Prevalence of anti-thyroid peroxidase antibodies in serum in the elderly: comparison with other tests for anti-thyroid antibodies.

Autoimmune thyroid disease, especially chronic thyroiditis, is prevalent in elderly women and is probably the major cause of hypothyroidism in this population. The reported prevalence of chronic thyroiditis is variable, depending on the area of residence and the method(s) used to detect the presence of anti-thyroid antibodies. The recent finding that thyroid peroxidase (TPO) is the antigen for the thyroid anti-microsomal antibody (AbM) has resulted in the development of sensitive radioimmunoassays (RIA) to detect the presence of AbTPO. We have determined the prevalence of AbTPO (by RIA) in ser…

The trigeminal sensory innervation to the middle ear, eustachian tube, and pharynx: a study by the horseradish peroxidase tracer method.

The trigeminal sensory innervations to the middle ear, the eustachian tube, and the pharynx were examined using the horseradish peroxidase (HRP) method in cats. After HRP application to the middle ear and the eustachian tube, HRP-labeled cells were found mainly in the posterolateral portion and the anteromedial portion of the trigeminal ganglion, respectively. In the case of injection into the epipharynx and the mesopharynx, HRP-labeled cells were found in the anteromedial portion and the posterolateral portion, respectively. HRP-labeled cells were hardly observed when HRP was applied to the h…

Abnormalities of blood selenium and glutathione peroxidase activity in patients with acquired immunodeficiency syndrome and aids-related complex.

Severe protein-calorie malnutrition is common in patients with AIDS and could contribute to the progressive deterioration characteristic of that disease. Selenium deficiency could also have a negative impact on immune function and other organ functions vital for recovery from infectious diseases. Therefore, to assess any role for selenium in AIDS we determined plasma and erythrocyte selenium levels and glutathione peroxidase activity in 13 patients with AIDS compared to 8 patients with AIDS-related complex (ARC) and 14 healthy controls. Plasma selenium levels were significantly reduced in AIDS…

Genetic mapping of katG, a locus that affects synthesis of the bifunctional catalase-peroxidase hydroperoxidase I in Escherichia coli.

A locus unlinked to either katE or katF that affected catalase levels in Escherichia coli was identified and localized between metB and ppc at 89.2 min on the genome. The locus was named katG. Mutations in katG which prevented the formation of both isoenzyme forms of the bifunctional catalase-peroxidase HPI were created both by nitrosoguanidine and by transposon Tn10 insertions. All katG+ recombinants and transductants contained both HPI isoenzymes. Despite the common feature of little or no catalase activity in four of the catalase-deficient strains, subtle differences in the phenotypes of ea…

Concanavalin A-peroxidase labeling in cervical exfoliative cytopathology. I. Labeling of normal squamous cells and the detection of cancer.

The lectin binding capacity of the cell surface of normal flattened exfoliated epithelial cells of the uterine cervix was investigated looking for differences between specimens from normal and cancer patients. The method used was a modified concanavalin A-horseradish peroxidase (Con A-HRP) labeling procedure. Both normal and cancer specimens contain labeled as well as unlabeled usual flattened cells. There is a distinct difference between the labeling intensity of labeled and that of unlabeled cells. Quantification of the labeling results has been achieved using a light microscope equipped wit…

Projections from the Edinger-Westphal complex of monkeys as studied by means of retrograde axonal transport of horseradish peroxidase.

By means of retrograde axonal transport of horseradish peroxidase, an experimental study was made in the Japanese monkey of the projection from the anteromedian nucleus (AM) and Edinger-Westphal nucleus (EW) to the ciliary ganglon, cerebellar nuclei and spinal cord. Special attention was paid on the labeled cell forms. Neurons projecting to the cerebellar nuclei or the spinal cord were small (8-11 microns in diameter) and spindle-shaped, and they were located in almost the entire parts of both AM and EW. On the other hand, cells which gave off their axons to the ciliary ganglion were large (25…

The organization of the thoracic spinal nerve projection in the rat dorsal horn demonstrated with transganglionic transport of horseradish peroxidase.

Transganglionic transport of horseradish peroxidase (HRP) was used to study the organization of the thoracic spinal nerve projection to the dorsal horn in rats. Labeling was found in the superficial dorsal horn 16-20 hours after application of HRP to the cut ends of various spinal nerve rami. Labeling was restricted to the outer part of the substantia gelatinosa at these stages. Longer survivals (25-48 hours) gave rise to labeling of the deep part of substantia gelatinosa and deeper parts of the dorsal horn as well. The dorsal ramus projected to the lateral third of the horn from half a segmen…

The reticulocerebellar projection to the pyramis copula pyramidis in the rat: an experimental study using retrograde transport of horseradish peroxidase.

The organization of the lateral reticulocerebellar projection to the pyramis and copula pyramidis in the rat was studied with the use of microinjections of horseradish peroxidase (HRP). These injections were made in different mediolateral positions along the width of this lobule. The objective was to determine whether reticulocerebellar parasagittal zones (Chan-Palay et al., '77) receive differential inputs from the lateral reticular nucleus (LRN). This material has previously been used to analyze the olivo and pontocerebellar projection to this same cortical region of the rat cerebellum (Eise…

The distribution of the cat's carotid sinus nerve afferent and efferent cell bodies using the horseradish peroxidase technique.

The location of both afferent and efferent carotid sinus nerve (CSN) cell bodies in the cat has been determined using the horseradish peroxidase (HRP) technique. Following a limited exposure of the central cut end of the CSN to HRP, labeled sensory ganglion cells were found in both the petrosal and superior ganglia of the IXth cranial nerve. An average of 387 in the former and 16 cells in the latter ganglion were labeled. Retrogradely labeled neurons were found only within the ipsilateral medulla. These cells were both round and spindle shaped and had an average somal diameter of 19 micrometer…

The olivocerebellar projection in the cat as studied with the method of retrograde axonal transport of horseradish peroxidase. II. The projection to the uvula.

