Profibrotic macrophage-derived CXCL4 promotes pericyte-to-myofibroblast transition after spinal cord injury.

INTRODUCTION: Spinal cord injury (SCI) induces fibrotic scarring that impairs axonal regeneration. Pericytes contribute to scar formation vi … d CXCL4 activates CXCR3/PI3K/Akt signaling in pericytes, driving their transition into scar-forming myofibroblasts after SCI. Blocking this axis mitigates fibrosis and enhances axonal regeneration and motor recovery. THE TRANSLATIONAL POTENTIAL OF THIS ARTICLE: This study indicates that targeting the CXCL4-driven pericyte-to-myofibroblast transition to reduce f …

Clinical characteristics and survival outcomes of adult inflammatory myofibroblastic tumor: a retrospective single-center study.

Inflammatory myofibroblastic tumor (IMT) is a rare mesenchymal neoplasm that can occur across all age groups and presents with heterogeneous clinical behavior. Clinical manifestations vary by tumor location and may include pain, swelling, or systemic inflammatory symptoms. Diagnosis relies on imaging and histopathological confirmation. A subset of IMTs harbor ALK gene rearrangements, supporting the use of targeted therapies. This study aimed to describe the demographic, clinical, pathological, and treatment-related characteristics of adult IMT patients, with emphasis on disease course, inflamm

Pediatric Mesenteric Inflammatory Myofibroblastic Tumor Presenting as a Diagnostic Challenge: A Case Report and Literature Review.

BACKGROUND: Inflammatory myofibroblastic tumor (IMT) is a rare mesenchymal neoplasm of intermediate malignant potential that predominantly affects children and young adults. Although pulmonary involvement is most frequently reported, mesenteric IMT is uncommon in pediatric patients. Its diagnosis remains challenging due to nonspecific clinical manifestations and significant radiologic and histopathologic overlap with other mesenchymal tumors. CASE PRESENTATION: We describe the case of a 5-year-old girl who presented with a three-month history of progressive abdominal distension, anorexia, and

Transoral Endoscopic Plasma Resection for Diagnosis and Management of Laryngeal Inflammatory Myofibroblastic Tumor: A Case Report.

Inflammatory myofibroblastic tumor (IMT) is an uncommon neoplasm that predominantly affects children and young adults. It can occur in soft tissues throughout the body, most commonly in the lungs, but it is rare in the larynx. A 24-year-old woman presented to the respiratory department of our hospital with breathing difficulties. Following a cervical computed tomography scan that identified a laryngeal mass, she was referred to our department for further management. The patient underwent transoral endoscopic plasma resection for laryngeal IMT under general anesthesia. The postoperative patholo

ALK-negative inflammatory myofibroblastic tumor with an undetermined differentiation direction: A case report and review of the literature.

RATIONALE: Inflammatory myofibroblastic tumor (IMT) is a rare mesenchymal neoplasm that can arise in various anatomical locations, while IMT originating from the broad ligament is exceptionally rare. Anaplastic lymphoma kinase (ALK) is a highly specific diagnostic biomarker for IMT. However, no standardized therapeutic algorithm has been established for ALK-negative IMT, largely due to the unclear risk of distant metastasis and recurrence. In cases with undefined tumor differentiation and no detectable molecular targets, close surveillance and individualized therapeutic strategies should be gu

An ALMS1 variant disrupts proximal centriole organization and promotes a myofibroblast-like phenotype that is regulated by THY1.

Primary endocardial fibroelastosis (pEFE) is characterized by accumulation of elastin-rich extracellular matrix (ECM) in the left ventricular endocardium in the absence of structural defects, but its pathophysiology remains unclear. We investigated dermal fibroblasts from a pEFE patient (proband) harboring a loss-of-function ALMS1 variant. Compared with control fibroblasts, proband cells displayed a myofibroblast-like phenotype, with increased ECM proteins and markers of fibroblast activation and endothelial-to-mesenchymal transition. Conversely, the thymus cell surface antigen (THY1) was down

Inflammatory Myofibroblastic Tumor of the Hard Palate and Gingivobuccal Sulcus Mimicking a Malignant Neoplasm: A Case Report.

Inflammatory myofibroblastic tumor (IMT) is an uncommon mesenchymal neoplasm of intermediate biological potential with rare occurrence in the oral cavity. Its broad histopathologic spectrum, coupled with variable immunohistochemical expression, renders it a significant diagnostic challenge, particularly in elderly patients with oral risk habits where malignancy is the primary clinical concern. We report a case of anaplastic lymphoma kinase (ALK)-negative IMT of the myxoid pattern arising simultaneously in the right hard palate and gingivobuccal sulcus in a 75-year-old female with a long-standi

Epigenetic Suppression of RASAL1 by HDAC3 and Cofactor YY1 Promotes Fibroblast-Myofibroblast Transition and Renal Fibrosis.

AIM: Astronauts return to Earth with reduced red cell masses and hypovolaemia. Not surprisingly, when they stand, their heart rates may speed inordinately, their blood pressures may fall, and some may experience frank syncope. We studied autonomic function in six male astronauts (average +/- SEM age: 40 +/- 2 years) before, during, and after the 16-day Neurolab space shuttle mission. METHOD: We recorded electrocardiograms, finger photoplethysmographic arterial pressures, respiration, peroneal nerve muscle sympathetic activity, plasma noradrenaline and noradrenaline kinetics, and cardiac outpu

Dipeptidyl peptidase-4 inhibitors suppress myofibroblastic differentiation and hyaluronan synthesis in Graves' orbitopathy fibroblasts.

