In the pursuit of novel antioxidant and anti-inflammatory agents, we investigated 5-hydroxyferulic acid (5-OHFA), a hydroxylated derivative of the well-known phenolic compound ferulic acid (FA). This study aimed to determine whether structural modification enhances the biological activity of FA. To this end, both compounds were subjected to a series of in vitro antioxidant assays (DPPH, ABTS, FRAP, and Fe(II)-chelating) and anti-inflammatory evaluations, complemented by in silico predictions. 5-OHFA consistently exhibited superior antioxidant capacity, with significantly lower IC50 values than

BACKGROUND AND AIMS: Sickle cell anemia (SCA) is an inherited red blood cell disorder resulting from the polymerization of Hemoglobin S. Acute vaso-occlusive crisis and multiple organ damage are the most common complications of SCA. Because of its multifunctional role in inflammation, endothelial dysfunction, and fibrosis, TGF-β1 could be involved in SCA development, which may explain a variety of symptoms associated with this disease. This study aimed to investigate the role of TGF-β1 as a noninvasive biomarker for predicting the types of SCA clinical manifestations and highlights the complic

INTRODUCTION: Alzheimer's disease (AD) is a common progressive and irreversible neurodegenerative disease. AD accounts for 60%-70% of all dementia cases, ranking as the seventh leading cause of death globally. Human umbilical cord mesenchymal stem cells (hUC-MSCs) characterized by their abundant availability and low immunogenicity, have demonstrated significant therapeutic potential for AD in both preclinical studies and clinical trials. The use of exosomes can help mitigate the issues associated with cellular therapies. However, the clinical application of hUC-MSCs remains challenging due to

BACKGROUND: Hepatocyte growth factor (HGF) is a broad-spectrum multifunctional cytokine with mitogenic, motogenic, morphogenic, and antiapoptotic functions in various types of cells. It is secreted by mesenchymal cells, including gingival fibroblasts, and its expression is induced by inflammatory cytokines, such as interleukin-1 and tumor necrosis factor-alpha, which play a potential role in periodontal destruction. Hence, the present study was carried out to assess HGF's concentration in gingival crevicular fluid (GCF) and to determine its association with periodontal disease progression.

Mutations in the Senataxin gene, SETX are known to cause the neurodegenerative disorders, ataxia with oculomotor apraxia type 2 (AOA2), and amyotrophic lateral sclerosis 4 (ALS4). However, the mechanism underlying disease pathogenesis is still unclear. The Senataxin N-terminal protein-interaction and C-terminal RNA/DNA helicase domains are conserved in the Saccharomyces cerevisiae homolog, Sen1p. Using genome-wide expression analysis, we first show alterations in key cellular pathways such as: redox, unfolded protein response, and TOR in the yeast sen1 ΔN mutant (N-terminal truncation). This m

BACKGROUND: Autosomal recessive ataxias represent a group of clinically overlapping disorders. These include ataxia with oculomotor apraxia type1 (AOA1), ataxia with oculomotor apraxia type 2 (AOA2) and ataxia-telangiectasia-like disease (ATLD). Patients are mainly characterized by cerebellar ataxia and oculomotor apraxia. Although these forms are not quite distinctive phenotypically, different genes have been linked to these disorders. Mutations in the APTX gene were reported in AOA1 patients, mutations in SETX gene were reported in patients with AOA2 and mutations in MRE11 were identified in

Cell transplantation for nucleus pulposus (NP) regeneration represents a promising strategy for intervertebral disc degeneration (IVDD). Nonetheless, the hostile microenvironment within the degenerated intervertebral discs, characterized by redox imbalance and elevated mechanical pressure, poses risks of low cell survival and inadequate cell colonization for efficient NP regeneration. To address these challenges, we developed a biomimetic, esterase-responsive composite hydrogel microsphere (GHKM) for cell delivery, consisting of gelatin methacrylate (GelMA) mixed with HAMA-KGN, a conjugate of

Recent studies have begun to reveal the complex and multifunctional roles of N6-methyladenosine (m6A) modifications and their associated writer, reader, and eraser proteins in infection by diverse RNA and DNA viruses. However, little is known about their regulation and functions during infection by several viruses, including poxviruses. Here, we show that members of the YTH Domain Family (YTHDF), in particular YTHDF2, are downregulated as the prototypical poxvirus, vaccinia virus (VacV) enters later stages of replication in a variety of natural target cell types, but not in commonly used trans