Following injections of small quantities of horseradish peroxidase (HRP) suspension in the uvula of the cat, the distribution of labeled cells in the inferior olive has been mapped. The findings confirm the conclusion made on the basis of studies of retrograde cell loss in the olive following ablations of the uvula (Brodal, '40b) that two small olivary subdivisions, the nucleus beta and the dorsomedial cell column project heavily to the uvula. In addition the HRP-study shows that the uvula receives a smaller number of fibers from two circumscribed areas of the contralateral medial accessory ol…

Human saliva peroxidase: microanalytical isoelectric fractionation and properties in normal persons and in cases with neuronal ceroid-lipofuscinosis.

Human saliva contains a high peroxidase activity that can be estimated spectrophotometrically with the hydrogen donor p-phenylenediamine and the substrate hydrogen peroxide from 20 mul of material. The pH optimum of the enzyme with citrate-phosphate buffer is 5.5. After microanalytical isoelectric fractionation 3 main isoenzyme components at pI 8.6, 6.5 and 4.3, and a number of isoenzyme subfractions at pI 9.5, 7.3 and 3.8 are detectable. In 3 patients with the juvenile form of neuronal ceroid-lipofuscinosis (type Spielmeyer-Vogt), in which a deficiency of leukocyte peroxidase had been reporte…

Integrated genomic, transcriptomic, and proteomic analyses prioritize cadmium-responsive class III peroxidases in Salix variegata.

Class III peroxidases (PRXs) link apoplastic redox regulation with cell-wall remodeling, yet their coordinated transcript- and protein-level responses to cadmium (Cd) remain poorly resolved in woody plants. We identified 80 high-confidence class III PRX genes in Salix variegata and characterized their genomic organization, evolutionary relationships, protein properties, and predicted regulatory features. Root RNA-seq and data-independent acquisition proteomic datasets from plants exposed to 0 or 100 μM CdCl2 for 10 d were then integrated to prioritize members supported across molecular layers.…

A geometry-guided computational framework for predicting functional immobilization orientations of plant peroxidases on chemically modified surfaces.

Enzyme immobilization depends not only on support chemistry, but also on the spatial orientation adopted by the protein after attachment. This study presents a computational framework guided by molecular geometry to screen favorable immobilization orientations of plant peroxidases on modified surfaces. Horseradish peroxidase, soybean peroxidase, and royal palm tree peroxidase were analyzed as three distinct case studies. Surface exposed anchoring residues were identified from experimental structures, and rigid body orientation sampling generated enzyme and surface configurations. Each configur…

Opposing effects of PM2.5 on TSH levels: effect modification by thyroid peroxidase antibody status in a preconception cohort.

BACKGROUND: Fine particulate matter (PM2.5) is a significant source of endocrine disrupting chemicals. However, epidemiological evidence linking PM2.5 exposure to thyroid function, particularly thyroid-stimulating hormone (TSH), remains inconsistent. We hypothesized that this inconsistency stems from unmeasured effect modification by thyroid autoimmunity status, specifically the presence of thyroid peroxidase antibodies (TPOAb). OBJECTIVE: To examine whether TPOAb status modifies the association between PM2.5 exposure and thyroid function in women planning pregnancy. METHODS: This cohort stu…

The clinical characteristics and treatment response of myasthenia gravis with positive thyroid peroxidase antibody or thyroid globulin antibody.

BACKGROUND AND PURPOSE: Myasthenia gravis (MG) frequently associates with thyroid peroxidase antibody (TPOAb) and thyroglobulin antibody (TgAb), indicating shared autoimmunity. This study investigated the impact of TPOAb/TgAb on MG severity and pathogenesis, to inform improved management. METHODS: We retrospectively analyzed 144 MG patients (Jan 2022-Dec 2024), stratified into four groups by TPOAb/TgAb status. We compared demographic and clinical profiles, analyzed the effects of disease duration, stage, and onset age on TPOAb/TgAb titers, and evaluated treatment responses across groups. RES…

Assessment of the participation of sulfhydryl proteins in the glutathione peroxidase mimicry of diphenyl diselenide in the presence of thiol alkylating agent.

Diphenyl diselenide (DPDS) is an organoselenium which has garnered profound interest due to its reported antioxidant viz-a-viz glutathione peroxidase (GPx) mimetic activity which is ultimately speculated to rely on oxidation of free thiols or protein thiols critical to the activities of some sulfhydryl proteins or enzymes. However, this hypothesis has not been fully established. This study was therefore carried out to investigate the antioxidant effect as well the possible involvement of proteins thiols in the in vitro GPx mimicry of DPDS. The effect of DPDS on Fe2+ (10 µM) and H2O2 (1mM)-indu…

Biosynthesis of nitric oxide in plants: An oxidative pathway orchestrated by the interplay of CYP79s, flavin-containing monooxygenases, and peroxidases.

Nitric oxide (NO) is in the pantheon of plant signal molecules and hormones controlling plant growth, development, and adaptation to environmental challenges. The route of NO biosynthesis in plants has remained enigmatic. Previous studies have shown the ability of peroxidases to utilize oximes for the production of NO. Peroxidases are widely spread and highly expressed in plant tissues. What then is the identity of the pathway signature enzymes determining tight, spatiotemporal regulation of NO production to effectuate its specific signal functions? What are the key selection criteria to be fu…
