BMP-2 restoration aids in recovery from liver fibrosis by attenuating TGF-β1 signaling.

Transforming growth factor-β (TGF-β) plays a central role in hepatic fibrogenesis. This study investigated the function and mechanism of bone morphogenetic protein-2 (BMP-2) in regulation of hepatic fibrogenesis. BMP-2 expression in fibrotic liver was measured in human tissue microarray and mouse models of liver fibrosis induced by bile duct ligation surgery or carbon tetrachloride administration. Adenovirus-mediated BMP-2 gene delivery was used to test the prophylactic effect on liver fibrosis. Primary hepatic stellate cells (HSC), HSC-T6 and clone-9 cell lines were used to study the interpla

Effect of Compound 21, a Selective Angiotensin II Type 2 Receptor Agonist, in a Murine Xenograft Model of Dupuytren Disease.

BACKGROUND: Although surgical excision and intralesional collagenase injection are mainstays in Dupuytren disease treatment, no effective medical therapy exists for recurrent disease. Compound 21, a selective agonist of the angiotensin II type 2 receptor, has been shown to protect against fibrosis in models of myocardial infarction and stroke. The authors investigated the potential use of compound 21 in the treatment of Dupuytren disease. METHODS: Human dermal fibroblasts were treated in vitro with compound 21 and assessed for viability using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazo

RET inhibition by selpercatinib suppresses TGF-β1-induced epithelial-mesenchymal transition in retinal pigment epithelium.

PURPOSE: The transforming growth factor-β (TGF-β) pathway is a master regulator of epithelial-mesenchymal transition (EMT) in retinal pigment epithelial (RPE) cells, a key process in fibrotic ocular diseases. However, key modulators fine-tuning this pathway in RPE are incompletely characterized. This study investigates the role of the receptor tyrosine kinase RET in TGF-β1-induced RPE-EMT. METHODS: The role of RET was assessed in vitro using the clinically approved RET inhibitor, Selpercatinib, in RPE cells stimulated with TGF-β1. Outcomes included EMT markers, migration, proliferation, and t

Diosgenin enhances transplanted human oligodendrocyte precursor cell-mediated myelination in preterm white matter injury by inhibiting matrix metalloproteinase-9-driven TGF-β/Smad2/3 signaling.

Preterm white matter injury (PWMI) is a major cause of long-term motor and cognitive disability, and there are no therapies that directly promote oligodendrocyte-lineage maturation. Here, we identify the natural steroid sapogenin diosgenin as a potent pro-myelinating modulator for transplanted human oligodendrocyte precursor cells (hOPCs) using a hypoxia-ischemia mouse model of PWMI, together with in vitro microglia-OPC co-culture systems. Diosgenin crosses the blood-brain barrier and promotes the differentiation of both transplanted hOPCs and endogenous OPCs into myelinating oligodendrocytes,

miR-27b-3p reduces muscle fibrosis during chronic skeletal muscle injury by targeting TGF-βR1/Smad pathway.

BACKGROUND: Fibrosis is a significant pathological feature of chronic skeletal muscle injury, profoundly affecting muscle regeneration. Fibro-adipogenic progenitors (FAPs) have the ability to differentiate into myofibroblasts, acting as a primary source of extracellular matrix (ECM). the process by which FAPs differentiate into myofibroblasts during chronic skeletal muscle injury remains inadequately explored. METHOD: mouse model with sciatic nerve denervated was constructed and miRNA expression profiles between the mouse model and uninjured mouse were analyzed. qRT/PCR and immunofluorescence

Hypoxia-induced retinal pigment epithelium cell-derived bFGF promotes the migration and angiogenesis of HUVECs through regulating TGF-β1/smad2/3 pathway.

Hypoxia promotes the secretion of basic fibroblast growth factor (bFGF) in retinal pigment epithelium (RPE), which plays an important part in retinopathy of prematurity (ROP). This study preliminarily explored the effect of hypoxia-induced RPE-derived bFGF on the biological functions of human umbilical vein endothelial cells (HUVECs). After cell culture in hypoxia conditions, the cell viability, apoptosis, and the expressions of bFGF and vascular endothelial growth factor A (VEGFA) in human RPEs were detected by 3-(4, 5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), flow cytometr

Wumei Wan restores the colonic mucus barrier by modulating TGF-β/Smad signaling to promote goblet cell differentiation in ulcerative colitis.

ETHNOPHARMACOLOGICAL RELEVANCE: Wumei Wan (WMW), a traditional Chinese medicine formula first documented in Treatise on Febrile Diseases (Eastern Han Dynasty), was originally indicated for chronic diarrhea or dysentery. Contemporary pharmacological studies have further confirmed its definite efficacy against ulcerative colitis (UC); however, the underlying molecular mechanisms remain elusive. AIM OF THE STUDY: Anti-inflammatory strategies fail to disrupt the "injury-immune activation-re-injury" vicious cycle in UC, rendering restoration of the epithelial barrier a promising therapeutic direct

Relationship between asporin expression and the pathological progression of murine temporomandibular joint osteoarthritis after partial discectomy.

OBJECTIVES: Temporomandibular joint osteoarthritis (TMJ-OA) involves cartilage degeneration and bone changes; however, the mechanisms of onset and progression are unclear. Recently, it has been shown that asporin, an extracellular matrix protein, increases as knee osteoarthritis progresses and suppresses the TGF-β pathway. However, the asporin levels in later stages of TMJ-OA, when cartilage destruction occurs, are not known. In this study, we examined the relationship between asporin and the pathological progression of TMJ-OA. METHODS: We constructed TMJ-OA models in 12-week-old male C57BL/6

Escherichia coli promotes myofibroblast differentiation in urethral fibrosis urethral stricture in vivo and vitro via regulating TGFβ/Smad signaling pathway.

