A finer examination of the role that negative affect plays in the relationship between paternal alcoholism and the onset of alcohol and marijuana use.

OBJECTIVE: This study was designed to separately examine the relations between four fundamental components of negative affect (sadness, fear, guilt, and hostility) and the onset of substance use. An additional goal was to examine the potential mediating roles that these components of negative affect play in the relationship between paternal alcoholism and the initiation of substance use. METHOD: The sample included 200 adolescents ages 15-19 years (62% girls; 68% white; 56% children of alcoholics [COAs]) and their fathers. The adolescents were followed up 5 years later when they were in early

Severe presentation of Beckwith-Wiedemann syndrome associated with high levels of constitutional paternal uniparental disomy for chromosome 11p15.

Beckwith-Wiedemann syndrome (BWS) is an overgrowth syndrome characterized by macrosomia, macroglossia, omphalocele, hemihyperplasia, and increased tumor risk. BWS can be associated with genetic and/or epigenetic alterations that modify imprinted gene expression on chromosome 11p15.5. Somatic mosaicism for paternal uniparental disomy (UPD) of chromosome 11p15, found in 20% of BWS patients, is associated with specific features of BWS including hemihyperplasia, Wilms tumor, and hepatoblastoma. The highly variable phenotypic spectrum of BWS associated with UPD may well reflect the level of UPD 11

Complementary roles of genes regulated by two paternally methylated imprinted regions on chromosomes 7 and 12 in mouse placentation.

Imprinted genes have prominent effects on placentation; however, there is limited knowledge about the manner in which the genes controlled by two paternally methylated regions on chromosomes 7 and 12 contribute to placentation. In order to clarify the functions of these genes in mouse placentation, we examined transcription levels of the paternally methylated genes, tissue differentiation and development and the circulatory system in placentae derived from three types of bi-maternal conceptuses that contained genomes of non-growing (ng) and fully grown (fg) oocytes. The genetic backgrounds of

Relationships of maternal and paternal birthweights to features of the metabolic syndrome in adult offspring: an inter-generational study in South India.

AIMS/HYPOTHESIS: The association between lower birthweight and metabolic syndrome may result from fetal undernutrition (fetal programming hypothesis) and/or genes causing both low birthweight and insulin resistance (fetal insulin hypothesis). We studied associations between the birthweight of parents and metabolic syndrome in the offspring. METHODS: We identified men and women (aged 35-68 years), who had been born in Holdsworth Memorial Hospital, Mysore, India. We also identified the offspring (20-46 years) of these men and women. In total, 283 offspring of 193 mothers and 223 offspring of 14

Kinetics of the pregnancy-induced humoral and cellular immune response against the paternal HLA class I antigens of the child.

Pregnancy can prime the maternal humoral immune response against paternal human leukocyte antigens (HLA) of the child. Previous studies have reported that formation of antibodies against inherited paternal HLA is associated with the presence of primed cytotoxic T lymphocytes (CTLs) specific for these antigens. Recently, we reported that primed CTLs can persist for more than 10 years after pregnancy even if the antibodies have disappeared. In the present study we studied the kinetics of the pregnancy induced immune response of the T-cell and B-cell compartment. In 12 women, who had specific ant

Somatic segregation errors predominantly contribute to the gain or loss of a paternal chromosome leading to uniparental disomy for chromosome 15.

Paternal uniparental disomy (UPD) for chromosome 15 (UPD15), which is found in approximately 2% of Angelman syndrome (AS) patients, is much less frequent than maternal UPD15, which is found in 25% of Prader-Willi syndrome patients. Such a difference cannot be easily accounted for if 'gamete complementation' is the main mechanism leading to UPD. If we assume that non-disjunction of chromosome 15 in male meiosis is relatively rare, then the gain or loss of the paternal chromosome involved in paternal and maternal UPD15, respectively, may be more likely to result from a post-zygotic rather than a

A maternally methylated CpG island in KvLQT1 is associated with an antisense paternal transcript and loss of imprinting in Beckwith-Wiedemann syndrome.

