A food-borne probiotic ameliorates depression by modulating tryptophan metabolism along the gut-brain axis.

Depression is increasingly conceptualized within a neuroimmune framework, where systemic inflammation originating from a compromised gut bar … nes (IL-23, IFN-γ) and oxidative stress. Concurrently, hippocampal metabolomics showed BC99 shifted tryptophan metabolism from the immune-activated, neurotoxic kynurenine pathway toward the neuroprotective serotonin pathway. Integrated hippocampal transcriptomics and proteomics identified the activation of the BDNF/TrkB/CREB neurotrophic signaling pathway an …

High-tryptophan diet mitigates parvalbumin-induced allergic responses via gut microbiome remodeling and immune modulation.

Parvalbumin-induced food allergy model in BALB/c mice were established to investigate the intervention effect of high- tryptophan (Trp) diet on allergic responses. Trp suppressed histamine and IL-6 release from the sensitized KU812 cells, effectively mitigated parvalbumin-induced cellular sensitization. High-Trp diet alleviated allergic symptoms, reduced the organ index of the spleen and serum specific IgE, IgG, IgG1 and IgG2a levels, alongside suppressed Th2-type immune responses in mice. Meanwhile, Trp not only influenced the relative abundance of allergy-related gut microbiome, such as Faec…

The gut microbiota-tryptophan metabolism-immune axis in inflammatory bowel disease: Mechanisms and therapeutic prospects.

The gut microbiota-tryptophan (TRP) metabolism-immune axis is one of the core pathogenic mechanisms in inflammatory bowel disease (IBD). Physiologically, gut microbes convert dietary TRP into bioactive metabolites that regulate intestinal immune balance and barrier function via AHR. In IBD, this axis enters a vicious cycle of dysbiosis, metabolic disruption, and immune imbalance - characterized by reduced beneficial bacteria, excessive 5‑HT production, and kynurenine pathway overactivation - which drives T‑cell dysregulation, aberrant macrophage polarization, and compromised barrier integrity.…

Rh-Catalyzed regioselective C7Ar-allylation of tryptophan with VCC and its late-stage peptide diversification.

N-Pivaloyl-directed Rh(III)-catalyzed regioselective C7Ar-allylation of protected tryptophans was achieved with vinyl-1,3-dioxolan-2-one, furnishing C7Ar-(hydroxymethyl)allylated tryptophan-based unnatural amino acids in high yields. Furthermore, the late-stage exemplification of the strategy was successfully accomplished in tryptophan-containing dipeptides, tripeptides and tetrapeptides. Slight modification in the reaction conditions enabled the formation of C7Ar-(formyl)allylated tryptophan-based unnatural amino acids and dipeptides in moderate yields.

Parkinson's disease: an integrative bioinformatics and machine learning analysis reveals tryptophan metabolism-associated gene biomarkers.

BACKGROUND: Parkinson’s disease (PD) represents a prevalent neurodegenerative disorder among the aging population, characterized by significant disruptions in neurological metabolic pathways. Recent evidence highlights the crucial role of tryptophan metabolism in the pathogenesis of PD; however, a comprehensive exploration of tryptophan metabolism genes (TMGs) and their specific contributions remains largely uncharted. METHODS: Employing bioinformatics, this study aimed to identify and validate TMGs associated with PD. A differential expression analysis was performed on a carefully selected se…

Tryptophan-Containing Antimicrobial Peptides Attenuate Colorectal Cancer Progression by Inhibiting the Growth of Fusobacterium nucleatum.

Colorectal cancer, which originates in the epithelial cells of the colon or rectum, is closely associated with dysbiosis of the gut microbiota. Increasing evidence has shown that Fusobacterium nucleatum plays a significant role in colorectal cancer progression by activating inflammatory responses, modulating the tumor microenvironment, and promoting tumor cell proliferation. Antimicrobial peptides targeting Fusobacterium nucleatum have the potential to serve as more effective and less toxic therapeutic agents compared to chemotherapy drugs. In this study, we systematically evaluated the antiba

Heterogeneous microalgal extracellular polymeric substances-phenanthrene interaction mechanisms modulated by time-dynamic secretion of tryptophan.

Microbial extracellular polymeric substances (EPS) act as critical receptors for environmental pollutants at the water-cell interface, providing binding sites, especially for tryptophan (Trp). A time variation in Trp secretion is expected during the proliferation of microalgal cells. However, how the change in Trp level regulates the micro-interfacial interactions between algal cells and pollutants remains poorly understood. We investigate the binding of a polycyclic aromatic compound (PAC) phenanthrene to EPS of Chlorella pyrenoidosa over time using a combination of in vivo and in silico anal…

CsERF21-l-CsASA2 module positively regulates cold tolerance by promoting tryptophan biosynthesis in tea plants.

Cold stress is a widely distributed abiotic stress that severely limits the yield and quality of tea plants. Tryptophan (Trp) and downstream indole compounds play an important role in plant growth and stress response. However, beyond its involvement in indole compounds synthesis, the other physiological functions of Trp in tea plants remain unknown. In this study, anthranilate synthetase (ASA2) was a positive regulator to enhance cold stress tolerance in Camellia sinensis. The expression of CsASA2 was strongly induced by cold stress. Suppression of CsASA2 expression in C. sinensis reduced the …

Tryptophan Metabolite ITE Attenuates LPS-Induced MMP-9 via NF-κB/AP-1 in Monocytes.