Colibactin induces DNA damage in mammalian cells and has been linked to the virulence of Escherichia coli and the promotion of colorectal cancer (CRC). By looking for mutants attenuated in the promoter activity of clbB encoding one of the key enzymes for the production of colibactin, we found that a mutant of the gene coding for the polyphosphate kinase (PPK) produced less colibactin than the parental strain. We observed this phenotype in different strains ranging from pathogens responsible for meningitis, urinary tract infection, or mouse colon carcinogenesis to the probiotic Nissle 1917. We

ConspectusIn biological systems, multivalent interactions orchestrate the hierarchical assembly of biomolecules into compartmentalized architectures, enabling spatial segregation and cooperative regulation of multiactive sites, thereby promoting complex reaction networks. Mimicking these assembly principles to develop biomimetic confined environments has emerged as a promising strategy for engineering highly efficient catalysts. Organic molecular cages (OMCs) possess discrete nanocavities and open windows, in which spatial confinement enables enzyme-mimetic encapsulation and stabilization of a

UNLABELLED: Cancer remains a leading cause of death worldwide. Colorectal cancer (CRC) is the most common type of gastrointestinal malignancy, with the combined effect of multiple etiological factors. Multifunctional nanocomposites present a promising platform for synergistic therapy of CRC. Elemene (EL), an active component in traditional Chinese medicine, exhibits both antitumor and immunostimulatory activities. However, its clinical application is limited by poor stability, low tumor accumulation, and the difficulty of effectively harnessing its dual activities. Herein, we developed an oral

This study presents the green synthesis of cobalt ferrite (CoFe2O4) and extract-mediated (phytochemical-capped) CoFe2O4-Ag nanocomposites using aqueous bark extract of Pterocarpus marsupium Roxb. The silver nanoparticles (Ag NPs) are uniformly decorated on the surface of CoFe2O4. The characterisation results from X-ray diffraction (XRD) analysis indicated a cubic spinel structure, with a crystallite size of about 23.85 nm and Ag-induced lattice strain, yielding a lattice parameter of a 0 = 8.1662 Å. The surface plasmon resonance (SPR) peak at 368 nm in the UV-vis spectroscopy spectrum revealed

Macrophages are central regulators of atherosclerosis, governing lipid accumulation, inflammatory signaling, and plaque stability. Lipin-1 is a multifunctional lipid-metabolic regulator that integrates cellular metabolism with inflammatory responses through its dual roles as a phosphatidic acid phosphatase enzyme and a transcriptional coregulator. However, its role in macrophage-driven atherosclerosis remains controversial. This review critically evaluates the domain-specific functions of lipin-1 and their impact on disease progression. Accumulating evidence indicates that lipin-1 exerts diver

Hypertension and arterial stiffening are major contributors to cardiovascular disease and are closely linked to vascular inflammation. Activation of the renin-angiotensin-aldosterone system promotes unfavorable immune responses that lead to vascular inflammation and remodeling; however, the exact molecular mediators connecting immune activation to arterial stiffening remain poorly understood. Transglutaminase 2 (TG2) is a multifunctional enzyme involved in extracellular matrix remodeling and inflammatory signaling, expressed in both vascular and immune cells. We hypothesized that TG2 in myeloi

BACKGROUND: APE1 is a multifunctional enzyme with 2 distinct functions-endonuclease activity in its C-terminal domain and redox function in its N-terminal domain. The redox activity of APE1 regulates a subset of transcription factors involved in cell survival, angiogenesis, and inflammation. However, its role in chronic rhinosinusitis (CRS) with nasal polyps is not fully understood.
OBJECTIVE: We sought to elucidate the role of APE1 redox function in the pathogenesis of CRS and to evaluate the therapeutic potential of targeting APE1 with redox inhibitors.
METHODS: APE1 expression and functio

Dry eye disease (DED) is currently the most prevalent condition seen in ophthalmology outpatient clinics, representing a significant public health issue. The onset and progression of DED are closely associated with oxidative stress-induced inflammation and damage. To address this, an aldehyde-functionalized F127 (AF127) hydrogel eye drop delivering multifunctional antioxidant Cu2-xSe nanoparticles (Cu2-xSe NPs) was designed. The research findings revealed that the Cu2-xSe nanoparticles exhibit unexpected capabilities in acting as superoxide dismutase and glutathione peroxidase. Additionally, C