Graves' orbitopathy (GO) is characterized by inflammation, hyaluronan accumulation, tissue expansion, and fibrosis of orbital tissues, while effective antifibrotic medical therapies remain limited. Dipeptidyl peptidase-4 inhibitors (DPP4i), widely used for type 2 diabetes, have been reported to modulate fibroblast activity and fibrotic responses in several organs. This study investigated the effects of DPP4i on primary human orbital fibroblasts and explored their potential relevance to GO. Orbital fibroblasts were obtained from patients with GO and non-GO controls. DPP4 protein expression was

Detection of an inflammatory myofibroblastic tumor in the testis by FDG-PET/CT; a rare presentation of a rare neoplasm.

FDG-PET/CT in a 73-year-old man revealed incidental FDG accumulation in the left testicle which was eventually diagnosed as an inflammatory myofibroblastic tumor (IMT): a rare localization of a rare tumor. Previous reports describe moderate to high FDG uptake in IMT but a localization in the testis on FDG-PET has not been described before. Although IMTs are often benign, they can affect nearby tissues and, in rare cases, spread to distant organs. FDG-PET can be used to guide therapy, with surgery being the preferred treatment, and to detect recurrent disease or to evaluate response to systemic

Unexpected complete resolution of intravesical inflammatory myofibroblastic tumor after immunotherapy for melanoma: A case report.

Inflammatory myofibroblastic tumors (IMT) are exceedingly rare, particularly when originating in the bladder. Complete surgical resection is the gold standard treatment for IMTs. We describe a case of IMT of the urinary bladder that achieved complete radiographic and endoscopic resolution after systemic treatment with ipilimumab and nivolumab directed towards metastatic melanoma. This case is noteworthy due to the tumor's serendipitous and remarkable response to immunotherapy.

Revisiting pulmonary fibrosis: inflammatory dynamics of the lipofibroblast-to-inflammatory lipofibroblast-to-activated myofibroblast reversible switch.

Idiopathic pulmonary fibrosis (IPF) is a chronic, progressive interstitial lung disease characterized by excessive extracellular matrix (ECM) deposition and irreversible lung damage. A key driver of disease progression is the phenotypic shift of lipofibroblasts (LIFs) into activated myofibroblasts (aMYFs), triggered by sustained epithelial injury, caused by inflammation, oxidative stress, viral infections (e.g., influenza, SARS-CoV-2), and metabolic dysfunction. Emerging evidence demonstrates that this transition is reversible, with pharmacological agents that promote aMYF-to-LIF reprogramming…

Inflammatory myofibroblastic tumor in the liver after bone marrow transplantation: case report and literature review.

INTRODUCTION: Inflammatory myofibroblastic tumor (IMT) is a rare low-grade malignant neoplasm in the liver. Timely diagnosis and treatment of IMT are challenging due to its atypical symptoms and imaging results. CASE REPORT: We report a 46-year-old woman who presented to our hospital with persistent hyperpyrexia and discomfort in the right upper abdomen for 2 months post bone marrow transplantation. Radiological findings revealed a space-occupying lesion of uncertain nature in the liver. Since the histological examination of the biopsy specimen indicated IMT, she underwent surgical resection.

PDGFRα inhibition reduces myofibroblast expansion in the fibrotic rim and enhances recovery after ischemic stroke.

Ischemic stroke is a major cause of disability in adults. Early treatment with thrombolytics and/or thrombectomy can significantly improve outcomes; however, following these acute interventions, treatment is limited to rehabilitation therapies. Thus, identification of therapeutic strategies that can help restore brain function in the post-acute phase remains a major challenge. Here we report that genetic or pharmacologic inhibition of the PDGF-CC/PDGFRα pathway, which has previously been implicated in stroke pathology, significantly reduced myofibroblast expansion in the border of the fibrotic

Inflammatory Myofibroblastic Tumor of the Bladder in an Adolescent Girl: A Case Report on Successful Management With Partial Cystectomy.

Inflammatory myofibroblastic tumors (IMTs) are rare bladder tumors that are difficult to differentiate from other malignant bladder tumors because of their non-specific presentation. Histopathology and immunohistochemistry serve as the main diagnostic tools in such cases. They have a very low malignant potential but can frequently recur. However, there are limited guidelines with regard to the choice of treatment for these tumors. This case scenario pertains to a 16-year-old female patient who had gross hematuria and fever, devoid of additional symptoms. Further evaluations uncovered a neoplas

EZH2-mediated macrophage-to-myofibroblast transition contributes to calcium oxalate crystal-induced kidney fibrosis.

Long-term nephrocalcinosis leads to kidney injury, fibrosis, and even chronic kidney disease (CKD). Macrophage-to-myofibroblast transition (MMT) has been identified as a new mechanism in CKD, however, the effect of MMT in calcium oxalate (CaOx)-induced kidney fibrosis remains unclear. In this study, abundant MMT cells are identified by immunofluorescence (IF) and flow cytometry in kidney tissues of patients with CaOx-related CKD, a male mouse model, and CaOx-treated macrophages. Clodronate liposome (CLO)-mediated macrophage depletion attenuates fibrosis in male nephrocalcinosis mice. Transcrip

Reduced expression of SMAD7 and consequent reduction of autophagy promotes endometrial stromal-myofibroblast transition and fibrosis.

Abnormal autophagy and the transforming growth factor-β (TGFβ)-SMAD3/7 signaling pathway play an important role in the development of intrauterine adhesions (IUAs); however, the exact underlying mechanisms remain unclear. In this study, we used IUA patient tissue and SMAD7 conditional knockout mice to detect whether SMAD7 effected IUA via regulation of autophagy and the TGFβ-SMAD3 signaling pathway. We applied a combination of techniques for the detection of p-SMAD3, SMAD7, autophagy and fibrosis-related proteins, autophagic flux, and analysis of the SMAD3 binding site. Endometrial tissue of p