Loss of imprinting at IGF2, generally through an H19-independent mechanism, is associated with a large percentage of patients with the overgrowth and cancer predisposition condition Beckwith-Wiedemann syndrome (BWS). Imprinting control elements are proposed to exist within the KvLQT1 locus, because multiple BWS-associated chromosome rearrangements disrupt this gene. We have identified an evolutionarily conserved, maternally methylated CpG island (KvDMR1) in an intron of the KvLQT1 gene. Among 12 cases of BWS with normal H19 methylation, 5 showed demethylation of KvDMR1 in fibroblast or lymphoc

Mutations in fibroblast growth-factor receptor 3 in sporadic cases of achondroplasia occur exclusively on the paternally derived chromosome.

More than 97% of achondroplasia cases are caused by one of two mutations (G1138A and G1138C) in the fibroblast growth factor receptor 3 (FGFR3) gene, which results in a specific amino acid substitution, G380R. Sporadic cases of achondroplasia have been associated with advanced paternal age, suggesting that these mutations occur preferentially during spermatogenesis. We have determined the parental origin of the achondroplasia mutation in 40 sporadic cases. Three distinct 1-bp polymorphisms were identified in the FGFR3 gene, within close proximity to the achondroplasia mutation site. Ninety-nin

Short-limb dwarfism and hypertrophic cardiomyopathy in a patient with paternal isodisomy 14: 45,XY,idic(14)(p11).

Uniparental disomy (UPD) has been shown to result in specific disorders either due to imprinting and/or homozygosity of mutant alleles. Here we present the findings in a child with paternal UPD14. Ultrasound evaluation was performed at 30 weeks of gestation because of abnormally large uterine size. Pertinent ultrasound findings included polyhydramnios, short limbs, abnormal position of hands, small thorax, and nonvisualization of the fetal stomach. Post-natally the infant was found to have a low birth weight, short birth length, contractures, short limbs, and a small thorax with upslanting rib

Decrease in the CGGn trinucleotide repeat mutation of the fragile X syndrome to normal size range during paternal transmission.

The fragile X syndrome, the most common inherited form of mental retardation, is caused by the expansion of a CGGn trinucleotide repeat in the FMR-1 gene. Although the repeat number usually increases during transmission, few cases with reduction of an expanded CGGn repeat back to the normal size range have been reported. We describe for the first time a family in which such reduction has occurred in the paternal transmission. The paternal premutation (delta = 300 bp) was not detected in one of the five daughters or in the son of this daughter, although he had the grandpaternal RFLP haplotype.

Preferential X inactivation in human placenta membranes: is the paternal X inactive in early embryonic development of female mammals?

In placenta membranes of newborn girls carrying electrophoretically distinguishable G6PD alleles, the maternally derived isozyme is expressed preferentially. This phenomenon cannot be explained by allelic differences in enzyme activity or by somatic selection directed against cells with particular G6PD phenotypes. Instead, it may be that in this tissue X inactivation is nonrandom. Preferential expression of the maternal X chromosome, as has been shown in marsupials and in extraembryonic membranes of rodents and now in man, may reflect the state of activity of the X chromosomes in the early sta

A Family With Discordant Germline BRCA1 and BRCA2 Variants: A BRCA1-Positive Mother and BRCA2-Positive Daughter With Possible Paternal Inheritance.

Hereditary breast and ovarian cancer (HBOC) is primarily caused by germline pathogenic variants in BRCA1 or BRCA2. Although a single pathogenic variant usually explains cancer predisposition within a family, independent pathogenic variants may coexist and complicate risk assessment and genetic counseling. We report a 43-year-old woman with right breast cancer in whom germline genetic testing identified a pathogenic BRCA2 variant, c.2808_2811del (p.Ala938ProfsTer21). Her mother developed primary peritoneal carcinoma at the age of 77. Tumor genomic testing identified a BRCA1 deletion of exons 1-

The Direction of Incongruence Matters: Low Maternal and High Paternal Marital Satisfaction Linked to Higher Depression in Emerging Adult Daughters.

