Susceptibility of gut indigenous lactic acid bacteria in BALB/c mice to oral administered Lactobacillus plantarum.

Cells of Lactobacillus plantarum strains AN1 and Tennozu-SU2 exert anti-inflammatory responses in ICR mouse models of inflammatory bowel disease and protective effects against S. Typhimurium infection in BALB/c mice, respectively. To clarify the existence of L. plantarum-susceptible gut indigenous bacteria, AN1 and Tennozu-SU2 cells were administered to BALB/c mice via drinking water. Gene amplicon sequencing of 16S rRNA of caecal content revealed that the AN1 and Tennozu-SU2 cells affected the abundance of caecal indigenous lactobacilli, but the effect on the dominant Clostridiales and Bacter…

Immunogenicity of eGFP-Marked Recombinant Lactobacillus casei against Transmissible Gastroenteritis Virus and Porcine Epidemic Diarrhea Virus.

Porcine transmissible gastroenteritis virus (TGEV) and porcine epidemic diarrhea virus (PEDV) are the causative agents of highly fatal acute diarrhea in pigs, resulting in enormous losses in the pig industry worldwide. To develop an effective bivalent oral vaccine against TGEV and PEDV infection, the D antigenic site of the TGEV spike (S) protein and the major antigen site (core neutralizing epitope-COE) of the PEDV S protein were used as immunogens, and the enhanced green fluorescent protein (eGFP) gene was used as a reporter to construct genetically engineered Lactobacillus casei rLpPGF-T7g1…

Lactobacillus plantarum producing a Chlamydia trachomatis antigen induces a specific IgA response after mucosal booster immunization.

Mucosal immunity is important for the protection against a wide variety of pathogens. Traditional vaccines administered via parenteral route … a high priority. In the present study, we have explored the use of a common lactic acid bacterium, Lactobacillus plantarum, as a vector for delivery of a C. trachomatis antigen to mucosal sites. The antigen, referred as Hirep2 (H2), was anchored to the surface of L. plantarum cells using an N-terminal lipoprotein anchor. After characterization, the constructed …

Oral administration of heat-killed Lactobacillus kunkeei YB38 improves murine influenza pneumonia by enhancing IgA production.

Influenza is one of the important respiratory tract infections that require special attention for maintaining health and hygiene. The removal of influenza virus (IFV) by secretory IgA produced by the respiratory epithelium has been reported to be a critical host defense mechanism. Therefore, we isolated Lactobacillus kunkeei YB38 (YB38), the promoter of the salivary IgA secretion in humans, from honeybee pollen and studied the effect of heat-killed YB38 treatment for preventing IFV infection in a mouse model. Female BALB/c mice received YB38 orally for 21 consecutive days and were then inocula…

Orally administered Lactobacillus plantarum reduces pro-inflammatory interleukin secretion in sera from Listeria monocytogenes infected mice.

Lactic acid bacteria have traditionally been thought to have immunomodulating effects. To verify this property, Lactobacillus plantarum was orally administered to mice (5 x 107 colony forming units (c.f.u.)), prior to infection with Listeria monocytogenes in order to evaluate the host resistance against an infectious micro-organism and to better define the influence of L. plantarum on such responses. Balb/c mice were treated daily with L. plantarum or received PBS (sham-treated mice as controls) for 4 weeks. Subsequently, mice were intravenously infected with a clinical isolate of L. monocytog…

Toward clinical translation: montmorillonite-enhanced Lactobacillus biofilm alleviates colitis by modulating the gut microbiota-bile acid axis.

Oral probiotics hold therapeutic potential for ulcerative colitis (UC), but their low bioavailability greatly limits clinical efficacy. Here, we designed a montmorillonite-Lactobacillus acidophilus biofilm (MLB) delivery strategy to enhance probiotic stability, intestinal adhesion, and therapeutic efficiency. MLB was prepared by inducing the clinically common strain Lactobacillus acidophilus to form biofilms on montmorillonite, an antidiarrheal agent widely used in clinics. The in vitro assays demonstrated that montmorillonite significantly promoted biofilm formation, thereby improving bacteri…

A fermented milk drink containing Lactobacillus casei strain Shirota modulates the esophageal microbiome composition in Barrett's esophagus.

Esophageal adenocarcinoma and its precursor, Barrett's esophagus (BE), are associated with a pro-inflammatory, Gram-negative-dominated esophageal microbiome. In a single-arm pilot study, 23 patients with BE consumed a fermented milk drink containing Lactobacillus casei strain Shirota (LcS) twice daily for 4 weeks, based on the hypothesis that this intervention could shift the microbiome toward a more beneficial Gram-positive profile. Post-intervention, metaplastic columnar epithelium showed a significant increase in Gram-positive Firmicutes (p < 0.01) without changes in overall Eubacterial abu…

Structure-property relationships of alginate-protein-polysaccharide composites for encapsulation and controlled release of Lactobacillus bulgaricus LB12.

This study investigates alginate-based composites for encapsulating the probiotic strain Lactobacillus delbrueckii ssp. bulgaricus (LB12). Alginate/casein, alginate/carrageenan and alginate/casein/κ-carrageenan composites and their formulations with LB12 were prepared by ionic gelation using calcium chloride. Combined analyses (FTIR, rheological, thermal, morphological, and bacterial cell release from formulations) reveal that the microparticle network structure and physicochemical properties depend on complex interactions among constituents. FTIR spectra confirm the formation of crosslinked c…

Oleogel fabrication from Lactobacillus-stabilized emulsion templates: The role of bacterial surface properties in network stabilization mechanisms.

Lactobacillus cells, owing to their combined interfacial stabilizing and potentially postbiotic functionalities, have emerged as promising natural Pickering stabilizers. However, the relationship between bacterial surface properties, emulsion-template stability, and oleogel formation remains poorly understood. In this study, eight Lactobacillus strains with distinct surface properties were employed to subsequently construct Pickering emulsion templates and oleogels. Principal component analysis revealed distinct relationships among bacterial surface properties and emulsion-template characteris…

Lactobacillus casei Fermentation Enhances the Sensory Quality of Shoumei Tea Infusion: Based on Flavor and Metabolic Analysis.

Shoumei white tea is widely consumed, yet its limited product diversity constrains its economic value. This study used Shoumei tea infusion as a fermentation substrate to examine how Lactobacillus casei modulates flavor and underlying biochemical pathways. During 72 h of fermentation, the pH decreased from 5.0 to 4.1, accompanied by a 95.5% reduction in soluble sugars (from 10,056 to 453 mg/L). The accumulation of organic acids, particularly lactic acid (rising from 873.90 to 1268.17 µg/mL) and malic acid (rising from 150.83 to 725.18 µg/mL), drove the increase in sourness. Concurrently, subst…
