet model of asphyxia-induced cardiac arrest, we investigated the effect of ASIC1a-specific blocker psalmotoxin-1 on neuronal injury. During asphyxia and the first 30min of recovery, brain tissue pH fell below 7.0, the approximate activation pH of ASIC1a. Psalmotoxin-1 injection at 20min before hypoxia, but not at 20min

A dynamic pharmacophore drives the interaction between Psalmotoxin-1 and the putative drug target acid-sensing ion channel 1a.

Acid-sensing ion channel 1a (ASIC1a) is a primary acid sensor in the peripheral and central nervous system. It has been implicated as a novel therapeutic target for a broad range of pathophysiological conditions including pain, ischemic stroke, depression, and autoimmune diseases such as multiple sclerosis. The only known selective blocker of ASIC1a is π-TRTX-Pc1a (PcTx1), a disulfide-rich 40-residue peptide isolated from spider venom. π-TRTX-Pc1a is an effective analgesic in rodent models of acute pain and it provides neuroprotection in a mouse model of ischemic stroke. Thus, understanding th

Altered ion channel formation by the Parkinson's-disease-linked E46K mutant of alpha-synuclein is corrected by GM3 but not by GM1 gangliosides.

Alpha-synuclein (alpha-syn) is an amyloidogenic protein that plays a key role in the pathogenesis of Parkinson's disease (PD). The ability of alpha-syn oligomers to form ionic channels is postulated as a channelopathy mechanism in human brain. Here we identified a ganglioside-binding domain in alpha-syn (fragment 34-50), which includes the mutation site 46 linked to a familial form of PD (E46K). We show that this fragment is structurally related to the common glycosphingolipid-binding domain (GBD) shared by various microbial and amyloid proteins, including Alzheimer's beta-amyloid peptide. alp

Changes in left ventricular repolarization and ion channel currents following a transient rate increase superimposed on bradycardia in anesthetized dogs.

INTRODUCTION: We previously demonstrated in dogs that a transient rate increase superimposed on bradycardia causes prolongation of ventricular refractoriness that persists for hours after resumption of bradycardia. In this study, we examined changes in membrane currents that are associated with this phenomenon. METHODS AND RESULTS: The whole cell, patch clamp technique was used to record transmembrane voltages and currents, respectively, in single mid-myocardial left ventricular myocytes from dogs with 1 week of complete AV block; dogs either underwent 1 hour of left ventricular pacing at 120

Expression of the P2X(2) receptor subunit of the ATP-gated ion channel in the cochlea: implications for sound transduction and auditory neurotransmission.

Extracellular ATP has multimodal actions in the cochlea affecting hearing sensitivity. ATP-gated ion channels involved in this process were characterized in the guinea pig cochlea. Voltage-clamped hair cells exhibited a P2 receptor pharmacology compatible with the assembly of ATP-gated ion channels from P2X(2) receptor subunits. Reverse transcription-PCR experiments confirmed expression of the P2X(2-1) receptor subunit mRNA isoform in the sensory epithelium (organ of Corti); a splice variant that confers desensitization, P2X(2-2), was the predominant subunit isoform expressed by primary audito…

[Gramicidin channels: a new mechanism for transmembrane transfer of ions (from high resolution x-ray structural studies of the antibiotic)].

The crystal structure of the membrane-active antibiotic-cyclopeptide gramicidin S complex with urea was determined by the X-ray structure analysis. The gramicidin S molecule possesses an antiparallel beta-structure, its slightly twisted 30-membered cycle has a roughly rectangular form about 4.8 x 13.6 A in size, with the lesser side being formed by the main chain atoms of Phe and Pro residues. The maximum size of the molecule is 22.9 A. A characteristic feature of the molecule is the position of the extended side chains of the Orn residues on one side of the molecular cycle in the form of pecu

Comparative effects of fast- and slow-ion channel blocking agents on reperfusion-induced arrhythmias in the isolated perfused rat heart.