Matrix metalloproteinase-9 (MMP-9) and lipopolysaccharide (LPS) levels are known to be elevated in obesity and contribute to metabolic dysfunction. 2-(1'H-indole-3'-carbonyl)-thiazole-4-carboxylic acid methyl ester (ITE), an endogenous ligand of the aryl hydrocarbon receptor (AhR), has been implicated in the regulation of inflammatory responses. This study aimed to determine whether ITE can inhibit LPS-induced MMP-9 expression in monocytic cells and to explore the underlying signaling mechanisms involved. Human monocytic THP-1 cells and primary human monocytes were treated with LPS in the pres

Associations Between Serum Gut-Derived Tryptophan Metabolites and Cardiovascular Health Markers in Adolescents with Obesity.

Background/Objectives: Gut-derived tryptophan (Trp) metabolites play important roles in metabolic and cardiovascular regulation. Although animal studies suggest their protective effects against metabolic dysfunction, data in adolescents, particularly those with obesity, remain limited. The objective of this study was to evaluate associations between circulating gut-derived Trp metabolites and markers of cardiometabolic, vascular, and platelet health in adolescents with obesity. Methods: Data were analyzed from 28 adolescents (ages 13-18; mean BMI = 36 ± 6.4 kg/m2). Fasting blood was collected …

Gut Microbial Tryptophan Metabolism Is Involved in Post-Cardiac Arrest Brain Injury via Pyroptosis Modulation.

AIMS: Post-cardiac arrest brain injury (PCABI) is a leading cause of death in cardiac arrest/cardiopulmonary resuscitation (CA/CPR) victims and long-term disability in CA/CPR survivors. Despite previous evidence indicating that the microbiota-gut-brain axis is critically involved in many neurological disorders, no research has hitherto established a connection between the gut microbiota and PCABI through this axis. This study aims to explore the biological roles of microbial tryptophan metabolites in the progression of PCABI. METHODS: To achieve this, we pretreated rats with a cocktail of bro…

Periconceptional Non-medical Maternal Determinants Influence the Tryptophan Metabolism: The Rotterdam Periconceptional Cohort (Predict Study).

BACKGROUND: The vital role of the maternal tryptophan (TRP) metabolism in maternal health and pregnancy is well established. However, non-medical maternal determinants influencing the TRP metabolism have been poorly investigated. We hypothesise that periconceptional maternal non-medical determinants alter the TRP metabolism, affecting both kynurenine (KP) and serotonin pathway (SP) metabolite concentrations. Therefore, we investigated the influence of non-medical maternal determinants on the TRP metabolism during the periconception period. METHODS: About 1916 pregnancies were included from th…

Dysrupted microbial tryptophan metabolism associates with SARS-CoV-2 acute inflammatory responses and long COVID.

Protection against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and risk of long COVID has been associated with the depletion or over-abundance of specific taxa within the gut microbiome. However, the microbial mechanisms mediating these effects are not yet known. We hypothesized that altered microbial production of tryptophan and its downstream derivatives might contribute to inappropriate immune responses to viral infection. In patients hospitalized with COVID-19 (n = 172), serum levels of tryptophan and indole-3-propionate (IPA) negatively correlated with serum lev…

Dual emission chiral carbon dots as fluorescent probe for fast chiral recognition of tryptophan enantiomers.

BACKGROUND: Chirality is an essential property of nature. Chiral recognition is of great significance to life sciences, pharmaceutical industry, food analysis, and so on. Chiral carbon dots (CCDs), as green nanomaterials, have great prospects in chiral sensing. However, CCDs with enantioselectivity for tryptophan (Trp) enantiomers are scarce. Moreover, most chiral sensing platforms depend on the difference of fluorescence intensity at the same emission wavelength to identify enantiomers, it is still a challenge to distinguish enantiomers by the positions of fluorescent emission peaks. RESULTS…

Novel Tryptophan Hydroxylase Inhibitor TPT-001 Reverses PAH, Vascular Remodeling, and Proliferative-Proinflammatory Gene Expression.

The serotonin pathway has long been proposed as a promising target for pulmonary arterial hypertension (PAH)-a progressive and uncurable disease. We developed a highly specific inhibitor of the serotonin synthesizing enzyme tryptophan hydroxylase 1 (TPH1), TPT-001 (TPHi). In this study, the authors sought to treat severe PAH in the Sugen/hypoxia (SuHx) rat model with the oral TPHi TPT-001. Male Sprague Dawley rats were divided into 3 groups: 1) ConNx, control animals; 2) SuHx, injected subcutaneously with SU5416 and exposed to chronic hypoxia for 3 weeks, followed by 6 weeks in room air; and 3…

Tryptophan As a New Member of RNA-Induced Silencing Complexes Prevents Colon Cancer Liver Metastasis.

Essential amino acids (EAA) and microRNAs (miRs) control biological activity of a cell. Whether EAA regulates the activity of miR has never been demonstrated. Here, as proof-of-concept, a tryptophan (Trp, an EAA) complex containing Argonaute 2 (Ago2) and miRs including miR-193a (Trp/Ago2/miR-193a) is identified. Trp binds miR-193a-3p and interacts with Ago2. Trp/Ago2/miR-193a increases miR-193a-3p activity via enhancing Argonaute 2 (Ago2) RNase activity. Other miRs including miR-103 and miR-107 in the Trp complex enhance miR-193a activity by targeting the same genes. Mechanistically, the Trp/A…