OBJECTIVE: Urethral stricture in children remains a challenging condition, with a high rate of postoperative recurrence. Previous studies employing 16 S rRNA sequencing and transcriptomic analyses have identified a positive association between urethral stricture, Escherichia coli (E. coli), and activation of the transforming growth factor-β (TGF-β)/Smad signaling pathway. Notably, even E. coli strains that do not cause overt urinary tract infection symptoms may contribute to fibrotic stricture formation. METHODS: In this study, we employed both in vitro cell culture and in vivo animal models

Aerobic Exercise Upregulates Renal-Derived ELABELA to Improve Myocardial Fibrosis by Activating APJ-AMPK-Sirt1 and Inhibiting TGFβ1-Smad2/3 Signaling Pathways in Myocardial Infarction Mice.

ELABELA (ELA) has been identified as a potential cardiovascular protective factor. However, the source of endogenous ELA and its molecular mechanism in myocardial fibrosis inhibition remain incompletely understood. Herein, we found that aerobic exercise significantly improved renal apoptosis caused by MI, inhibited inflammation, attenuated structural damage, enhanced renal function, and increased expression and secretion levels of renal ELA. Aerobic exercise stimulated the circulation of renal-derived ELA to reach the MI heart and played a protective role. Under aerobic exercise intervention,

Discovery of 2, 4-disubstituted pyrimidine derivatives as selective HIPK2 inhibitors for potential treatment of chronic kidney disease.

Chronic kidney disease (CKD) remains a major global health burden, and renal fibrosis is a key pathological driver of CKD progression. Homeodomain-interacting protein kinase 2 (HIPK2) has emerged as a potential therapeutic target because of its involvement in TGF-β/Smad3-mediated fibrotic signaling and NF-κB-driven inflammatory responses. In this study, a series of 2, 4-disubstituted pyrimidine derivatives were designed and synthesized as HIPK2 inhibitors. Among them, A6 (AF-1) and B11 (AF-2) exhibited potent HIPK2 inhibitory activity, with IC50 values of 0.36 and 0.56 μM, respectively, togeth

Pharmacologic Inhibition of Histone Deacetylase 6 Prevents the Progression of Chlorhexidine Gluconate-Induced Peritoneal Fibrosis by Blockade of M2 Macrophage Polarization.

Peritoneal fibrosis contributes to ultrafiltration failure in peritoneal dialysis (PD) patients and thus restricts the wide application of PD in clinic. Recently we have demonstrated that histone deacetylase 6 (HDAC6) is critically implicated in high glucose peritoneal dialysis fluid (HG-PDF) induced peritoneal fibrosis, however, the precise mechanisms of HDAC6 in peritoneal fibrosis have not been elucidated. Here, we focused on the role and mechanisms of HDAC6 in chlorhexidine gluconate (CG) induced peritoneal fibrosis and discussed the mechanisms involved. We found Tubastatin A (TA), a selec

Can the AGE/RAGE/ERK signalling pathway and the epithelial-to-mesenchymal transition interact in the pathogenesis of chronic rhinosinusitis with nasal polyps?

Chronic rhinosinusitis with nasal polyps (CRSwNP) is a persistent sinonasal mucosa inflammatory disease with still unclear pathophysiologic mechanisms that imply events of tissue repair and structural remodelling. Several cascades seem to have a considerable role in the onset and progression of mucosa hyperproliferation in nasal polyps including transforming growth factor β/Small mother against decapentaplegic (TGFβ/Smads), mitogenactivated protein kinases (MAPKs), advanced glycosylation end-products (AGEs) together with epithelial-tomesenchymal transition (EMT). Since many inflammatory mediat

Extracellular miR-145, miR-223 and miR-326 expression signature allow for differential diagnosis of immune-mediated neuroinflammatory diseases.

BACKGROUND: Although misdiagnosis of neuromyelitis optica spectrum disorder (NMOSD) with neuropsychiatric systemic lupus erythematosus (NPSLE) or multiple sclerosis (MS) is not infrequent, reliable biomarkers remains an unmet need. Extracellular microRNAs (miRNAs) represent a worthy avenue to identify biomarkers for differential diagnosis. We aimed to explore the potential role of some selected circulating miRNAs as biomarkers for the differential diagnosis in immune-mediated neuroinflammatory diseases. METHODS: A total of 80 subjects were enrolled in the present study, including 37 patients

Transforming growth factor-β directly induces p53-up-regulated modulator of apoptosis (PUMA) during the rapid induction of apoptosis in myc-driven B-cell lymphomas.

c-Myc transformed human Burkitt's lymphoma (BL) cells are highly sensitive to TGF-β-induced apoptosis. Previously we demonstrated that TGF-β-mediated cell death in BL cells is regulated via the mitochondrial intrinsic apoptosis pathway, which is dependent on the activation of BAX and/or BAK. TGF-β directly induces transcription of the BH3-only protein BIK and represses expression of the pro-survival factor BCL-X(L) but has no effect on the direct BAX/BAK "activators" BIM or BID (tBID). Here we show that TGF-β induces the BH3-only activator PUMA to aid induction of the intrinsic cell death path
