In an attempt to characterize immunological reactions in patients with progressive systemic sclerosis (PSS) mononuclear cells were tested for their ability to secrete interleukin-2 (II-2), and for their response to the T-cell independent B-cell mitogen anti-mu sepharose. Compared with patients with systemic lupus erythematosus (SLE), patients with PSS showed a marked enhancement in their response to anti-mu. With regard to II-2 secretion mononuclear cells from PSS patients tended to show increased responses as well. Our data provide evidence for hyperreactivity at both the T- and B-cell level

An imbalance in immunoregulatory cells might explain the increase in autoimmune phenomena associated with hyposplenism. Using the monoclonal antibodies OKT3, OKT4 and OKT8 to identify T-lymphocyte subsets, we have studied 19 patients with treated coeliac disease, 9 of whom had hyposplenism, 10 splenectomized subjects, 10 gastrointestinal control patients and 10 normal subjects. Baseline immunologic abnormalities were identified in 16 hospitalized intravenous drug abusers ( IDAs ) without acquired immune deficiency syndrome (AIDS). Twelve (75%) of 16 had inverted helper-suppressor ratios. In seven patients (44%), the abnormal ratio resulted from an increase in the absolute number of suppressor cells with a normal number of helper T lymphocytes. In five patients (31%), the reduced ratio resulted primarily from decreased helper cells, immunophenotypic findings similar to those seen in patients with AIDS. In 72 workers having occupational contact with benzene, toluene or xylene during 31 to 122 months, the T lymphocyte functions were evaluated. Workers after a service of 55-122 months showed a decreased T lymphocyte count without, however, alterations in their functions. No clinical signs of diminished immunological reactivity were noted in the subjects studied. The E rosette test is of importance in the evaluation of early effects on the lymphocyte system of the toxic action of benzene and its homologues.

T lymphocytes were fractionated according to their receptors for IgG (TG) or IgM (TM) and scored in 37 patients with Graves' disease (17 hyperthyroid and untreated. 10 euthyroid on antithyroid drugs, 10 in long-term remission after radioiodine therapy). TG percentages were very low both in untreated and in drug-treated patients. By contrast, normal TG levels were observed in patients in long-term remission. These data are consistent with the hypothesis of a defective suppressor cell activity in Graves' disease.
