DNA barcoding of sand flies from Chandipura-affected districts of Gujarat, India.

Phlebotomine sand flies are important vectors of various pathogens, including the Chandipura virus (CHPV), a neurotropic rhabdovirus associated with fatal pediatric encephalitis outbreaks in India. Gujarat, a CHPV-endemic region, lacks comprehensive molecular data on sand fly species. Accurate species identification is important for effective vector surveillance and control strategies. 2950 sand flies were collected from 14 districts using aspirators and traps. Morphological identification was performed by using standard taxonomic keys. DNA barcoding of the mitochondrial cytochrome c oxidase s

Epidemiological landscape in a Mediterranean hotspot of human leishmaniosis in Spain under a One Health approach.

Integrated One Health studies are essential to assess the potential risks associated with leishmaniosis hotspots in Europe. Thus, the aim was to holistically evaluate Leishmania infantum epidemiology in a rural hotspot in Spain with a high incidence of human leishmaniosis. Samples from 145 humans (blood), 41 dogs (blood and hairs), and 41 wild lagomorphs (blood, skin, and spleen) were collected during 2022-2023. Sandflies were captured with CDC-traps, and blood-feeding was evaluated. L. infantum exposure was assessed using indirect immunofluorescence and/or quantitative PCR. Positivity was det

An update on Leishmania martiniquensis infections: Transmission, clinical characteristics, and treatment.

Leishmaniasis, caused by intracellular protozoa of the Leishmania genus, continues to be a global health issue, with approximately 700,000 to 1 million new cases occur annually worldwide. The disease is transmitted via the bite of infected female sand flies of the genus Phlebotomus, resulting in a range of symptoms known as cutaneous, mucocutaneous, and visceral leishmaniasis. The species Leishmania (Mundinia) martiniquensis, discovered in 1995, has been linked to cases in individuals with HIV, presenting with diverse clinical pictures. Interestingly, biting midges, not sandflies, has proved t

Diversity, Abundance and Leishmania infantum Infection Rate of Phlebotomine Sandflies in an Area with Low Incidence of Visceral Leishmaniasis in Northern Tunisia.

We report the study of sandfly Leishmania infection in an area of low incidence of visceral leishmaniasis in Tunisia. Sandflies were collected monthly using CDC light-traps set in houses and animal shelters during May-November 2016 and 2017. All males were identified at the species level. A sample of 878 females including all gravid specimens was subjected to kDNA qPCR for Leishmania detection and parasite load estimation. Leishmania species were determined by ITS1 PCR sequencing, and species identification of infected sandflies was performed by DNA barcoding. Phlebotomus perfiliewi and P. per

Comparative study of kala-azar vector control measures in eastern Nepal.

This study was conducted to explore the most effective vector control tool among indoor residual spraying (IRS), long lasting insecticidal nets (LLINs) and ecological vector management (EVM) as a part of the regional visceral leishmaniasis elimination initiative. Alpha-cypermethrin as IRS, PermaNet as LLINs and plastering the inner walls of houses with lime as EVM were the interventions. One baseline and three follow-up entomological surveys were carried out in all arms using CDC miniature light traps (LT) and mouth aspirators. Comparisons were made between intervention arms and control arms w

Vector-borne diseases and climate change: a European perspective.

Climate change has already impacted the transmission of a wide range of vector-borne diseases in Europe, and it will continue to do so in the coming decades. Climate change has been implicated in the observed shift of ticks to elevated altitudes and latitudes, notably including the Ixodes ricinus tick species that is a vector for Lyme borreliosis and tick-borne encephalitis. Climate change is also thought to have been a factor in the expansion of other important disease vectors in Europe: Aedes albopictus (the Asian tiger mosquito), which transmits diseases such as Zika, dengue and chikungunya

Bioactivity guided fractionation of Moringa oleifera Lam. flower targeting Leishmania donovani.

Leishmaniases is a group of diseases caused by the protozoan parasite belonging to the genus Leishmania. At least 20 species of Leishmania are known to infect humans transmitted by female sandflies, Phlebotomus spp. Leishmania donovani causes visceral leishmaniasis, considered most lethal among the common three forms of leishmaniasis. Lack of appropriate vaccines, emergence of drug resistance and side effects of currently used drugs stress the need for better alternative drugs, particularly from natural sources. Here, we conducted in vitro and in vivo experiments to study the efficacy of diffe

Leishmaniasis in Malta and the Mediterranean basin.

Leishmaniasis is a public-health problem in most countries bordering the Mediterranean littoral. In Malta, where the disease has been recognized for many years, Phlebotomus perniciosus is the established vector and dogs act as reservoir hosts. Visceral leishmaniasis (VL) was the only form of the disease recorded in Malta until the early 1980s, when cutaneous leishmaniasis (CL) was recognized. Although the incidence of CL has recently increased, the overall numbers of cases of leishmaniasis have markedly decreased since the 1960s. Prior to 1963, almost all cases were aged < 10 years. Although l

A Bayesian modeling and retrospective analysis of cutaneous leishmaniasis in the Sahara Desert.

