Lipopolysaccharide biogenesis and transport at the outer membrane of Gram-negative bacteria.

The outer membrane (OM) of Gram-negative bacteria is an asymmetric lipid bilayer containing a unique glycolipid, lipopolysaccharide (LPS) in its outer leaflet. LPS molecules confer to the OM peculiar permeability barrier properties enabling Gram-negative bacteria to exclude many toxic compounds, including clinically useful antibiotics, and to survive harsh environments. Transport of LPS poses several problems to the cells due to the amphipatic nature of this molecule. In this review we summarize the current knowledge on the LPS transport machinery, discuss the challenges associated with this p

The site-2 protease.

The site-2 protease (S2P) is an unusually-hydrophobic integral membrane protease. It cleaves its substrates, which are membrane-bound transcription factors, within membrane-spanning helices. Although structural information for S2P from animals is lacking, the available data suggest that cleavage may occur at or within the lipid bilayer. In mammalian cells, S2P is essential owing to its activation of the sterol regulatory element binding proteins (SREBPs); in the absence of exogenous lipid, cells lacking S2P cannot survive. S2P is also important in the endoplasmic reticulum (ER) stress response

The origin of parasitophorous vacuole membrane lipids in malaria-infected erythrocytes.

During invasion of an erythrocyte by a malaria merozoite, an indentation develops in the erythrocyte surface at the point of contact between the two cells. This indentation deepens as invasion progresses, until the merozoite is completely surrounded by a membrane known as the parasitophorous vacuole membrane (PVM). We incorporated fluorescent lipophilic probes and phospholipid analogs into the erythrocyte membrane, and followed the fate of these probes during PVM formation with low-light-level video fluorescence microscopy. The concentration of probe in the forming PVM was indistinguishable fr

Extracellular Vesicles and the Placenta-Brain Axis: Mechanisms of Shared Risk in Pregnancy and Perinatal Psychiatry.

Disorders of pregnancy and psychiatric conditions in mothers and affected children have high rates of comorbidity and underlying mechanistic overlap. We propose that extracellular vesicles (EVs) may subserve this shared psycho-obstetric risk. EVs are small lipid bilayer-bound particles containing regulatory nucleic acids and proteins that are released from cells into circulation, where they readily cross blood-brain and placental barriers. In pregnancy, EVs are produced in large numbers by the placenta and have been implicated in pregnancy-related medical and psychiatric conditions via vascula

Nuclear pores form de novo from both sides of the nuclear envelope.

Nuclear pore complexes are multiprotein channels that span the double lipid bilayer of the nuclear envelope. How new pores are inserted into the intact nuclear envelope of proliferating and differentiating eukaryotic cells is unknown. We found that the Nup107-160 complex was incorporated into assembly sites in the nuclear envelope from both the nucleoplasmic and the cytoplasmic sides. Nuclear pore insertion required the generation of Ran guanosine triphosphate in the nuclear and cytoplasmic compartments. Newly formed nuclear pore complexes did not contain structural components of preexisting p

Molecular Rationale for Improved Dynamic Nuclear Polarization of Biomembranes.

Dynamic nuclear polarization (DNP) enhanced solid-state NMR can provide orders of magnitude in signal enhancement. One of the most important aspects of obtaining efficient DNP enhancements is the optimization of the paramagnetic polarization agents used. To date, the most utilized polarization agents are nitroxide biradicals. However, the efficiency of these polarization agents is diminished when used with samples other than small molecule model compounds. We recently demonstrated the effectiveness of nitroxide labeled lipids as polarization agents for lipids and a membrane embedded peptide. H

Ca(2+)-dependent annexin self-association on membrane surfaces.

Annexin self-association was studied with 90 degrees light scattering and resonance energy transfer between fluorescein (donor) and eosin (acceptor) labeled proteins. Synexin (annexin VII), p32 (annexin IV), and p67 (annexin VI) self-associated in a Ca(2+)-dependent manner in solution. However, this activity was quite labile and, especially for p32 and p67, was not consistently observed. When bound to chromaffin granule membranes, the three proteins consistently self-associated and did so at Ca2+ levels (pCa 5.0-4.5) approximately 10-fold lower than required when in solution. Membrane orientation regulates GAPR-1 oligomerization that is suitable for interaction with beclin 1.

Golgi-associated plant pathogenesis-related protein 1 (GAPR-1) is a beclin 1-interacting peripheral membrane protein that is anchored to the Golgi complex via an N-myristoyl-group and ionic interactions. Here we investigated the determinants of GAPR-1 membrane binding using molecular dynamics (MD) simulations and applied the results to investigate the subsequent oligomerization into biomolecular condensates and interaction with beclin 1. MD simulations revealed a specific orientation of GAPR-1 upon docking to a negatively charged lipid bilayer involving lysines at position 7 and 33 (Lys7 and L

Alpha-actinin associates with polycystin-2 and regulates its channel activity.