With studies primarily focusing on the influence of individual parents' marital satisfaction, less is known about the combined influence of fathers and mothers. Using polynomial regressions and response surface analyses, this study aims to explore the congruence and incongruence between fathers' marital satisfaction and mothers' marital satisfaction on child depression by the child's developmental stage and gender. Participants were 902 Chinese one-child families (in terms of child gender, 41.7% were girls and 58.3% were boys; in terms of child age, 49.7% were emerging adults and 50.3% were mi

Integrating Osteology and Ancient DNA: Cranial Variation, Hemoglobin S, and Paternal Lineage in a Roman-Period Individual from Anatolia.

(1) Background: Integrated bioarchaeological approaches combining osteological and ancient DNA analyses provide powerful insights into health, disease, and population history in past societies. However, the relationship between rare skeletal variations, genetic disorders, and ancestry remains insufficiently explored within single individuals. This study aimed to investigate the combined osteological, paleopathological, and genetic characteristics of a Roman-period individual from southwestern Anatolia. (2) Methods: A multidisciplinary approach was applied to the skeletal remains of an adult ma

Impact of advanced paternal age on reproductive outcomes in preimplantation genetic testing cycles of young female: a retrospective cohort study.

INTRODUCTION: Although advanced paternal age (APA) is increasingly scrutinized in reproductive medicine, its independent impact on embryo development and clinical outcomes remains contentious, particularly when controlling for maternal age and embryo ploidy. METHODS: This retrospective cohort study analyzed 357 preimplantation genetic testing for aneuploidy (PGT-A) cycles from couples with non-advanced maternal age (≤35 years). Cycles were stratified by paternal age into three groups: <35, 35-39, and ≥40 years. We compared sperm DNA fragmentation index (DFI), embryo development metrics, and c

Protective Relationships, Adultification, and Sexual Risk Among Black Adolescent Girls: Examining Maternal, Paternal, Peer, and Community Influences.

The adultification of Black adolescent girls, viewing them as less innocent and more adult-like, has implications for their sexual and psychological well-being. Adultification creates bias, which hyper-sexualizes Black girls, increasing vulnerability to early sexual activity, sexual abuse, and lack of support, while hindering their ability to seek protection. However, protection from parents, peers, and communities may play a critical role in mitigating this risk for Black girls. The current study examined the role of maternal, paternal, peer friendships, and communal protective factors in med

Adverse childhood experiences and paternal mental health during the transition to fatherhood: Findings from a cohort study of Australian men.

BACKGROUND: Adverse childhood experiences (ACEs) are associated with poorer mental health across the life course. However, research examining ACEs during the transition to parenthood has largely focused on mothers, with limited evidence on fathers. OBJECTIVE: To examine associations between cumulative and specific ACEs and mental health and wellbeing among first-time fathers during the first year after their child's birth. PARTICIPANTS AND SETTING: Participants were 718 first-time fathers (Mage = 34.69 years; SD = 6.98 years) drawn from the Ten to Men Study, a population-representative cohor

Paternal age and adverse pregnancy outcomes: a spline analysis from the nulliparous pregnancy outcomes study: monitoring mothers-to-be study.

OBJECTIVES: Extremes of maternal age are associated with an increased risk of adverse pregnancy outcomes (APOs). Less is known about the independent effects of extremes of paternal age on reproductive risk. METHODS: We conducted a secondary analysis using data from the Nulliparous Pregnancy Outcomes Study: Monitoring Mothers-to-Be (nuMoM2b) using restricted cubic spline logistic regression to examine the non-linear association between paternal age and APOs and adverse fetal/neonatal outcomes. RESULTS: After adjusting for maternal health and sociodemographic and paternal sociodemographic conf