Cutaneous leishmaniasis (CL) is one of the most widespread neglected tropical diseases, transmitted by Phlebotomus sandflies and caused by protozoan parasites of the genus Leishmania. It poses a major global public health concern, particularly in arid and semi-arid regions where ecological and socio-environmental factors favor vector proliferation. In Algeria, CL is endemic across several regions, yet few studies have explored its long-term epidemiological dynamics in the Sahara Desert, where the disease continues to expand. This study aimed to investigate the spatio-temporal evolution and epi

Canine leishmaniosis in a low-but-non-zero risk setting: A practice-oriented review from Hungary.

Canine leishmaniosis (CanL) caused by Leishmania infantum is traditionally endemic in the Mediterranean basin; however, imported infections and sporadic autochthonous cases are increasingly often reported in Central Europe. Recent field surveys and curated vector-distribution datasets indicate that several phlebotomine sand fly species occur in the Carpathian Basin and climate-based suitability models suggest that ecologically favourable microhabitats may be present beyond the historical endemic zone. In this narrative, practice-oriented review, we synthesise literature published between 2004

Real-time PCR using high-resolution melting analysis technology for diagnosis of Leishmania and determination of types of clinical samples.

BACKGROUND/AIM: Leishmaniasis is a disease group carried by Phlebotomus and Lutzomyia sand flies infected with Leishmania and is mostly observed in rural areas. In this study, using high-resolution melting analysis (HRMA), we aim to identify the active types of leishmaniasis, which are inadequately identified by classical methods. MATERIALS AND METHODS: Samples of 85 patients were examined in the study. Six of the patients were suspected of having visceral leishmaniasis (VL) and 79 cutaneous leishmaniasis (CL). The slides prepared from the samples were stained with Giemsa stain and examined u

Prevalence of canine leishmaniasis in Beichuan County, Sichuan, China and phylogenetic evidence for an undescribed Leishmania sp. in China based on 7SL RNA.

BACKGROUND: Leishmaniasis is a vector-borne disease, which is still endemic in the west and northwest area of China. Canines are the major reservoirs of Leishmania, the etiological agent of human visceral leishmaniasis. Phlebotomus chinensis is the main transmission vector of zoonotic visceral leishmaniasis (ZVL). METHODS: In this study, rK39 dip-stick, ELISA and PCR methods were used to investigate the prevalence of canine leishmaniasis (CanL) in Beichuan County, Sichuan Province, China. RESULTS: Among the 86 dogs which were included in the study, 13 dogs were positive using the dip-stick t

Epidemiology of paediatric visceral leishmaniasis in Hebron district, Palestine.

Seventy-six cases of visceral leishmaniasis (VL) were reported from the Hebron district of the West Bank, Palestine between 1993 and 2007. All cases were in children less than 9 years old (median age 2 years). The average number of cases was 5.06/year and the average annual incidence was 3.02/100000 children. Ribosomal internal transcribed spacer 1 (ITS1) PCR-RFLP was performed using DNA extracted from two cultures and 36 archived Giemsa-stained slides from VL patients. Leishmania infantum was revealed as the causative agent of VL in the focus. Isoenzyme analysis identified two isolates as zym

Global Distribution of Canine Visceral Leishmaniasis and the Role of the Dog in the Epidemiology of the Disease.

Visceral leishmaniasis is a disease caused by protozoa of the species Leishmania (Leishmania) infantum (syn = Leishmania chagasi) and Leishmania (Leishmania) donovani, which are transmitted by hematophagous insects of the genera Lutzomyia and Phlebotomus. The domestic dog (Canis familiaris) is considered the main urban reservoir of the parasite due to the high parasite load on its skin, serving as a source of infection for sandfly vectors and, consequently, perpetuating the disease in the urban environment. Some factors are considered important in the perpetuation and spread of canine visceral

Cutaneous leishmaniasis is one of the most commonly diagnosed dermatological condition in travel medicine after diarrhoeal diseases and febrile status. The disease is transmitted by Phlebotomus and Lutzomyia sandflies. It appears in various clinical forms, the most common of which is a painless ulcer with raised edges, usually present on exposed parts of the body on the side where the insect bite occurred. Annually, over a million new cutaneous leishmaniasis (CL) cases are reported globally. We present two cases of affliction, the first occurring in Patient 1, who attempted treatment through t

Towards a Sustainable Vector-Control Strategy in the Post Kala-Azar Elimination Era.

Visceral leishmaniasis (VL) is a potentially deadly parasitic disease. In the Indian sub-continent, VL is caused by Leishmania donovani and transmitted via the bite of an infected Phlebotomus argentipes female sand fly, the only competent vector species in the region. The highest disease burden is in the northern part of the Indian sub-continent, especially in the state of Bihar. India, Bangladesh, and Nepal embarked on an initiative, coordinated by World Health Organization, to eliminate VL as a public health problem by the year 2020. The main goal is to reduce VL incidence below one case per
