Treponema pallidum was demonstrated in a cutaneous, tertiary syphilitic lesion by indirect immunofluorescence microscopy, but not by darkfield microscopy illumination or silver stain. The numerous organisms observed by this method may help explain the histologically vigorous tissue reaction in tertiary syphilis, despite the scarcity of organisms demonstrable by other methods.

Searching for a chelator with increased sensitivity for use as cadmium stain, a number of benzothiazolylazonaphthol derivatives were synthesized and their staining properties for cadmium and zinc were examined. Benzothiazolylazo-beta-naphthol and -naphthol AS (BTAN-AS) stained cadmium with the highest staining intensity and the most precise localization. The frequencies of dicentric chromosomes and of partial premature chromosome condensation (PCC) in definitely first, second, or third mitoses after X-irradiation of human leukocytes, as determined with a bromodeoxyuridine (BUdR)/ Giemsa labeling technique, were investigated. The results show that practically no PCCs occur in first mitoses after irradiation and culture start in contrast to second or third ones. The frequencies of dicentrics from first to second, as well as from second to third mitoses after irradiation decline by about 50% for each division step.

A description of vitally unrecognized cryptococcal meningitis and tumour-like cryptococcosis of the adrenals is presented. Comparison of the results of various stainings for degeneratively changes and phagocytized fungi suggests that cryptococci in histological sections should be stained by Zihl-Nilsen's method (methylene blue and basic fuchsin).

The application of hydrogen peroxide and egg albumin to paraffin sections before immunostaining prevents nonspecific staining by immunoperoxidase techniques. This method is more effective than pretreating secions with normal sera, or using either egg albumin or hydrogen peroxide separately, or using diluted antisera with prolonged incuabations in the staining procedure.

Histochemical differentiation of Onchocerca volvulus microfilariae from 164 patients in West African rain-forest (Liberia), Sudan-savanna (Upper Volta), Guatemala and the Yemen has been carried out using a staining method for the demonstration of acid phosphatase. An aniline blue fluorescence staining technique specific for eosinophils was compared to haematoxylin and eosin to determine the percentages of eosinophils in the peripheral blood of patients with atopic or with allergic contact dermatitis. The aniline blue technique proved to be excellent for demonstrating eosinophilic granulocytes. An improved method of preparing Coxiella burnetii natural phase II antigen of high purity enabled its staining for use in the microagglutination reaction (MAR).