The effects of bepridil (1-4 microM), a new antianginal agent which reduces the intensity of both the fast and slow inward ionic currents in myocardium, on reperfusion-induced arrhythmias (RA) in the isolated perfused rat heart were compared with those of tetrodotoxin (0.16-1.57 microM), verapamil (0.5-2 microM), diltiazem (1-2 microM), nifedipine (0.02-0.2 microM), and nitrendipine (0.02-0.2 microM). In comparable negative inotropic concentrations, neither nifedipine nor nitrendipine reduced the incidence of RA, whereas the other four agents did. Protection against RA does not appear to be re

Spatial organization of voltage-gated ion channel expression across molecularly defined neuronal populations in the mouse mammillary bodies.

The mammillary bodies (MB) are a hypothalamic component of the limbic Papez circuit that plays a critical role in spatial and episodic memory in mammals. Degeneration of the MB occurs in disorders associated with cognitive impairment, including Korsakoff's syndrome and Alzheimer's disease, yet the molecular organization and intrinsic properties of MB neurons remain poorly understood. Recent single-cell RNA sequencing identified multiple transcriptionally distinct neuronal populations within the MB and suggested that they differentially express voltage-gated ion channels that regulate neuronal …

Exploratory Immunohistochemical Mapping of Mechanosensitivity-Associated Ion Channel Proteins in Terminal Glial Cells of Human Meissner and Pacinian Corpuscles.

Terminal glial cells (TGCs) of cutaneous end-organ complexes have traditionally been regarded as structural and trophic elements, although they may also participate in mechanosensory processing. We performed an exploratory immunohistochemical and immunofluorescence study of mechanosensitivity-associated ion channel proteins in human Meissner and Pacinian corpuscles using archival formalin-fixed, paraffin-embedded glabrous skin from 64 donors and several anatomical sites. Immunoreactivity was assessed separately in mechanoreceptor axons and TGCs. In Meissner corpuscles, TGC-associated immunorea

Advances in neuropharmacology: Innovative drug strategies targeting synaptic plasticity, neuroinflammation, and ion channel regulation for future CNS treatments.

As our understanding of the molecular and cellular mechanisms underlying central nervous system (CNS) disorders expands, neuropharmacology is undergoing a paradigm shift. Despite the increasing burden of neurodegenerative and neuropsychiatric diseases, challenges such as blood-brain barrier (BBB) impermeability, drug resistance, and complex pathophysiology continue to limit effective therapeutic development. This review highlights key emerging targets, including ion channel regulation, neuroinflammation, and synaptic plasticity, as important entry points for next-generation neurotherapeutics.

Selective pulmonary vasorelaxant effects of an N2,N4-diaminoquinazoline PDE5 inhibitor involving endothelial signaling and ion channel modulation.

Pulmonary arterial hypertension (PAH) is a progressive disease characterized by elevated pulmonary vascular resistance and right ventricular overload. Phosphodiesterase type-5 (PDE5) inhibitors are standard therapy but are limited by systemic vasodilation and side effects. N2,N4-diaminoquinazoline and N2,N6-diaminopurine derivatives have been identified as selective PDE5 inhibitors, however, achieving pulmonary vascular selectivity remains challenging. This study therefore evaluated the vasorelaxant effects of seven potent derivatives developed from the initial lead compound A with novel biois

The calcium ion channel protein TRPML is indispensable to the reproduction of Nilaparvata lugens and the transovarial transmission of its yeast-like symbionts.

Transient receptor potential mucolipin (TRPML, also known as mucolipin-3), a critical cation channel protein in mammals, mediates ion transport, and regulates diverse signaling pathways and cellular physiological functions. However, its functional significance in insects remains poorly characterized. In this study, we cloned TRPML of Nilaparvata lugens (NlTRPML), employed bioinformatics tools to predict its structure and function, elucidating its tertiary architecture and potential binding sites for the small-molecule agonist mucolipin synthetic agonist 1(ML-SA1). Quantitative real-time (qRT)-
