Progesterone (P) facilitation of sexual receptivity in rodents has been achieved by intracranial administration to the ventral hypothalamus; the preoptic area; and midbrain areas such as central gray, mesencephalic reticular formation, and ventral tegmental nucleus. In our laboratory, by far the most effective site in rats has been the ventromedial nucleus of the hypothalamus (VMN). However, several reports of sensitivity to P in the midbrain of rats and other rodent species led us to investigate whether stimulation of the ventral midbrain of female rats might contribute to facilitation of sex…

Since cryptococcosis is characterized by cryptococcoma formation, the antimycotic effect of amphothericin B was examined in view of such pathological-anatomical conditions. In white mice (NMRI), cryptococcoma formation was induced by intramuscular injection of Cryptococcus neoformans strain W71 into the hind leg (STAIB, 1962), using a suspension (0.2 ml) containing approximately 2.8 times 10-7 cells/ml. The mice were treated daily with 1 mg amphotericin B in 5% dimethyl sulfoxide by gastric intubation. Course of infection and effectivity of therapy were assessed by microbiological and patho-hi

Prolactin-releasing peptide (PrRP), originally isolated from the hypothalamus, is highly localized in the cardiovascular regions of the medulla, and intracerebroventricular administration of PrRP causes a pressor response. In the present study we investigated the cardiovascular effects of PrRP applied to functionally different areas of the ventrolateral medulla (VLM), and to the nucleus tractus solitarius (NTS) and the area postrema (AP). In urethane-anesthetized rats, microinjection of PrRP into the pressor area of the most caudal VLM, recognized as the caudal pressor area in the rat, elicite

Rett syndrome is a neurodevelopmental disorder caused by mutations in the transcriptional repressor methyl-CpG-binding protein 2 (MeCP2) and represents the leading genetic cause for mental retardation in girls. MeCP2-mutant mice have been generated to study the molecular mechanisms of the disease. It was suggested that an imbalance between excitatory and inhibitory neurotransmission is responsible for the behavioral abnormalities, although it remained largely unclear which synaptic components are affected and how cellular impairments relate to the time course of the disease. Here, we report th

General pharmacological effects of [Ethyl p-(6-guanidinohexanoyloxy)benzoate] methanesulfonate (FOY), a new antiproteolytic agent, were studied and the following results were obtained. Acute administration of large doses of FOY in conscious dogs and rabbits caused a decrease in spontaneous motility, ataxia, cyanosis, collapse, mydriasis, and respiratory paralysis. The agent had no effect on the central nervous system and exhibited hypotensive effects in dogs in doses of more than 1 mg/kg. Hypotensive responses were not inhibited by treatment with atropine or hexamethonium. FOY had no effects o

PURPOSE: Sulfur mustard (SM) is a highly reactive chemical warfare agent that causes severe cutaneous injury, including blistering, necrosis, inflammation, and delayed wound repair. This exploratory study characterized cellular and molecular changes associated with SMinduced skin injury and repair over 60 days.
MATERIALS AND METHODS: Göttingen minipigs received a single SM exposure using a vapor cap model. Skin samples were analyzed for changes in epidermal architecture, proliferation, adhesion, differentiation, and dermal remodeling using histopathology and immunochemistry. Markers included

Deoxynivalenol (DON), a Fusarium-derived mycotoxin, disrupts immune homeostasis and induces immunotoxicity. Macrophage extracellular traps (METs) are key effectors in innate immunity and inflammation, yet whether DON triggers METs formation and the underlying mechanisms remain unclear. This study aims to investigate the molecular mechanism by which DON induces METs generation through circadian rhythm immune checkpoints involving the PER2-c-Myc axis. Using RAW264.7 and THP-1 macrophage models, we combined siRNA-mediated Per2 knockdown, and pharmacological inhibitors to dissect the mechanisms of

Tertiary lymphoid structures (TLS) are associated with inflammatory kidney diseases, but their pathogenesis is unclear. A chronic high phosphate diet (HPD) in mice increases serum levels of phosphate and fibroblast growth factor 23 (FGF23) which is associated with progressive tubule damage and fibrosis. We hypothesized that chronic HPD induces TLS in the kidneys and thereby promotes progressive kidney injury. After only 2 months, mice on HPD showed increased tubule damage accompanied by the formation of perivascular immune cell clusters of B and T cells in the corticomedullary zone and in the

BACKGROUND: Kindler syndrome is a rare, inherited skin disease characterized by acral bullae formation, fusion of fingers and toes, and generalized progressive poikiloderma. The purpose of this study was to clarify the nature of the bullous component of Kindler syndrome and to determine whether this inherited skin disorder represents a variant of dystrophic epidermolysis bullosa or a unique independent clinical entity.
OBSERVATIONS: Two unrelated patients with Kindler syndrome were studied. Electron microscopy demonstrated marked duplication of the lamina densa, and clefts were observed in ar