Polycystin-2 (PC2) is the product of the PKD2 gene, which is mutated in 10-15% patients of autosomal dominant polycystic kidney disease (ADPKD). PC2 is an integral transmembrane protein and acts as a calcium-permeable cation channel. The functional modulation of this channel by other protein partners remains largely unknown. In the present study, using a yeast two-hybrid approach, we discovered that both intracellular N- and C-termini of PC2 associate with alpha-actinins, actin-binding and actin-bundling proteins important in cytoskeleton organization, cell adhesion, proliferation and migratio

The effect of merulinic acid on biomembranes.

Merulinic acid (heptadecenylresorcinolic acid, resorcinolic acid) is one of the members of resorcinolic lipids, the natural amphiphilic long-chain homologues of orcinol (1,3-dihydroksy-5-methylbenzene). In the present study, membrane properties of merulinic acid were investigated. Merulinic acid exhibits strong haemolytic activity against sheep erythrocytes (EH50 of 5+/-2 microM) regardless of the form of its application-direct injection into the erythrocyte suspension or injection as merulinic acid-enriched liposomes. The lysis of erythrocytes induced by merulinic acid was inhibited by the pr

Antioxidant and Membrane Binding Properties of Serotonin Protect Lipids from Oxidation.

Serotonin (5-hydroxytryptamine, 5-HT) is a well-known neurotransmitter that is involved in a growing number of functions in peripheral tissues. Recent studies have shown nonpharmacological functions of 5-HT linked to its chemical properties. Indeed, it was reported that 5-HT may, on the one hand, bind lipid membranes and, on the other hand, protect red blood cells through a mechanism independent of its specific receptors. To better understand these underevaluated properties of 5-HT, we combined biochemical, biophysical, and molecular dynamics simulations approaches to characterize, at the mole

Research progress on exosomal proteins as diagnostic markers of gastric cancer (review article).

Gastric cancer (GC) is one of the most common types of tumors and the most common cause of cancer mortality worldwide. The diagnosis of GC is critical to its prevention and treatment. Available tumor markers are the crucial step for GC diagnosis. Recent studies have shown that proteins in exosomes are potential diagnostic and prognostic markers for GC. Exosomes, secreted by cells, are cup-shaped with a diameter of 30-150 nm under the electron microscope. They are also surrounded by lipid bilayers and are widely found in various body fluids. Exosomes contain proteins, lipids and nucleic acid. T…

Extracellular Vesicles: New Tools for Early Diagnosis of Breast and Genitourinary Cancers.

Breast cancers and cancers of the genitourinary tract are the most common malignancies among men and women and are still characterized by high mortality rates. In order to improve the outcomes, early diagnosis is crucial, ideally by applying non-invasive and specific biomarkers. A key role in this field is played by extracellular vesicles (EVs), lipid bilayer-delimited structures shed from the surface of almost all cell types, including cancer cells. Subcellular structures contained in EVs such as nucleic acids, proteins, and lipids can be isolated and exploited as biomarkers, since they direc

Exosome-mediated cell-cell communication in tumor progression.

Exosomes, which are 30-150 nm lipid bilayer vehicles, have been recognized as one of the most crucial components of the tumor microenvironment. Exosomes transfer specific lipid, nucleic acids, proteins and other bioactive molecules from the donor cells to the recipient cells. Accumulating evidence has suggested that cancer cells and the tumor associated stromal cells can release and receive exosomes, inside of which the components and amounts are greatly changed. Pioneering studies have revealed that these exosomes play essential roles in tumor progression. Here we summarize the recent advance

New Insights into the Role of Exosomes in the Heart After Myocardial Infarction.

Intercellular communications play a pivotal role in several cardiac pathophysiological processes. One subtype of extracellular vesicles, so-called exosomes, became known as important intercellular communication mediators in the heart. Exosomes are lipid bilayer biological nanovesicles loaded with diverse proteins, lipids, and mRNAs/microRNAs. All major cardiac cell types can modulate recipient cellular function via the release of exosomes. After myocardial infarction (MI), exosomes, especially those secreted by different cardiac stem cells, have been shown to confer cardioprotective effects, a

New insights into regulation of erythrocyte shape.

Two ideas explain the mechanism of shape regulation: the lipid bilayer coupled theory and the protein network scaffold theory. Recently, several important articles have been published on the former theory. However, many phenomena argue against the theory, including behavior of ghosts and triton shells, various types of manipulation of proteins, and fixation of the shape by the addition of large reagents outside the cell. Moreover, hereditary spherocytosis shows normal, uneven distribution of phospholipids, and hereditary and artificial defects of a membrane protein show spherocytes or elliptoc

From Liposomes to Virosomes: Evolution of Phospholipid Nanocarriers in Drug Delivery.

Phospholipid-based nanocarriers are an adaptable and chemically tunable class of drug delivery systems that self-assemble into bilayered vesicles due to the amphiphilic nature of phospholipids. Phospholipid-based nanocarriers can encapsulate both hydrophilic and hydrophobic drugs through non-covalent interactions and modulation of lipid phase behavior. This review explores the molecular and supramolecular principles governing the formation, stability, and function of key phospholipid-derived nanocarriers, including liposomes, transferosomes, ethosomes, invasomes, phytosomes, pharmacosomes, and
